Heterologous expression of hydroxylase coupled with pathway optimization enables highly efficient ʟ-DOPA production in Corynebacterium glutamicum.

ʟ-3,4-dihydroxyphenylalanine (ʟ-DOPA), the direct precursor of the neurotransmitter dopamine, is a cornerstone therapeutic agent for Parkinson's disease. In this study, Corynebacterium glutamicum was used as a microbial chassis to systematically evaluate its potential as a cell factory for ʟ-DOPA production. The strain was initially evaluated for ʟ-DOPA tolerance to confirm its suitability for production. Key hydroxylases, HpaB and HpaC, were introduced, and promoter and ribosome-binding site (RBS) optimization was applied to establish a de novo ʟ-DOPA biosynthetic pathway. To minimize flux to

Enhancing the production of isopropanol with reduced CO2 emission via protein and metabolic engineering using Corynebacterium glutamicum.

Isopropanol (IPA), a versatile chemical with applications in various fields of industries, yet its petroleum-based production raises environmental concerns. In this study, Corynebacterium glutamicum was engineered to enhance IPA production while mitigating CO2 emissions. First, the rational design of secondary alcohol dehydrogenase (SADH) variants with shifted cofactor specificity from NADPH to NADH, resulting in an 11.11-fold increased NADH oxidation rate and 6.02-fold increased enzyme activity. Secondly, modified SADH was used in combination with the Ncgl1676 promoter in order to separate th

Development strategy of non-GMO organism for increased hemoproteins in Corynebacterium glutamicum: a growth-acceleration-targeted evolution.

Heme, found in hemoproteins, is a valuable source of iron, an essential mineral. The need for an alternative hemoprotein source has emerged due to the inherent risks of large-scale livestock farming and animal proteins. Corynebacterium glutamicum, regarded for Qualified Presumption of Safety or Generally Recognized as Safe, can biosynthesize hemoproteins. C. glutamicum single-cell protein (SCP) can be a valuable alternative hemoprotein for supplying heme iron without adversely affecting blood fat levels. We constructed the chemostat culture system to increase hemoprotein content in C. glutamic

Engineering of sugar metabolism of Corynebacterium glutamicum for production of amino acid L-alanine under oxygen deprivation.

Corynebacterium glutamicum was genetically engineered to produce L-alanine from sugar under oxygen deprivation. The genes associated with production of organic acids in C. glutamicum were inactivated and the alanine dehydrogenase gene (alaD) from Lysinibacillus sphaericus was overexpressed to direct carbon flux from organic acids to alanine. Although the alaD-expressing strain produced alanine from glucose under oxygen deprivation, its productivity was relatively low due to retarded glucose consumption. Homologous overexpression of the gapA gene encoding glyceraldehyde 3-phosphate dehydrogenas

Metabolic Engineering of Corynebacterium glutamicum for the Production of the Four-Carbon Platform Chemicals γ-Hydroxybutyrate and γ-Butyrolactone.

γ-Hydroxybutyrate (GHB) is an important C4 platform chemical, serving as a crucial precursor for the synthesis of various bulk chemicals, including γ-butyrolactone (GBL) and 1,4-butanediol (1,4-BDO). In this study, we report the systematic metabolic engineering of Corynebacterium glutamicum for the biological production of GHB from glucose via the introduction of a glutamate-derived pathway. We showed that C. glutamicum is a promising host for producing GHB due to its higher tolerance to GHB as compared to other chassis. By screening key enzymes capable of converting glutamate into GHB and blo

Production of pure all-trans retinal from agricultural byproducts by metabolically engineered Corynebacterium glutamicum using two-phase cultivation.

Retinoids, including retinal, retinol (vitamin A), and retinoic acid, comprise vitamin A and its biologically active metabolites, which are widely used in pharmaceutical, healthcare, and cosmetic applications. Among these, retinal has attracted increasing attention in skincare applications because of its favorable combination of high biological efficacy with low skin irritation, and its potent antibacterial activity. However, microbial production of retinal generally yields mixtures with other retinoids and requires expensive nutrient media. Here, we report the production of pure retinal from

Investigation of phosphorylation status of OdhI protein during penicillin- and Tween 40-triggered glutamate overproduction by Corynebacterium glutamicum.

Glutamate overproduction by Corynebacterium glutamicum is triggered by treatment with penicillin or Tween 40 and is accompanied by a decrease in 2-oxoglutarate dehydrogenase complex (ODHC) activity. We have reported that de novo synthesis of OdhI, which inhibits ODHC activity by interacting specifically with the E1o subunit of ODHC (OdhA), is induced by penicillin, and that odhI overexpression induces glutamate overproduction in the absence of any triggers for glutamate overproduction. In this study, to determine the function of OdhI in glutamate overproduction by C. glutamicum, changes in Odh

[Protein engineering of PgsA combined with Bayesian optimization drives efficient poly-γ-glutamic acid synthesis by Corynebacterium glutamicum].

Poly-γ-glutamic acid (γ-PGA) is a biopolymer polymerized from l-glutamic acid (l-Glu) and/or d-glutamic acid monomers, exhibiting broad application prospects in pharmaceuticals, cosmetics, and other fields. The low yields remain a key factor limiting large-scale production of γ-PGA in heterologous expression systems. To enhance the biosynthetic efficiency, this study employed Corynebacterium glutamicum as the chassis cell and utilized the γ-PGA synthase PgsBCA from Bacillus licheniformis to catalyze γ-PGA synthesis. First, a polycistron expression system was constructed, increasing the γ-PGA y

Bio-isopropanol production in Corynebacterium glutamicum: Metabolic redesign of synthetic bypasses and two-stage fermentation with gas stripping.

Isopropanol is a commodity chemical widely used as a biofuel, fuel additive, rubbing alcohol and intermediate in various fields. Here, an engineered Corynebacterium glutamicum overproducing isopropanol was developed. To our knowledge, despite a representative industrial host to produce valuable chemicals, the high-level production of isopropanol in C. glutamicum has never been reported. First, the problem of the inability to produce isopropanol was solved by finding a key factor in its metabolism. The consolidation and modular optimization of synthetic bypasses including succinate and mevalona

Genes and enzymes of the acetyl cycle of arginine biosynthesis in Corynebacterium glutamicum: enzyme evolution in the early steps of the arginine pathway.

A cluster of arginine biosynthetic genes of Corynebacterium glutamicum ATCC 13032, comprising argJ, argB and argD as well as part of argC and argF, has been cloned by heterologous complementation of an Escherichia coli argE mutant. The gene order has been established as argCJBDF by sequencing the entire 4.4 kb cloned DNA fragment. The C. glutamicum argB gene can be transcribed in E. coli cells from an internal promoter located in the coding part of the preceding argJ gene, whereas transcription of the argJ gene appears vector-dependent. Expression of the corynebacterial argB gene is repressed

Regulatory mechanism of ferrous ion in enhancing the biosynthesis of L-homoserine, L-threonine, and L-isoleucine in Corynebacterium glutamicum.

Iron is a vital nutrient for almost all living organisms, predominantly existing in its ferrous Fe2+ and ferric Fe3+ states within cells. The limited bioavailability of Fe3+ due to its poor solubility necessitates Fe2+ supplementation in bacterial culture media to meet cellular demands. During fermentation studies with three Corynebacterium glutamicum strains, elevated Fe2+ concentrations were observed to enhance biomass accumulation, accelerate glucose consumption, maintain pH homeostasis, and promote product synthesis. To elucidate the precise transmission of iron signaling to amino acid bio

Integrated Transporter Engineering and Semi-Rational Enzyme Design in Corynebacterium glutamicum for High-Level Natural Blue Pigment Indigoidine Production.

Indigoidine is a sustainable natural blue pigment with broad industrial prospects, while its microbial production is hampered by growth inhibition of the producing strain, enzyme-host incompatibility, and insufficient industrial-scale titer. In this study, de novo biosynthesis of indigoidine was achieved in Corynebacterium glutamicum. Efflux pump engineering was employed to alleviate growth inhibition of the production strain, and semi-rational design of the key synthetase BpsA generated the optimal mutant Y433Q with enhanced catalytic activity. Isobutanol production in Corynebacterium glutamicum: Suppressed succinate by-production by pckA inactivation and enhanced productivity via the Entner-Doudoroff pathway.

On the basis of our previous studies of microbial L-valine production under oxygen deprivation, we developed isobutanol-producing Corynebacterium glutamicum strains. The artificial isobutanol synthesis pathway was composed of the first three steps of the L-valine synthesis pathway; and the subsequent Ehrlich Pathway: pyruvate was converted to 2-ketoisovalerate in the former reactions; and the 2-keto acid was decarboxylated into isobutyraldehyde, and subsequently reduced into isobutanol in the latter reactions. Although there exists redox cofactor imbalance in the overall reactions, i.e., NADH

The effect of reactive oxygen species (ROS) and ROS-scavenging enzymes, superoxide dismutase and catalase, on the thermotolerant ability of Corynebacterium glutamicum.

The function of two reactive oxygen species (ROS) scavenging enzymes, superoxide dismutase (SOD) and catalase, on the thermotolerant ability of Corynebacterium glutamicum was investigated. In this study, the elevation of the growth temperature was shown to lead an increased intracellular ROS for two strains of Corynebacterium glutamicum, the wild-type (KY9002) and the temperature-sensitive mutant (KY9714). In order to examine the effects of ROS-scavenging enzymes on cell growth, either the SOD or the catalase gene was disrupted or overexpressed in KY9002 and KY9714. In the case of the KY9714 s

Overexpression of the phosphofructokinase encoding gene is crucial for achieving high production of D-lactate in Corynebacterium glutamicum under oxygen deprivation.

We previously reported on the impacts of the overexpression of individual genes of the glycolytic pathway encoding glucokinase (GLK), glyceraldehyde phosphate dehydrogenase (GAPDH), phosphofructokinase (PFK), triosephosphate isomerase (TPI), and bisphosphate aldolase (FBA) on D-lactate productivity in Corynebacterium glutamicum under oxygen-deprived conditions. Searching for synergies, in the current study, we simultaneously overexpressed the five glycolytic genes in a stepwise fashion to evaluate the effect of the cumulative overexpression of glycolytic genes on D-lactate production. 5'-End Translationalization: Iterative Assembly of Leaderless Polycistronic Amplifiers for Context-Independent Expression in the Food-Grade Bacterium Corynebacterium glutamicum.

Corynebacterium glutamicum is a crucial food-grade (GRAS) bacterial chassis widely utilized for the industrial production of amino acids and nutraceuticals. However, the efficient production of recombinant proteins and secondary metabolites in this host remains limited by context dependence and low translational efficiency. To overcome this, we introduce a 5'-end translationalization strategy. By repurposing passive 5' untranslated regions (5'UTRs) into actively translated fore-cistrons, we converted conventional monocistronic designs into context-independent, leaderless polycistronic designs

Enhancing d-Pantothenic Acid Production in Corynebacterium glutamicum via Integrating Dynamic Regulation With Structure-Guided Protein Engineering of Ketopantoate Reductase.

d-Pantothenic acid (DPA) is an essential vitamin with broad applications. In this study, we engineered Corynebacterium glutamicum for high-titer DPA production by integrating dynamic pathway regulation with structure-guided protein engineering. Functional characterization revealed that endogenous CgPanE and BsPanE2 function as α-hydroxy acid dehydrogenases rather than ketopantoate reductases, whereas heterologous EcPanE and BsPanE, bifunctional activity toward both ketoisovalerate and ketopantoate, possess larger active cavities. A stationary-phase promoter (P4-N14) was employed to delay ketop

Functional Identification of AcsR, a MarR Family Transcriptional Regulator Involved in the Regulation of Aromatic Compound-Degrading Genes in Corynebacterium glutamicum.

The MarR (multiple antibiotic resistance regulator) family regulators, which are widely conserved across various organisms, play pivotal roles in metabolism, stress response mechanisms, and virulence factor production. However, the regulatory functions of these factors in the degradation of aromatic compounds within Corynebacterium glutamicum remain largely uncharacterized. In this study, we identified a MarR-type regulator, designated AcsR (encoded by ncgl2425), which directly represses the expression of the catechol 2,3-dioxygenase gene ncgl2007 (c23o) and the heavy metal (nickel) transport

Corynebacterium glutamicum sigmaE is involved in responses to cell surface stresses and its activity is controlled by the anti-sigma factor CseE.

In this study, we demonstrate that sigma(E), an alternative sigma factor of Corynebacterium glutamicum, is involved in cell surface stresses. Cells in which the sigE gene was deleted evidenced increased sensitivity to magnesium deficiency, as well as to SDS, lysozymes, EDTA and heat. We utilized physiological analyses to show that the downstream gene, designated cseE, encodes an anti-sigma factor. The retarded growth of the cseE mutant cells under ordinary growth conditions could be recovered by an additional deletion of sigE encoding sigma(E). Under stress conditions, the phenotype of the cse
