Small Mammals as Reservoir for Zoonotic Agents in Afghanistan.

INTRODUCTION: Rodents and other small mammals can serve as reservoirs for a large number of zoonotic pathogens. A higher risk of infection with rodent-borne pathogens exists for humans with direct contact to rodents and/or their excretions, e.g., soldiers in operation areas. To date, little is known about endemic human pathogenic disease agents that are naturally associated with small mammals in Afghanistan. The aim of this study was to screen abundant rodents and insectivores collected from 2009 to 2012 in four field camps of the German Federal Armed Forces (Bundeswehr) in Northern Afghanista

A Flexible Hybrid Site-Specific Integration-Based Expression System in CHO Cells for Higher-Throughput Evaluation of Monoclonal Antibody Expression Cassettes.

The implementation of site-specific integration (SSI) systems in Chinese hamster ovary (CHO) cells for the production of monoclonal antibodies (mAbs) can alleviate concerns associated with production instability and reduce cell line development timelines. SSI cell line performance is driven by the interaction between genomic integration location, clonal background, and the transgene expression cassette, requiring optimization of all three parameters to maximize productivity. Systematic comparison of these parameters has been hindered by SSI platforms involving low-throughput enrichment strateg

Optimized Split Zeocin Selection System Enhances Monoclonal Antibody Productivity and Reduces High Mannose Levels in CHO Cells.

The limited variety of selection markers presents a challenge in the development and production of complex biologic therapeutics. Here, we developed and optimized a split zeocin selection system using a split bleomycin binding protein (BBP) to address these limitations. Two split bleomycin binding protein strategies (the N-terminal fragment paired with the heavy chain + the C-terminal fragment paired with the light chain (LC), or the reciprocal arrangement) were evaluated in multiple monoclonal antibodies. Our results demonstrate that the split zeocin system achieves comparable or higher titer

Positive feedback during sulfide oxidation fine-tunes cellular affinity for oxygen.

UNLABELLED: Sulfide (H2S in the gas form) is the third gaseous transmitter found in mammals. However, in contrast to nitric oxide (NO) or carbon monoxide (CO), sulfide is oxidized by a sulfide quinone reductase and generates electrons that enter the mitochondrial respiratory chain arriving ultimately at cytochrome oxidase, where they combine with oxygen to generate water. In addition, sulfide is also a strong inhibitor of cytochrome oxidase, similar to NO, CO and cyanide. The balance between the electron donor and the inhibitory role of sulfide is likely controlled by sulfide and oxygen availa

Host traits shape flea infestation patterns in small mammals: a case study of Spermophilus undulatus and associated flea species in northern Xinjiang, China.

OBJECTIVE: Fleas are obligate ectoparasites of mammals and play an important role in the transmission of zoonotic pathogens, including Yersinia pestis. Describing empirical patterns of flea infestation across host communities is essential for understanding host-parasite associations in plague-endemic regions. The aim of this study is to investigate the relationship between fleas and their host animals by examining how host traits influence flea parasitism, with the goal of offering novel insights for future plague prevention strategies and related research. METHODS: From 2022 to 2025, small m

Systematic review and data-driven insights into CHO cell engineering for next-generation antibody production.

Chinese hamster ovary (CHO) cells remain the dominant platform for therapeutic antibody and biopharmaceutical production, yet productivity bottlenecks persist, particularly for complex molecules. To identify overarching trends in host cell optimization, a systematic review and quantitative cross-study analysis of 164 publications (2011-2024) reporting CHO cell engineering strategies with effects on titer or specific productivity was conducted. Data from 466 engineered targets were extracted and analyzed by strategy, pathway, and production context. The field - driven largely by antibody produc

Rapid expression of therapeutic antibodies in mammalian cells via mRNA transfection.

Messenger RNA (mRNA) has emerged as a powerful tool for protein expression in clinical settings, yet its potential as a platform for biologics manufacturing remains underexplored. Here, we evaluate transient mRNA transfection in Chinese hamster ovary (CHO) cells as a rapid and versatile system for protein production. Using reporter mRNAs, we optimize transfection efficiency and benchmark performance against industry-standard plasmid transfection and stable cell line methods. We demonstrate that co-transfection of heavy and light chain mRNAs enables the efficient synthesis, assembly and secreti

A design space for the filtration of challenging monoclonal antibodies using Planova™ S20N, a new regenerated cellulose virus removal filter.

Virus removal by filtration is a crucial step in ensuring the safety of therapeutic antibodies and other biopharmaceutical products by mitigating the risk of endogenous and adventitious viral contamination. However, there are monoclonal antibodies (mAb) that are difficult to filter effectively using virus removal filters (i.e., challenging mAbs), necessitating the creation of guidelines for designing suitable filtration conditions for these challenging molecules. This study presents a filtration design space for filtration conditions using a new regenerated cellulose membrane filter with a rep

Methoxylated Modification of Glutathione-Mediated Metabolism of Halobenzoquinones In Vivo and In Vitro.

Xenobiotics were generally detoxified in organisms through interaction with endogenous molecules, which may also generate metabolites of increased toxicity. Halobenzoquinones (HBQs), a group of highly toxic emerging disinfection byproducts (DBPs), can be metabolized by reacting with glutathione (GSH) to form various glutathionylated conjugates (SG-HBQs). In this study, the cytotoxicity of HBQs in CHO-K1 cells showed a wavy curve as a function of increased GSH dosage, which was inconsistent with the commonly recognized progressive detoxification curve. We hypothesized that the formation and cyt

Multiple mechanisms of action of pyridoxine in primary hyperoxaluria type 1.

Primary hyperoxaluria type 1 (PH1) is a rare hereditary calcium oxalate kidney stone disease caused by a deficiency of the liver-specific pyridoxal-phosphate-dependent peroxisomal enzyme alanine:glyoxylate aminotransferase (AGT). About one third of patients are responsive to pharmacological doses of pyridoxine (vitamin B6), but its mechanism of action is unknown. Using stably transformed Chinese Hamster Ovary (CHO) cells expressing various normal and mutant forms of AGT, we have shown that pyridoxine increases the net expression, catalytic activity and peroxisomal import of the most common mis

Development and Optimization of a Hollow Fiber-Based Countercurrent Dialysis Process for Continuous Manufacturing of Monoclonal Antibodies.

Countercurrent dialysis using hollow fiber modules offers several advantages over both inline diafiltration (ILDF) and hemodialyzer based countercurrent dialysis in terms of buffer requirements and compatibility with GMP manufacturing of biologics. In this work, we evaluate GMP-grade hollow fiber modules of varying lengths (20, 41.5, and 65 cm) and benchmark their performance against an ILDF unit as a reference process for continuous exchange of buffer in manufacturing of monoclonal antibodies. Hollow fiber modules outperformed the ILDF configuration in terms of buffer consumption and pressure

Perfusion-Based Antibody Production in the Ambr® 250 Modular.

The perfusion mode has become increasingly important in biopharmaceutical production in recent years. A bioreactor system used in many laboratories for the development of monoclonal antibodies (mAbs) production processes is the Sartorius' Ambr. 250 system. Vessels designed for perfusion mode are only available for its high throughput version, while the modular version of the Ambr 250 is not designed for perfusion mode. In this study, perfusion processes for the production of a mAb with Chinese Hamster Ovary (CHO) cells were realized in the Ambr 250 Modular in combination with Repligen's ATF 1

Use of Raman spectroscopy for the label-free discrimination of CHO cells associated with antibody expression.

The performances of host cells that produce therapeutic proteins is important for the efficient production of drug substances, to minimize production costs, and to ensure competitive prices for commercial drugs in the biopharmaceutical industry. However, the selection of cells that exhibit high performances in the expression of proteins and in the construction of master cell banks is a complicated and time-consuming process. Consequently, novel methods are required to assess the expression levels of drug molecules and to select dominant cells at an early stage. In this study, Raman spectroscop

The signaling and selectivity of α-adrenoceptor agonists for the human α2A, α2B and α2C-adrenoceptors and comparison with human α1 and β-adrenoceptors.

α2-adrenoceptors, (α2A, α2B and α2C-subtypes), are Gi-coupled receptors. Central activation of brain α2A and α2C-adrenoceptors is the main site for α2-agonist mediated clinical responses in hypertension, ADHD, muscle spasm and ITU management of sedation, reduction in opiate requirements, nausea and delirium. However, despite having the same Gi-potency in functional assays, some α2-agonists also stimulate Gs-responses whilst others do not. This was investigated. Agonist responses to 49 different α-agonists were studied (CRE-gene transcription, cAMP, ERK1/2-phosphorylation and binding affinity)

Sensitive and Robust Capillary Electrophoretic Analysis of Host Cell Residual DNA in Biopharmaceutical Samples.

Host cell residual DNA (rDNA) is a critical process-related impurity in biopharmaceutical production, involving trace amounts of DNA fragments originating from yeast, bacteria, CHO (Chinese hamster ovary), and other host cells. Monitoring both the size and quantity of rDNA is essential because of potential safety risks, including oncogenicity and immunogenicity, in biological products such as vaccines and other protein therapeutics. Regulatory guidelines from the WHO and U.S. FDA recommend limits of less than 10 ng/dose and below 200 base pairs (bp) for the rDNA content and size in final produ

Separation and Purification of CHO Secretome and Extracellular Vesicles for Proteome Analysis.

For decades, host cell proteins (HCPs) have been investigated as putative contaminants in downstream processing of biopharmaceutical products of Chinese hamster ovary (CHO) cells. However, little is still known about the composition of the entire protein and vesicle environment in CHO cultivations. Ever evolving mass spectrometry techniques allow more and more insights into cell-cell communication processes and the composition of extracellular matrix, proteases, and further actively segregated compounds such as extracellular vesicles (EVs). EVs themselves are a heterologous group consisting of

Accelerating biopharmaceutical cell line selection with label-free multimodal nonlinear optical microscopy and machine learning.

The selection of high-performing cell lines is crucial for biopharmaceutical production but is often time-consuming and labor-intensive. We investigated label-free multimodal nonlinear optical microscopy for non-perturbative profiling of biopharmaceutical cell lines based on their intrinsic molecular contrast. Employing simultaneous label-free autofluorescence multiharmonic (SLAM) microscopy with fluorescence lifetime imaging microscopy (FLIM), we characterized Chinese hamster ovary (CHO) cell lines at early passages (0-2). A machine learning (ML)-assisted analysis pipeline leveraged high-dime

Assessing the probability of clonality achieved by single-cell cloning of CHO cells through cell deposition combined with imaging using distinguishable cells.

Mammalian cell lines used for clinical studies and post-approval production of recombinant DNA-derived biotherapeutics are expected to be derived from a single cell, and regulatory submissions are expected to provide robust evidence of monoclonality. Imaged single-cell deposition followed by whole-well imaging using specialized instruments has, in many cell line development labs, replaced the "gold standard" of two rounds of limiting dilution due to its increased speed and the assurance of clonality provided by orthogonal images. However, there is still a lack of information on how the procedu
