Coagulation studies after transfusion of hydroxyethyl starch protected frozen blood in primates.

This-study evaluates the effects of the transfusion of frozen-thawed blood protected by hydroxyethyl starch (HES) on shocked monkeys. Particular attention is directed toward the study of coagulation in view of previous reports of coagulopathies caused by hydroxyethyl starch. In vivo studies were done in 13 shocked monkeys that received either homologous or autologous blood which had been frozen and thawed with 14 per cent HES as the cryoprotective agent. Multiple coagulation studies were obtained on the monkeys, and histopathological evaluations were performed. There was no evidence of increas

Electron microscopic study of the effects of calcitonin on bone cells and their extracellular milieu.

This is a scanning and transmission electron microscopic study of the changes produced by small doses of calcitonin (0.15 mU/g body weight) in the lining cells and their microenvironment at the endosteal surfaces of the tibia of neonate rats. The techniques used included "freeze substitution" preparation, staining with lead acetate, and with lanthanum added to the fixative. Rats were sacrificed 5, 15, and 30 minutes after subcutaneous injection of salmon calcitonin. The following observations were made; within 5 minutes following calcitonin injection, the response of the endosteal lining cells…

Acellular amniotic membrane: an appropriate scaffold for fibroblast proliferation.

BACKGROUND: Various materials have been investigated as possible skin substitutes to repair skin defects such as burns. Because of its unique characteristics, acellular amniotic membrane seems to provide a good scaffold for cell cultures. AIM: To investigate the proliferation of fibroblasts on an amniotic membrane scaffold, and the preparation of a temporary skin substitute using this method. METHODS: Neonatal foreskin tissue was harvested after circumcision and used for isolation of skin fibroblasts. The skin sample was refrigerated in cell-culture solution, and later treated with trypsin,

Cryotherapy of the liver: a histological review.

INTRODUCTION: Cryotherapy has been largely used in the past for palliation of unresectable liver tumors, but high local recurrence rates and peculiar systemic complications have determined its progressive abandonment. This review analyzes the histological changes produced to provide the basis for the local recurrences. MATERIALS AND METHODS: A detailed literature search was performed on studies focusing on liver cryotherapy. Included were only those that described the histological characteristics in detail. RESULTS: A total of 22 studies were found, one clinical and the others in-vivo animal

Examination of murine tear film.

PURPOSE: To define spatially any free aqueous layer in murine tear film. METHOD: A pre-zeroed microelectrode was touched to the superficial corneal epithelium and then raised in steps of 1 micro m through the murine tear film into the air and then retraced along the same path. Other murine tear films were partially probed with a spatial resolution of 0.1 micro m. The reference microelectrode was placed in a fragment of 3% polyacrylamide gel equilibrated against 154 mM NaCl and located on the nasal quadrant of the scleral conjunctiva. Other murine corneas were quick frozen in melting isopentan

Cryofixation, cryosubstitution, and immunoelectron microscopy: potential role in diagnostic pathology.

In immunoelectron microscopic investigations, retention of antigenic sites is crucial. Methods for preparing samples for conventional electron microscopy involve chemical fixatives followed by dehydration in organic solvents and embedding in plastic resins, all procedures potentially detrimental to antigenicity. Cryomethods provide a physical fixation alternative for the preparation of biological samples for ultrastructural, immunocytochemical, and microanalysis studies without the use of any chemicals. This can be particularly useful in diagnostic pathology, providing an alternative to conven

Effect of different resuspension media on the post-thaw characteristics of frozen blood.

Red blood cells frozen by the low-glycerol fast-freezing technique were thawed, deglycerolized and resuspended in various media. The use of ACD-saline for resuspension markedly reduced in vitro haemolysis such that the red cells could be transfused up to 5 d after thawing. At this time the cells contained satisfactory levels of potassium and organic phosphates, while bacterial contamination was negligible. For the past 4 years we have operated a small bank of previously frozen red blood cells which were thawed and resuspended in this way. Over 3500 units have been transfused after 1--5 d post-

Hybrid gelatin-sulfated alginate scaffolds as dermal substitutes can dramatically accelerate healing of full-thickness diabetic wounds.

Diabetic foot ulcers (DFUs) are defined as chronic and non-healing wounds that cause skin disorders. Here, we introduce a novel biodegradable gelatin/sulfated alginate hybrid scaffold as a dermal substitute to accelerate the healing of full-thickness diabetic ulcers in a diabetic mouse model. The hybrid scaffold possessing different weight ratios of sulfated alginate, from 10 % up to 50 %, were prepared through chemical crosslinking by carbodiimide chemistry and further freeze-drying. Based on the in vitro cytotoxicity experiments, the hybrid scaffolds not only showed no cytotoxicity, but the

Bilateral maxillary sinus floor augmentation with tissue-engineered autologous osteoblasts and demineralized freeze-dried bone.

The pneumatization of the maxillary sinus often results in a lack of sufficient alveolar bone for implant placement. In the last decades, maxillary sinus lift has become a very popular procedure with predictable results. Sinus floor augmentation procedures are generally carried out using autologous bone grafts, bone substitutes, or composites of bone and bone substitutes. However, the inherent limitations associated with each of these, have directed the attention of investigators to new technologies like bone tissue engineering. Bone marrow stromal cells have been regarded as multi-potent cell

Occipitocervical or C1-C2 fusion using allograft bone in pediatric patients with Down syndrome 8 years of age or younger.

Sufficient internal fixation for occipitocervical (OC) or C1-C2 instability in pediatric patients with Down syndrome is difficult owing to small osseous structures, congenital deformities, and immature ossification. The purpose of this study was to evaluate the clinical outcomes of patients aged 8 years or younger with Down syndrome who underwent OC or C1-C2 fusion using freeze-dried allograft as bone graft substitute. The participants included seven consecutive patients aged 8 years or younger with Down syndrome who were treated for upper cervical disorders with posterior OC or C1-C2 fusion u

Alveolar ridge preservation using an open membrane approach for sockets with bone deficiency: A randomized controlled clinical trial.

BACKGROUND: Various approaches are used for alveolar ridge preservation (ARP); however, there is no standard method or material. PURPOSE: To investigate the effect of ARP with a dense polytetrafluoroethylene (d-PTFE) membrane and freeze-dried irradiated allogenic bone for sockets with bone deficiency. MATERIALS AND METHODS: Thirty-four patients (with sockets exhibiting ≥3 mm hard tissue loss in ≥1 walls) were randomized to undergo natural socket healing (control) or ARP with a d-PTFE membrane and freeze-dried irradiated allogenic bone (test group). After 4 months, horizontal and vertical rid

The effects of preservation procedures on antibacterial property of amniotic membrane.

Amniotic membrane (AM), the innermost layer of the fetal membranes, has been widely employed in the surgical reconstruction and tissue engineering. Expression of the antimicrobial peptides such as defensins, elafin and SLPI which are essential elements of the innate immune system results in antibacterial properties of the AM. Preservation is necessary to reach a ready-to-use source of the AM. However, these methods might change the properties of the AM. The aim of this study was to evaluate antibacterial properties of the AM after preservation. Antibacterial property of the fresh AM was compar
