Comparison of the Level of Substance P and Neurokinin A in Gingival Crevicular Fluid of Sound and Symptomatic Carious Primary Teeth by ELISA.

OBJECTIVES: Pulpal inflammation is often associated with odontogenic pain. Dental pulp is abundantly innervated with sensory fibers encompassing neuropeptides. Neurokinin A (NKA) and substance P (SP) are important neuropeptides in the dental pulp that can cause neurogenic inflammation. Since no previous study has assessed dental pulp neuropeptides in children, this study aimed to compare the level of NKA and SP in gingival crevicular fluid (GCF) of sound and symptomatic carious primary teeth. MATERIALS AND METHODS: Samples of GCF were obtained of 20 sound and 20 painful carious primary teeth.

Nitric oxide synthase in healthy and inflamed human dental pulp.

Nitric oxide synthase (NOS) plays a significant role in the pathogenesis of pulpitis. In this study, we hypothesized the existence of endothelial (eNOS) and inducible (iNOS) enzyme isoforms in human dental pulp. Extracted third molar pulps were divided into groups based on clinical diagnosis: healthy, hyperemic, and irreversible pulpitis. We have localized the eNOS and iNOS by immunohistochemistry and have tested their mRNA expression by RT-PCR and protein levels by Western blots. eNOS is present in the endothelial cells and odontoblasts of the healthy pulp, but an elevation of eNOS mRNA and p

A 2-year retrospective cohort study of pulp-related outcomes of permanent anterior teeth with uncomplicated crown fractures.

BACKGROUND: Uncomplicated crown fractures (UCFs) are the most common form of traumatic dental injury in permanent dentition. However, studies on long-term pulp outcome in South Asian populations are limited. This study aimed to analyse pulp survival of permanent anterior teeth with uncomplicated crown fractures in a Sri Lankan population. METHODS: A retrospective cohort study was conducted following STROBE (Strengthening the Reporting of Observational Studies in Epidemiology) guidelines. Records of 257 teeth with uncomplicated crown fractures in patients attending a tertiary dental care centr

Loss of dental pulp potentially increased the risk of Alzheimer's dementia.

BACKGROUND/PURPOSE: Chronic periodontitis and tooth loss contribute to cognitive decline. Since many biological processes are shared by loss of teeth and loss of pulps, this study investigated the potential association between loss of pulp and the development of dementia. MATERIALS AND METHODS: A retrospective cohort analysis was conducted to investigate the association between dental treatment and the development of dementia. The records of dental treatment during the 10 years prior to the first diagnosis of dementia were extracted from the Elderly Cohort Database of the National Health Info

Bacterial invasion of non-exposed dental pulp.

Anaerobic procedures were adopted to demonstrate the early bacterial invasion of non-exposed dental pulps, and to isolate and identify the bacteria. Of 19 freshly extracted teeth which originally exhibited deep dentinal lesions, clinical examination and electric pulp testing showed that nine of them had no pulpal exposure. Thus the pulps of these teeth were covered by clinically sound dentine beneath the carious lesion. Bacteria were found to have invaded the pulps of six of these nine teeth. The predominant bacteria were obligate anaerobes belonging to the genera Eubacterium, Propionibacteriu

FoxO1 mediates odontoblast differentiation of hDPSCs via B cell-derived ANGPTL1 in dental caries: A laboratory investigation.

AIM: Clinical and in vitro evidence indicates that chronic inflammatory responses initiated by dental caries can persist in the dental pulp even after treatment, necessitating the formation of reparative dentin to restore tissue homeostasis and health. Human dental pulp stem cells (hDPSCs) serve as crucial precursors in this reparative process. This study explores the role of B cells and their secreted factor, Angiopoietin Like 1 (ANGPTL1), in promoting hDPSCs differentiation into odontoblasts under carious conditions, with a particular focus on the activation of Forkhead box O1 (FoxO1). Fabrication of a Vertically Stacked Dentin-Pulp Complex Organ-on-a-Chip Device Using a Human Dentin Disc.

Conventional preclinical models used in dental research, including animal models and static cell cultures, present important limitations in reproducing the physiological conditions of the dentin-pulp interface and predicting human biological responses to dental biomaterials. Organ-on-a-chip (OoC) technology provides an alternative approach by enabling the recreation of tissue-specific microenvironments within controlled microfluidic systems. This manuscript describes a step-by-step protocol for the fabrication and assembly of a dentin-pulp complex OoC device incorporating a human dentin disc w

Quantitation of catecholamines in inflamed human dental pulp by high-performance liquid chromatography.

Catecholamines may play an important role in the control of intrapulpal pressure as mediators of vasoconstriction. A baseline level of catecholamines (dopamine, epinephrine, and norepinephrine) in the uninflamed human dental pulp was previously reported using high-performance liquid chromatography. The purpose of this study was to compare the level of catecholamines present in the inflamed human dental pulp with the baseline level established in virgin teeth. Twelve uninflamed pulps were analyzed as a control and to validate previous findings. Pulp tissue was obtained from 10 vital and inflame

Successful Healing of Periapical Pathology with Partial Pulpotomy in a Mature Permanent Molar: A Case Report.

BACKGROUND Vital pulp therapy approaches are preferred over non-surgical root canal treatment, giving the advantage of preserving the vitality of the dental pulp and thus maintaining its benefits. Such approaches can be performed in teeth having normal apical area; however, performing vital pulp therapy approaches in teeth associated with periapical pathology remains controversial. CASE REPORT We present a case of a mature mandibular right first permanent molar tooth in a medically fit 10-year-old female diagnosed as asymptomatic irreversible pulpitis with asymptomatic apical periodontitis wit

Dental pulp inflammation; experimental studies in human and monkey teeth.

Data from several studies performed during the last 15 years indicate that bacteria and their products are the main etiological factor for dental pulp inflammation. The present series of investigations was conducted in order to determine the irritating effect from various oral bacterial components and restorative measures on dental pulpal tissue and to ascertain reaction patterns of inflamed pulp tissue. The different bacterial materials were tested in an experimental model where the test substance was applied and enclosed in buccal class V cavities prepared in human or monkey teeth. The pulp

Diagnosis dilemmas in vital pulp therapy: treatment for the toothache is changing, especially in young, immature teeth.

The literature is almost devoid of scientific studies of diagnosis of pulpal pathology in primary and permanent teeth with open apices. Most reports are empirical or retrospective studies without adequate prior knowledge of preexisting conditions or histologic findings leading to the necessity of pulpal procedures. Appropriate diagnostic tests and their effectiveness are documented for both groups. This article reviews the available literature and current techniques of indirect pulp therapy, pulp capping, and pulpotomy for primary teeth and permanent teeth with open apex. The apical barrier wi

Feasibility of high-resolution DWI of the healthy dental pulp in human using a hybrid TGSE BLADE sequence.

Clinical evaluation of the human dental pulp is limited to imprecise, subjective methods, necessitating the development of an accurate and objective diagnostic parameter. Diffusion-weighted imaging (DWI) could address this gap; however, no in vivo data are available. To illustrate the feasibility of DWI imaging of the human pulp and to investigate physiological patterns. A turbo gradient spin echo (TGSE) BLADE diffusion MRI sequence was optimized for dental pulp imaging in three volunteers, defining an optimal b-value range and assessing intraindividual fluctuations at 3 T. Apparent diffusion

Feasibility and Outcomes of Cell-based Regenerative Endodontic Therapy in Postautogenous Transplantation of a Mature Tooth: A Case Report.

Autogenous tooth transplantation (ATT) offers advantages; however, success rates depend on factors like socket formation, donor tooth manipulation, and endodontic treatment. Root canal treatment in a mature post-ATT tooth remains challenging. Cell-based regenerative endodontic therapy (RET) shows promise for regenerating the pulp-dentin complex in mature teeth. However, its application in a post-ATT tooth is unexplored. This case report demonstrates the feasibility and outcomes of cell-based RET in a post-ATT tooth. A 37-year-old male underwent ATT for a missing maxillary left first molar. The
