Cellulose-based hybrid glycosilicones via grafted-to metal-catalyzed hydrosilylation: "When opposites unite".

Hydrosilylation catalyzed by the rhodium(I) complex [Rh(acac)(CO)2] or platinum(0)-based Karstedt's catalyst was employed to combine hydrophilic propargylated hydroxyethyl cellulose and hydrophobic hydride-terminated polydimethylsiloxane to give polymer hybrid structures. The final polymers were characterized by FTIR, solid state 1H, 13C and 29Si NMR, contact angle, microcalorimetry and thermogravimetry measurements. The grafting degree was controlled by the catalyst choice and by the reagent load variations; an increase of the polysiloxane load and a change from Karstedt's to the rhodium cata

Biocompatible patterning of proteins on wettability gradient surface by thermo-transfer printing.

We develop a simple and biocompatible method of patterning proteins on a wettability gradient surface by thermo-transfer printing. The wettability gradient is produced on a poly(dimethylsiloxane) (PDMS)-modified glass substrate through the temperature gradient during thermo-transfer printing. The water contact angle on the PDMS-modified surface is found to gradually increase along the direction of the temperature gradient from a low to a high temperature region. Based on the wettability gradient, the gradual change in the adsorption and immobilization of proteins (cholera toxin B subunit) is a

Applying mechanical forces on Drosophila tissues in vivo using the StretchCo, a 3D-printable device.

Applying mechanical forces to tissues helps to understand morphogenesis and homeostasis. Additionally, recording the dynamics of living tissues under mechanical constraints is needed to explore tissue biomechanics. Here, we present a protocol to 3D-print a StretchCo device and use it to apply uniaxial mechanical stress on the Drosophila pupal dorsal thorax epithelium. We describe steps for 3D printing, polydimethylsiloxane (PDMS) strip cutting, and glue preparation. We detail procedures for PDMS strip mounting, tissue compaction, and live imaging upon force application. For additional details

Long-lasting biofouling formation on transparent fouling-release coatings for the construction of efficient closed photobioreactors.

In order to build an efficient closed-photobioreactor (PBR) in which biofouling formation is avoided, a non-toxic coating with high transparency is required, which can be applied to the interior surface of the PBR walls. Nowadays, amphiphilic copolymers are being used to inhibit microorganism adhesion, so poly(dimethylsiloxane)-based coatings mixed with poly(ethylene glycol)-based copolymers could be a good option. The 7 poly(dimethylsiloxane)-based coatings tested in this work contained 4% w/w of poly(ethylene glycol)-based copolymers. All were a good alternative to glass because they present

Anti-marine biofouling adhesion performance and mechanism of PDMS fouling-release coating containing PS-PEG hydrogel.

Polystyrene microspheres compounded with polyethylene glycol-based hydrogel (PS-PEG)/polydimethylsiloxane (PDMS) coatings were prepared using the physical blending method. The chemical structure, surface and interface properties, interlayer adhesion, and tensile properties were tested in this paper. Furthermore, the antifouling performance was evaluated through bovine serum albumin fluorescent protein adsorption testing, marine bacteria adhesion testing, and benthic diatom adhesion testing. The results showed that the coating performance was best when 20 wt% PS-PEG hydrogel was added. Its surf

Increased efficiency of charge-mediated fusion in polymer/lipid hybrid membranes.

Due to their augmented properties, biomimetic polymer/lipid hybrid compartments are a promising substitute for natural liposomes in multiple applications, but the protein-free fusion of those semisynthetic membranes is unexplored to date. Here, we study the charge-mediated fusion of hybrid vesicles composed of poly(dimethylsiloxane)-graft-poly(ethylene oxide) and different lipids and analyze the process by size distribution and the mixing of membrane species at μm and nano scales. Remarkably, the membrane mixing of oppositely charged hybrids surpasses by far the degree in liposomes, which we c

Injectable polydimethylsiloxane microfiller coated with zwitterionic polymer for enhanced biocompatibility.

Silicone-based fillers have been applied in several branches of medicine, such as soft tissue augmentation, because of their stability and durability. However, the inherently hydrophobic surfaces of silicone occasionally cause excessive deposition of the fibrous matrix in vivo, leading to severe fibrosis. In this study, we evaluated the use of a zwitterionic copolymer to offer a facile surface treatment method for silicone-based fillers and performed a preclinical trial of the formulation as-prepared. The copolymer has amphiphilic moieties, which act as macromolecular surfactants that can func

Nanoconfinement Controls Mechanical Properties of Elastomeric Thin Films.

Mastering mechanical properties of polymers at nanometer scale is highly demanded yet remains challenging. Pioneering advances determined Young's modulus in ultrathin polymer films and attained unprecedented results including rubbery stiffening. However, many viscoelastic properties such as dynamic mechanical behavior of freestanding nanoconfined polymer films are still unknown. Here we demonstrate striking changes of stiffness and the ratio between elastic and viscous responses in thin PDMS films, using a microvibrational system which enables direct measurements of dynamic stress-strain relat

Preparation of an AgNPs@Polydimethylsiloxane (PDMS) multi-hole filter membrane chip for the rapid identification of food-borne pathogens by surface-enhanced Raman spectroscopy.

The consumption of food infected with food-borne pathogens has become a global public health problem. Therefore, it is monitor food-borne infections to avoid health and financial consequences. The rapid detection and differentiation of bacteria for biomedical and food safety applications continues to be a significant challenge. Herein, we present a label-free surface-enhanced Raman scattering approach for separating harmful bacteria from food. The method relies on the ascorbic acid reduction method to synthesize silver nanoparticles (AgNPs) and a polydimethylsiloxane (PDMS) multi-hole filter m

Transoral laser microsurgery for polydimethylsiloxane (VOX Implant®) explantation.

INTRODUCTION: Polydimethylsiloxane (VOX Implant®) injection into the larynx is a surgical technique used to restore volume to paralysed or atrophic vocal folds. We present a case series that demonstrates the feasibility of explantation of this non-resorbable material in the event of complications secondary to this injection. DESCRIPTION: We report four cases of VOX Implant® explantation. In every case, explantation was performed following complications such as dysphonia and/or dyspnoea secondary to injection of this material. VOX Implant® explantation was performed via cordotomy using transor

Protocol for microfluidic-based high-precision general polarization fluorescence microscopy of lipid packing in membrane vesicles.

Lipid packing plays a critical role in membrane organization and function, but its precise quantification remains challenging in fluidic environments. Here, we present a protocol to integrate confocal fluorescence generalized polarization (GP) imaging with a polydimethylsiloxane (PDMS)-based microfluidic platform. We describe steps for lipid preparation, giant unilamellar vesicle (GUV) fabrication, microfluidic trapping, and GP analysis using polarity-sensitive dyes. This protocol enables robust measurement of membrane order and is readily adaptable to diverse lipid compositions and aqueous en

From Fiber Bundles to Architected Membranes: Triply Periodic Minimal Surface Architectures for Biohybrid Artificial Lungs.

Artificial lung systems rely almost exclusively on hollow fiber membrane (HFM) bundles, where gas exchange is constrained by heterogeneous flow distribution and thrombogenic blood-material interfaces. Here, we introduce an architecture-driven design framework for artificial lungs based on additively manufactured triply periodic minimal surface (TPMS) membranes. In contrast to discrete fiber bundles, TPMS membranes form continuous three-dimensional architectures that simultaneously regulate perfusion pathways, diffusion interfaces, and blood-material interactions. Computational fluid dynamics a

Generation of vascularized retinal organoids containing microglia based on a PDMS microwell platform.

Retinal organoids (ROs) offer a biomimetic in vitro model for investigating human retinal development and disease. However, current ROs face several limitations, such as the absence of vascular networks and microglial cells (MGs). Here, we developed a vascularized retinal organoids (vROs) model by coculturing vascular organoids (VOs) with ROs in a V-bottom polydimethylsiloxane (PDMS) microwell platform. Through coculturing for 30 to 120 days, we observed the presence of tubular blood vessels at the center of vROs. Transcriptomic analysis revealed that the vascularization in ROs was associated

A comprehensive protocol for PDMS fabrication for use in cell culture.

Cells exhibit remarkable sensitivity to the mechanical properties of their surrounding matrix, particularly stiffness changes, a phenomenon known as cellular mechanotransduction. In vivo, tissues exhibit a wide range of stiffness, from kilopascals (kPa) to megapascals (MPa), which can alter with aging and disease. Traditional cell culture methods employ plastic substrates with stiffness in the gigapascal range, which does not accurately mimic the physiological conditions of most biological tissues. Therefore, employing substrates that can be engineered to span a wide range of stiffnesses, clos

A type of multifunctional cellulose nanocrystal composite silicone-based polymer coating for marine antibiofouling.

Nanocomposite polymer coatings are being used as a new generation of marine antibiofouling coatings because of their toxin-free chemical composition and ease of large-scale adoption. Cellulose nanocrystal (CN) exhibits significant potential for composite reinforcement. Herein, CN was surface-modified via α,ω-bis(3-(2-hydroxyl-terminated polydimethylsiloxane (HTPDMS), resulting in dihydroxyl-terminated poly(dimethylsiloxane)-grafted CN (HP-g-CN). The amine-terminated PDMS as the foundational component was sequentially reacted with isophorone diisocyanate, isophthalaldehyde, and carbon disulfide

Surface photosterilization of implantable silicone biomaterials: structural and functional characterization.

Hospital-acquired infections (HAIs) remain one of the major challenges faced by the global healthcare system. The increasing rate of pathogenic resistance against antibiotics suggests that alternative treatments are needed to control recurrent infections. Catheter-associated urinary tract infections (CAUTIs) are the third most common type of HAI worldwide, and this is mainly due to indwelling devices being excellent substrates for bacterial adhesion and growth. Subsequent biofilm formation on the implant surface acts as a constant nidus of bacteria and infection, thereby contributing to increa

Contact photolithography-free integration of patterned and semi-transparent indium tin oxide stimulation electrodes into polydimethylsiloxane-based heart-on-a-chip devices for streamlining physiological recordings.

Controlled electrical stimulation is essential for evaluating the physiology of cardiac tissues engineered in heart-on-a-chip devices. However, existing stimulation techniques, such as external platinum electrodes or opaque microelectrode arrays patterned on glass substrates, have limited throughput, reproducibility, or compatibility with other desirable features of heart-on-a-chip systems, such as the use of tunable culture substrates, imaging accessibility, or enclosure in a microfluidic device. In this study, indium tin oxide (ITO), a conductive, semi-transparent, and biocompatible material

Long-term Outcomes of Silastic Arthroplasty of the Thumb Metacarpophalangeal Joint.

Background: Silastic metacarpophalangeal arthroplasty (SMPA) has proven to be a durable option for end-stage arthritis in the non-thumb digits, while fusion has been the mainstay procedure for the thumb metacarpophalangeal joint (MP). Few studies exist to comment on the viability of thumb MP arthroplasty. This study reports both survival and objective outcomes following SMPA of the thumb. Methods: In an institutional review board-approved retrospective study, we identified 18 patients who underwent thumb SMPA at a tertiary academic center by 3 board-certified hand surgeons. Primary outcome mea
