Structural and functional analysis of EntV reveals a 12 amino acid fragment protective against fungal infections.

Fungal pathogens are a continuing challenge due to few effective antifungals and a rise in resistance. In previous work, we described the inhibition of Candida albicans virulence following exposure to the 68 amino acid bacteriocin, EntV, secreted by Enterococcus faecalis. Here, to optimize EntV as a potential therapeutic and better understand its antifungal features, an X-ray structure is obtained. The structure consists of six alpha helices enclosing a seventh 16 amino acid helix (α7). The individual helices are tested for antifungal activity using in vitro and nematode infection assays. Inte

Local Population Structure and Patterns of Western Hemisphere Dispersal for Coccidioides spp., the Fungal Cause of Valley Fever.

UNLABELLED: Coccidioidomycosis (or valley fever) is a fungal disease with high morbidity and mortality that affects tens of thousands of people each year. This infection is caused by two sibling species, Coccidioides immitis and C. posadasii, which are endemic to specific arid locales throughout the Western Hemisphere, particularly the desert southwest of the United States. Recent epidemiological and population genetic data suggest that the geographic range of coccidioidomycosis is expanding, as new endemic clusters have been identified in the state of Washington, well outside the established

Structural insights into the substrate specificity and transglycosylation activity of a fungal glycoside hydrolase family 5 β-mannosidase.

β-Mannosidases are exo-acting glycoside hydrolases (GHs) that catalyse the removal of the nonreducing end β-D-mannose from manno-oligosaccharides or mannoside-substituted molecules. They play important roles in fundamental biological processes and also have potential applications in various industries. In this study, the first fungal GH family 5 β-mannosidase (RmMan5B) from Rhizomucor miehei was functionally and structurally characterized. RmMan5B exhibited a much higher activity against manno-oligosaccharides than against p-nitrophenyl β-D-mannopyranoside (pNPM) and had a transglycosylation a

Stage-Specific Differences in Fungal Community Structure and Functional Potential During Litter Decomposition in a Lava Plateau.

Litter-inhabiting fungi drive organic matter mineralization, regulate nutrient cycling, and support ecosystem stability. Understanding their dynamics in unique geological habitats is essential for predicting ecological recovery on volcanic landforms. Using high-throughput ITS sequencing and physicochemical analyses, we investigated litter-inhabiting fungal communities across four stand types on the Jingpo Lake lava plateau-shrub forest (SF), deciduous broad-leaved forest (DB), coniferous and broad-leaved mixed forest (CB), and coniferous forest (CF)-at the early (t1) and late (t2) stages of de

Structural basis for sensitivity and acquired resistance of fungal cap guanine-N7 methyltransferases to the antifungal antibiotic sinefungin.

The essential enzyme messenger RNA (mRNA) (guanine-N7) methyltransferase catalyzes S-adenosylmethionine (SAM)-dependent conversion of GpppRNA ends to the m7GpppRNA cap structure characteristic of eukaryal mRNAs. The antibiotic sinefungin (SFG) is a SAM analog in which the S-CH3 sulfonium moiety of SAM is replaced by a C-NH2 amine. Available evidence indicates that the antifungal activity of SFG is exerted via inhibition of fungal cap methyltransferase Abd1. Here we report that recombinant Kluyveromyces lactis and Saccharomyces cerevisiae Abd1 are 240-fold and 485-fold more sensitive to inhibit

A chemical screen identifies structurally diverse metal chelators with activity against the fungal pathogen Candida albicans.

Candida albicans, one of the most prevalent human fungal pathogens, causes diverse diseases extending from superficial infections to deadly systemic mycoses. Currently, only three major classes of antifungal drugs are available to treat systemic infections: azoles, polyenes, and echinocandins. Alarmingly, the efficacy of these antifungals against C. albicans is hindered both by basal tolerance toward the drugs and the development of resistance mechanisms such as alterations of the drug's target, modulation of stress responses, and overexpression of efflux pumps. Thus, the need to identify nove

A Structure-Aware Convolutional Neural Network for Automatic Diagnosis of Fungal Keratitis with In Vivo Confocal Microscopy Images.

Fungal keratitis (FK) is a common and severe corneal disease, which is widely spread in tropical and subtropical areas. Early diagnosis and treatment are vital for patients, with confocal microscopy cornea imaging being one of the most effective methods for the diagnosis of FK. However, most cases are currently diagnosed by the subjective judgment of ophthalmologists, which is time-consuming and heavily depends on the experience of the ophthalmologists. In this paper, we introduce a novel structure-aware automatic diagnosis algorithm based on deep convolutional neural networks for the accurate

In vivo structural modification of type II arabinogalactans with fungal endo-β-1, 6-galactanase in Arabidopsis.

Arabinogalactan-proteins (AGPs) are mysterious extracellular glycoproteins in plants. Although AGPs are highly conserved, their molecular functions remain obscure. The physiological importance of AGPs has been extensively demonstrated with β-Yariv reagent, which specifically binds to AGPs and upon introduction into cells, causes various deleterious effects including growth inhibition and programmed cell death. However, structural features of AGPs that determine their functions have not been identified with β-Yariv reagent. It is known that AGPs are decorated with large type II arabinogalactans

Spatial genetic structure of a symbiotic beetle-fungal system: toward multi-taxa integrated landscape genetics.

Spatial patterns of genetic variation in interacting species can identify shared features that are important to gene flow and can elucidate co-evolutionary relationships. We assessed concordance in spatial genetic variation between the mountain pine beetle (Dendroctonus ponderosae) and one of its fungal symbionts, Grosmanniaclavigera, in western Canada using neutral genetic markers. We examined how spatial heterogeneity affects genetic variation within beetles and fungi and developed a novel integrated landscape genetics approach to assess reciprocal genetic influences between species using co

Structure of a rice beta-glucanase gene regulated by ethylene, cytokinin, wounding, salicylic acid and fungal elicitors.

A rice beta-glucanase gene was sequenced and its expression analyzed at the level of mRNA accumulation. This gene (Gns1) is expressed at relatively low levels in germinating seeds, shoots, leaves, panicles and callus, but it is expressed at higher levels in roots. Expression in the roots appears to be constitutive. Shoots express Gns1 at much higher levels when treated with ethylene, cytokinin, salicylic acid, and fungal elicitors derived from the pathogen Sclerotium oryzae or from the non-pathogen Saccharomyces cereviseae. Shoots also express Gns1 at higher levels in response to wounding. Exp

Single-season responses of rhizosphere fungal community structure and diversity to combined application of different organic materials in potato.

Long-term application of chemical fertilizers can lead to a decline in soil microbial diversity and an imbalance in community structure, thereby constraining the sustainable development of the potato industry. In this study, a single-season field experiment was conducted in Yongdeng County, Lanzhou City, Gansu Province, with four treatments: chemical fertilizer alone (CF), sheep manure combined with chemical fertilizer (T1), biochar combined with chemical fertilizer (T2), and corn stover combined with chemical fertilizer (T3). High-throughput sequencing was employed to investigate the single-s

Investigation of the effects of polycations on Candida albicans membrane models and fungal cells. The effect of polymer structure and molecular weight.

Cationic polymers (polycations) represent a promising class of antimicrobial compounds whose physicochemical and biological properties can be tailored through appropriate structural design. The positive charge of these macromolecules indicates possible interactions with biological membranes. In this work, polycations differing in chemical structure (poly(2-(dimethylamino)ethyl methacrylate (PDMAEMA), poly(3-methacrylamido propyl trimethyl ammonium chloride) (PMAPTAC)) and molecular weight were investigated for their antifungal activity. The studies focused on the interactions of the polycation

Discovery of Potential Fungal Dihydroorotate Dehydrogenase Inhibitors Via Structure-based Virtual Screening for Plant Disease Control.

Dihydroorotate dehydrogenase (DHODH), a key enzyme in the de novo pyrimidine biosynthesis pathway of phytopathogenic fungi, has emerged as a promising target for fungicide discovery. In this study, compound VS-24 was identified from a commercial chemical library using molecular docking-based virtual screening and bioactivity assays. Antifungal tests demonstrated that at a concentration of 50 mg/L, VS-24 has broad-spectrum inhibitory effects with inhibition rates of 96.0%, 76.7%, 75.3%, and 76.5% against Valsa mali, Botrytis cinerea, Gaeumannomyces graminis, and Magnaporthe oryzae, respectively

Fungal-augmented biofertilizer increases soil nutrient availability, alters rhizosphere community structure, and improves plant growth.

Converting lignocellulosic waste into biofertilizers offers a sustainable practice for waste valorization, yet the mechanistic links underpinning how such amendments affect the soil-plant system remain poorly elucidated. We hypothesized that fungal-augmented biofertilizers improve plant growth through direct nutrient release and shifts in rhizosphere microbial community composition. To test this hypothesis, we isolated a white-rot fungus, Bjerkandera adusta strain QX3-19, which exhibited high ligninolytic activity. Inoculation with QX3-19 during composting of forest residues substantially enha

Effect of Intensified Fermentation with Wickerhamomyces anomalus on Fungal Community Structure of Fermented Grains and Flavor Compounds of Xiaoqu Baijiu.

In order to explore the effect of Wickerhamomyces on the production of flavor compounds in Xiaoqu Baijiu (XQBJ), this study examined the correlation between the fungal communities in Xiaoqu and ester compounds. It was hypothesized that Wickerhamomyces contributes to the aroma of Xiaoqu. Intensified fermentation methods were used to validate the role of Wickerhamomyces anomalus in XQBJ brewing. Compared to traditional Xiaoqu, intensified fermentation significantly increased the acidity of fermented grains and starch utilization rate (p < 0.05). The fungal communities in fermented grains were an

New findings on ascus structure reveal forcible spore discharge mechanism in the fungal wheat pathogen Fusarium graminearum.

Fungi in the phylum Ascomycota disperse their sexual spores primarily by forcible discharge from the spore sac, the ascus, in which they develop. These fungi cause some of the most devastating diseases of agricultural crops worldwide. The mechanism of spore discharge is not well understood, despite its primary role in disease spread. Using confocal microscopy, we have elucidated the ascus structure and development as related to function for spore dispersal in Fusarium graminearum, a pathogen of small grains and corn. To determine the mechanism of spore discharge in this species, we examined as

The structural language of fungal cell wall sugars in WHO priority fungi: the determinant of pathogenicity and the gateway to new therapies.

Carbohydrates, far beyond their nutritional role, are fundamental biomolecules that encode biological information and mediate critical cellular interactions. In pathogenic fungi, cell wall (CW) glycans serve as both a protective armor and a dynamic interface with the host, directly influencing virulence and immune evasion. This review discusses the intricate architecture and composition of the CW in the four WHO-critical priority fungal pathogens: Aspergillus fumigatus, Candidozyma auris, Cryptococcus neoformans, and Candida albicans. We detail how distinct carbohydrate polymers, including α-
