Two isoenzymes of carbonate dehydratase were identified in green leaf tissue of Lactuca sativa. Their molecular weights were found to be 195 000 and 250 000. The lighter isoenzyme (I) was further characterized. It is localized in the chloroplast fraction. With polyacrylamide gel electrophoresis in the presence of dodecyl sulphate, subunits with a molecular weight of 34 000 and higher aggregates of this size could be detected. The standard method for estimating cyanide liberated from sodium nitroprusside (SNP) has been colorimetric. Using a cyanide ion-selective electrode technique, it was found that SNP remained unchanged in the presence of whole blood, plasma and washed erythrocytes, no cyanide being detected. Previously reported releases of cyanide from SNP in vivo and in vitro are probably inaccurate. Values obtained could have arisen not from reaction with blood, but from photo-decomposition during the lengthy analysis required by the colorimetric method or during infusion, if the solution was left uncovered. Based on average mean concentrations compared with all-method mean concentration, hexokinase methods are the most accurate. Silver stainability of nucleolus organizer regions (NORs) appears to be correlated with the presence of grouped sulfhydryl (SH) side chains of proteins. In fact, heavy metals with high affinity for SH groups, such as Hg and Cu, do prevent the silver staining reaction. A quantitative cytochemical method for the measurement of beta-galactosidase activity in cultured human skin fibroblasts has been developed using 5-bromo-4-chloro-3-indolyl-beta-D-galactopyranoside as the indigogenic substrate. Needle-shaped cytoplasmic liver cell inclusions are considered to represent a feature typical of porphyria cutanea tarda (PCT). Histological sections of 849 liver biopsies were stained with ferricyanide reduction reaction for demonstration of the inclusions. Cytochrome cd1 from the denitrifying bacterium Pseudomonas aeruginosa catalyzes the oxygenation of carbon monoxide by dioxygen. The use of OHB12 in the prophylaxis of CN intoxication resulting from SNP infusions is well documented in the literature. However, data is lacking to support its use as an antidote once signs and symptoms have developed. Because of the efficacy of oxygen therapy combined with supportive therapy and nitrite/sodium thiosulfate administration, the use of OHB12 as an antidote cannot be recommended at this time. Further studies are needed to determine efficacy, dosing, mode of administration and comparative trials to other antidotes.
