Fusobacterium nucleatum infections: clinical spectrum and bacteriological features of 78 cases.

PURPOSE: Few series describe the clinical spectrum of Fusobacterium spp. infections. Among them, fewer discuss F. nucleatum, even though there are many clinical cases. METHODS: We performed a retrospective study over 8 years (from 2007 to 2014) in Limoges University Hospital, France, to assess clinical and bacteriological aspects of infections due to F. nucleatum. RESULTS: Eighty-one patients with F. nucleatum positive cultures were included in this study, irrespective of sample origin. Abscesses (n = 43), bacteraemia (n = 18) and bone infections (n = 8) were the most common types of infecti

Isolation of Primary RNA for Differential RNA Sequencing in Fusobacterium nucleatum.

Understanding gene expression in Fusobacterium nucleatum and related species is essential for uncovering the molecular mechanisms that underlie their adaption to various environments. However, the isolation of high-quality RNA from these Gram-negative anaerobes is challenging because of their cell wall structure and the limitations of conventional column-based RNA purification kits, particularly their transcript size bias. Here, we present a robust, phenol-based RNA extraction protocol optimized for F. nucleatum that consistently yields high-integrity, DNA-free RNA suitable for transcriptomic …

The Dual Face of Fusobacterium nucleatum in Cancer: Foe, Friend or Both?

Fusobacterium is a genus of anaerobic Gram-negative bacteria that has been increasingly implicated in a range of diseases, including periodontitis and cancer. This review critically evaluates the reported role of Fusobacterium nucleatum in cancer, highlighting new findings that suggest a more nuanced cross-talk with the disease. We contextualise current evidence on the interactions with the wider tumour micro-environment, including the roles of polymicrobial communities, microbial metabolites and taxonomic heterogeneity. Collectively, the evidence suggests that Fusobacterium nucleatum should n…

The role of adhesins of Fusobacterium nucleatum in colorectal cancer - a structural perspective.

Fusobacterium nucleatum is frequently found in the colon microbiome of colorectal cancer (CRC) patients. The bacterium is not only a passive bystander of CRC, but is actively involved in disease progression through mediating tumor growth stimulation, metastasis, and immune evasion. These outcomes are achieved through the action of several adhesins that are located on the outer membrane surface of F. nucleatum, which bind to different receptors on tumor or immune cells. Adhesin-receptor interaction as the initial step of host-pathogen interaction then triggers signal transduction pathways respo…

Ethanolamine-induced assembly of microcompartments is required for Fusobacterium nucleatum virulence.

UNLABELLED: Many bacteria metabolize ethanolamine as a nutrient source through cytoplasmic organelles named bacterial microcompartments (BMCs). Here we investigated the molecular assembly, regulation, and function of BMCs in Fusobacterium nucleatum-a Gram-negative oral pathobiont that is associated with adverse pregnancy outcomes. The F. nucleatum genome harbors a conserved ethanolamine utilization (eut) locus with 21 genes that encode several putative BMC shell proteins and a two-component signal transduction system (TCS), in addition to the enzymes for ethanolamine transport and catabolism. …

Investigation of Fusobacterium Nucleatum in saliva and colorectal mucosa: a pilot study.

As evidence has been linking the oral bacterium Fusobacterium nucleatum (F. nucleatum) to colorectal tumorigenesis, we aimed to produce preliminary data on the expression of F. nucleatum in both oral and colorectal body sites in cases diagnosed with colorectal neoplasms (CRN) and CRN-free controls. We conducted a pilot hospital-based case-control study among patients who underwent colonoscopy examination. Saliva samples and biopsies from healthy colon mucosa from CRN cases and CRN-free controls, and from tumors in cases, were collected, as well as data on periodontal condition and potential CR…

Tea polyphenols inhibit the growth and virulence properties of Fusobacterium nucleatum.

Fusobacterium nucleatum plays a key role in creating the pathogenic subgingival biofilm that initiates destructive periodontitis. It is also a common resident of the human gastrointestinal tract and has been associated with inflammatory bowel disease. The aim of the present study was to investigate the effects of green and black tea extracts as well as two of their bioactive components, EGCG and theaflavins, on the growth and virulence properties of F. nucleatum. The tea extracts and components displayed various degrees of antibacterial activity that may involve damage to the bacterial cell me…

Characteristics of Porphyromonas gingivalis lipopolysaccharide in co-culture with Fusobacterium nucleatum.

Porphyromonas gingivalis is associated with chronic periodontitis and forms multi-species biofilms. They can communicate within species as well as with other species found in the subgingiva, which may induce changes in the growth ratio and virulence of periodontopathogens. The lipopolysaccharide (LPS) of P. gingivalis shows different virulence by growth condition. The purpose of this study was to investigate the characteristics of P. gingivalis LPS when co-cultured with Fusobacterium nucleatum. After culture of P. gingivalis in the presence or absence of F. nucleatum, P. gingivalis LPS was ext…

Microbe-driven immune suppression in colorectal cancer: the Fusobacterium nucleatum playbook.

Fusobacterium nucleatum, a key oral-pathobiont in colorectal cancer (CRC), has evolved from a presumed "passenger" to an active "co-conspirator" in tumorigenesis. This review systematically delineates its core role as an "architect of the immune microenvironment," whereby it remodels T-cell immune responses through precise mechanisms to establish an immunosuppressive tumor microenvironment. Specifically, F. nucleatum employs virulence factors such as Fap2 and FadA for specific colonization, and achieves immune evasion by inhibiting NK and T-cell function and recruiting myeloid-derived suppress…

2'-Fucosyllactose Attenuates Fusobacterium nucleatum Virulence and Modulates the Oral Microbiota.

Fusobacterium nucleatum (F. nucleatum) is a key periodontal pathobiont associated with oral inflammation. This bacterium forms biofilms and expresses adhesins that facilitate its adhesion to and invasion of gingival epithelial cells. These processes disrupt the epithelial barrier and trigger oral inflammation, and in some cases, systemic inflammation. Conventional antimicrobial strategies predominantly depend on the utilization of antibiotics. Nevertheless, this can result in the proliferation of drug-resistant strains and the disruption of the oral microbiome equilibrium. As the predominant h…

Fusobacterium nucleatum infection leading to rare hepatorenal abscess: a case report.

Fusobacterium nucleatum is a gram-negative anaerobic bacterium commonly associated with periodontal disease. However, its role in extraoral infections, particularly in immunocompetent individuals, is increasingly recognized. We report a rare case of hepatorenal abscess caused by F. nucleatum in a previously healthy woman, initially suspected to have a malignant tumor based on PET-CT findings. Next-generation sequencing (NGS) of abscess aspirate confirmed the pathogen. The patient responded well to targeted antibiotic therapy. This case highlights the importance of considering anaerobic pathoge…

An Orbital Abscess Caused by Fusobacterium nucleatum Mimicking Tolosa-Hunt Syndrome.

Fusobacterium species, particularly Fusobacterium nucleatum, are anaerobic gram-negative bacteria found in the oral cavity and gastrointestinal tract. Infections caused by these organisms exhibit diverse clinical manifestations. Here we report a rare case of orbital abscess caused by Fusobacterium nucleatum, which was initially misdiagnosed as Tolosa-Hunt syndrome. The patient was controlled conservatively, and surgical intervention was withheld due to poor prognosis, diabetes, and logistical limitations. This case report highlights the need to consider infection even when imaging and laborato…

Effect of High Intensity Blue Light on Fusobacterium nucleatum Membrane Integrity.

Oral malodour is considered to be caused mainly by the production of volatile sulfide compounds (VSC) by anaerobic gram-negative oral bacteria. Previous studies showed that these bacteria were susceptible to blue light phototoxicity mediated by the production of reactive oxygen species (ROS). In the present study, we tested the effect of blue light on the integrity Fusobacterium nucleatum's membrane, cellular proteins and DNA. Bacterial samples were exposed to high intensity blue light for 0, 70, 140 and 280 s (i.e. fluences of 0, 96, 192 and 384 J cm-2 , respectively). Following light exposur…

Fusobacterium nucleatum exacerbates the progression of pulpitis by regulating the STING-dependent pathway.

Bacterial infection is the main cause of pulpitis. However, whether a dominant bacteria can promote the progression of pulpitis and its underlying mechanism remains unclear. We provided a comprehensive assessment of the microbiota alteration in pulpitis using 16S rRNA sequencing. Fusobacterium nucleatum was the most enriched in pulpitis and played a pathogenic role accelerating pulpitis progression in rat pulpitis model. After odontoblast-like cells cocultured with F. nucleatum, the stimulator of interferon genes (STING) pathway and autophagy were activation. There was a float of STING express…
