BACKGROUND AND OBJECTIVES: In December 2010 four, lions and one tiger died at the Tehran zoo. Out of all samples, Burkholderia mallei (causative agent of Glanders) was isolated just from ulcer sample of the tiger which was imported to Iran from Russia.
MATERIALS AND METHODS: One nasal swab from a tiger and fifteen blood samples with anticoagulant belonging to one tiger and fourteen lions (four dead lions and eleven live lions) were collected and were inoculated directly onto the selective media. The isolate was identified by morphological and biochemical and API BBL tests and PCR using specif…

Glanders is a zoonotic infection inducing acute forms of the disease (pneumonia, sepsis) in humans and animals under certain conditions, which even with the use of modern chemotherapy have unfavourable prognosis. Insufficient of efficacy of antibiotics with in vitro low MIC for planktonic bacterial suspension of Burkholderia mallei in chemotherapy of acute forms of glanders was due to the capacity of the pathogen for intracellular survival and formation of biofilms. Under such conditions the susceptibility of B. mallei to antibiotics lowered by several orders of magnitude. Chemotherapy of the …

Burkholderia mallei, the etiologic agent of the disease known as glanders, is primarily a disease affecting horses and is transmitted to humans by direct contact with infected animals. The use of B. mallei as a biological weapon has been reported and currently, there is no vaccine available for either humans or animals. Despite the history and highly infective nature of B. mallei, as well as its potential use as a bio-weapon, B. mallei research to understand the pathogenesis and the host responses to infection remains limited. Therefore, this minireview will focus on current efforts to elucida…

The pathomorphology and cell-mediated response of the body to Burkholderia mallei in laboratory animals, highly sensitive and resistant to these bacteria. In the comparative study of the pathomorphology and pathogenesis of glanders in golden hamsters and white rats quantitative and qualitative differences in the histogenesis of response reaction and the morphology of immunocompetent organs were established. Cell-mediated reactions play a greater role in the limitation of the early spread of B. mallei in the host body than antigen-mediated mechanisms.

Electron-microscopic and electron-cytochemical method were used to indicate the capability of Pseudomonas mallei (str. N 10230) to produce extracellular slime during the agent growth on the meat-peptone agar. In case of guinea pigs infection the agent forms a capsule that defends the pathogen from phagocytosis.

Double immunodiffusion in gel test was used to determine the antigenic composition of the preparations of membranes isolated from the lysozyme spheroplasts of glanders (strain No. 10230) and melioidosis (strain No. C-141) causative agents. The membranes of these microbes proved to contain antigens of cell walls, lipopolysaccharides and the thermolabile membrane antigen proper. A study of antimembrane sera in the agglutination and immunofluorescence tests demonstrated a heterogeneity of the glanders and melioidosis strains under study by the membrane thermolabile antigen.
