Tonic GABAergic inhibition of sympathetic preganglionic neurons: a novel substrate for sympathetic control.

The sympathetic tone is primarily defined by the level of activity of the sympathetic preganglionic neurons. We report a novel inhibitory influence on sympathetic activity, that of tonic GABAergic inhibition which could have a profound global effect on sympathetic outflow. Recording from identified SPNs in the intermediolateral cell column (IML) of rat spinal cord slices, application of the GABA receptor antagonist bicuculline, but not gabazine, elicited a change in voltage that lasted for the duration of application. This response was mediated by a direct effect on SPNs since it persisted in

Binge ethanol exposure delays development of GABAergic miniature postsynaptic currents in septal neurons.

Whole cell GABA(A)R currents of septal neurons isolated from rat pups increase rapidly during the first weeks of life when inhibitory synapses are forming. Early postnatal binge ethanol intubation on days 4-9 delays this maturational up-regulation in septal neurons isolated several days later suggesting inhibitory synapse formation could be disrupted [S.-H. Hsiao, J.L. Acevedo, D.W. DuBois, K.R. Smith, J.R. West, G.D. Neuronal hyperexcitability in dystrophin-deficient mdx hippocampal neurons: the importance of interleukin-6 and GABAergic regulation.

Duchenne Muscular Dystrophy (DMD) is a severe neuromuscular disorder arising from loss of the structural protein, dystrophin. It also often presents with cognitive deficits and susceptibility to epilepsy. Expressed in neurons of the hippocampus, dystrophin plays an important role in synapse formation, specifically the post-synaptic organisation of γ-aminobutyric acid A receptors (GABAARs). This study explored possible interactions between interleukin (IL)-6, which is elevated in DMD, and GABAAR signalling in cultured hippocampal neurons of dystrophic mdx mice. Immunofluorescent imaging reveale

RHO GTPase by L1 GABAergic neurons in frontal cortex and L6 glutamatergic neurons in prefrontal cortex differentiates states of unconsciousness.

Unconsciousness can be induced by sleep and general anaesthesia alike; however, the extent to which these states differ is not well understo … s of genes were enriched in the RHO GTPase pathway. Based on these results, we hypothesised that L1 GABAergic neurons in the frontal cortex and L6 glutamatergic neurons in the prefrontal cortex use RHO GTPase to differentiate between the states of unconsciousness induced by general anaesthesia and sleep.

GABAergic neuron fate specification and lineage allocation: from development to disorder.

The mammalian telencephalon contains molecularly and functionally diverse inhibitory neuron subtypes that are essential for balanced neural circuit function and dynamics. The developmental process that generates this diversity from undifferentiated neuronal progenitor cells is governed by cis-regulatory elements (CREs) and the transcription factors (TFs) that bind to them. In combination with associated transcription co-regulators and epigenetic regulators, these transcription factors form gene regulatory networks that control the establishment of lineage-specific patterns of gene expression,

The microcephaly gene Donson is essential for progenitors of cortical glutamatergic and GABAergic neurons.

Biallelic mutations in DONSON, an essential gene encoding for a replication fork protection factor, were linked to skeletal abnormalities an … kx2.1-Cre-mediated Donson deletion in subpallial progenitors ablated 75% of Nkx2.1-derived cortical GABAergic neurons. Thus, the early telencephalic neuroepithelium depends critically on Donson function. Our findings help explain why the neocortex is most severely affected in individuals with DONSON mutations and suggest that DONSON-dependent microcephaly might be …

Ptf1a, a bHLH transcriptional gene, defines GABAergic neuronal fates in cerebellum.

The molecular machinery governing glutamatergic-GABAergic neuronal subtype specification is unclear. Here we describe a cerebellar mutant, c … y. Introduction of Ptf1a into glutamatergic neuron precursors in the dorsal telencephalon generated GABAergic neurons with representative morphological and migratory features. Our results suggest that Ptf1a is involved in driving neural precursors to differentiate into GABAergic neurons in the cerebellum.

GABAergic nature of hypothalamic leptin target neurones in the ventromedial arcuate nucleus.

Leptin is an adipose tissue-derived cytokine hormone, which reduces body weight via interactions with hypothalamic neurones. Leptin receptors capable of activating the JAK-STAT signal transduction pathway are expressed at high levels in the hypothalamus, particularly in the arcuate nucleus. In order to identify the chemical mediators of leptin's action in the hypothalamus, we have examined whether GABA neurones of the hypothalamic arcuate nucleus contain leptin receptors and the leptin-activated transcription factor STAT3. GABAergic neurones, as visualized by antisera to the GABA-synthesizing

Post-tetanic potentiation of GABAergic IPSCs in cultured rat hippocampal neurones.

1. Dual whole-cell patch-clamp recording was used to investigate post-tetanic potentiation (PTP) of GABAergic IPSCs evoked between pairs of cultured rat hippocampal neurones. Tetanization of the presynaptic neurone at frequencies (f) ranging from 5 to 100 Hz resulted in PTP of the IPSCs. Maximum PTP had a magnitude of 51.6 % just after the stimulus train, and lasted up to 1 min. PTP was shown to be dependent on the number of stimuli in the train, but independent of f at frequencies > or =5 Hz. 2. Blocking postsynaptic GABAA receptors with bicuculline during the tetanus did not affect the expre

GABAergic neurons and circuits in the pretectal nuclei and the accessory optic system of mammals.

Two classes of GABAergic cell bodies have been described. They probably can be divided into GABAergic local interneurons and GABAergic projection neurons. GABAergic cell bodies receive few terminals which is in contrast to non-GABAergic somata, which receive many synaptic contacts. GABAergic dendrites that originate from GABAergic cell bodies, however, receive numerous terminals, both GABAergic and nonGABAergic. It can therefore be concluded that somatic inhibition is not present on GABAergic neurons, but does occur on nonGABAergic neurons. Furthermore, dendrites traverse large parts of the NO…

[Influence of GABAergic neurons in the brain on central effects of kainate on the cardiovascular system].

Microinjection of kainate (KA 0.125, 0.25, or 0.5 microgram), a special agonist for KA receptor, into rat lateral cerebral ventricle produced dose-dependent increases in blood pressure and heart rate. Pretreatment with semicarbazide (SCZ 140 mg/kg, ip), an inhibitor of GABA biosynthesis, or picrotoxin (PIC 1 mg/kg, iv), an antagonist for GABA, significantly enhanced the effects of KA on the cardiovascular system. These effects of KA were reduced by aminoxyacetic acid (AOAA 25 micrograms/rat, icv), an inhibitor of GABA aminotransferase. All these results described above imply that the function

Endocannabinoid metabolism at VTA GABAergic neurons gates motivation for social, but not food, reward.

The endocannabinoid system functions as a gatekeeper of motivational processes. Here, we show that CRISPR-mediated inhibition of the degrading enzyme for the brain endocannabinoid, 2-AG, at ventral tegmental area GABAergic interneurons promotes social, but not food-maintained, operant responding. These results reveal a region-specific role for endocannabinoid signaling in social motivation.

Mu-Opioids Suppress GABAergic Synaptic Transmission onto Orbitofrontal Cortex Pyramidal Neurons with Subregional Selectivity.

The orbitofrontal cortex (OFC) plays a critical role in evaluating outcomes in a changing environment. Administering opioids to the OFC can alter the hedonic reaction to food rewards and increase their consumption in a subregion-specific manner. However, it is unknown how mu-opioid signaling influences synaptic transmission in the OFC. Thus, we investigated the cellular actions of mu-opioids within distinct subregions of the OFC. Using in vitro patch-clamp electrophysiology in brain slices containing the OFC, we found that the mu-opioid agonist DAMGO produced a concentration-dependent inhibiti

Aberrant expression of S-SCAM causes the loss of GABAergic synapses in hippocampal neurons.

The duplication and deletion mutations of the S-SCAM/MAGI-2 gene are associated with schizophrenia and infantile spasms, respectively. S-SCAM is a unique synaptic scaffolding protein that localizes to both excitatory and GABAergic synapses. However, consequences of aberrant S-SCAM expression on GABAergic synapses is little studied. Here we report the effect of S-SCAM knockdown and overexpression on GABAergic synapses. S-SCAM knockdown in cultured hippocampal neurons caused a drastic loss of both pre- and post-synaptic components of GABAergic synapses, indicating its essential role in GABAergic

GABAergic synaptic inputs of locus coeruleus neurons in wild-type and Mecp2-null mice.

Rett syndrome is an autism spectrum disorder resulting from defects in the gene encoding the methyl-CpG-binding protein 2 (MeCP2). Deficiency of the Mecp2 gene causes abnormalities in several systems in the brain, especially the norepinephrinergic and GABAergic systems. The norepinephrinergic neurons in the locus coeruleus (LC) modulate a variety of neurons and play an important role in multiple functions in the central nervous system. In Mecp2(-/Y) mice, defects in the intrinsic membrane properties of LC neurons have been identified, while how their synaptic inputs are affected remains unclea

Acidosis and alkalosis impair brain functions through weakening spike encoding at cortical GABAergic neurons.

Acidosis and alkalosis, associated with metabolic disorders, lead to the pathological changes of cognition and behaviors in clinical practices of neurology and psychology. Cellular mechanisms for these functional disorders in the central nervous system remain unclear. We have investigated the influences of acidosis and alkalosis on the functions of cortical GABAergic neurons. Both acidosis and alkalosis impair the ability of encoding sequential spikes at these GABAergic neurons. The impairments of their spiking are associated with the increases of refractory periods, threshold potential and af…

Signaling mechanisms of angiotensin II-induced attenuation of GABAergic input to hypothalamic presympathetic neurons.

The hypothalamic paraventricular nucleus (PVN) is an important site for the regulation of sympathetic outflow. Angiotensin II (Ang II) can activate AT(1) receptors to stimulate PVN presympathetic neurons through inhibition of GABAergic input. However, little is known about the downstream pathway involved in this presynaptic action of Ang II in the PVN. In this study, using whole cell recording from retrogradely labeled PVN neurons in rat brain slices, we determined the signaling mechanisms responsible for the effect of Ang II on synaptic GABA release to spinally projecting PVN neurons.
