Micro-RNA-27a/b negatively regulates hepatic gluconeogenesis by targeting FOXO1.

In the context of hepatic insulin resistance, hepatic gluconeogenesis is abnormally increased, which results in increased hepatic glucose production and hyperglycemia, but the underlying mechanisms remain to be fully elucidated. Micro-RNAs (miRNAs) have been identified as critical regulators of diabetes and other metabolic disorders. In this study, we found that the expressions of miRNA-27 family members miRNA-27a and miRNA-27b (miR-27a/b) decreased significantly in the livers of diabetic mice. Moreover, the levels of miR-27a/b increased in the serum of patients with type 2 diabetes. Our prese…

Dapagliflozin Aggravates Renal Injury via Promoting Gluconeogenesis in db/db Mice.

BACKGROUND/AIMS: A sodium-glucose co-transporter-2 inhibitor dapagliflozin is widely used for lowering blood glucose and its usage is limite … tion was mainly derived from the disturbance of glucose homeostasis with elevated hepatic and renal gluconeogenesis induced by dapagliflozin. Although it had no effect on insulin sensitivity and glucose tolerance, dapagliflozin further induced the expression of gluconeogenic key rate-limiting enzymes through increasing the expression levels of FoxO1 in the kidney …

Targeting pyruvate carboxylase reduces gluconeogenesis and adiposity and improves insulin resistance.

We measured the mRNA and protein expression of the key gluconeogenic enzymes in human liver biopsy specimens and found that only hepatic pyruvate carboxylase protein levels related strongly with glycemia. We assessed the role of pyruvate carboxylase in regulating glucose and lipid metabolism in rats through a loss-of-function approach using a specific antisense oligonucleotide (ASO) to decrease expression predominantly in liver and adipose tissue. Pyruvate carboxylase ASO reduced plasma glucose concentrations and the rate of endogenous glucose production in vivo. Interestingly, pyruvate carbox

Loss of TR4 orphan nuclear receptor reduces phosphoenolpyruvate carboxykinase-mediated gluconeogenesis.

OBJECTIVE: Regulation of phosphoenolpyruvate carboxykinase (PEPCK), the key gene in gluconeogenesis, is critical for glucose homeostasis in response to quick nutritional depletion and/or hormonal alteration. RESEARCH DESIGN/METHODS AND RESULTS: Here, we identified the testicular orphan nuclear receptor 4 (TR4) as a key PEPCK regulator modulating PEPCK gene via a transcriptional mechanism. TR4 transactivates the 490-bp PEPCK promoter-containing luciferase reporter gene activity by direct binding to the TR4 responsive element (TR4RE) located at -451 to -439 in the promoter region. Binding to TR…

Hepatic gluconeogenesis and PDK3 upregulation drive cancer cachexia in flies and mice.

Cachexia, a severe wasting syndrome characterized by tumour-induced metabolic dysregulation, is a leading cause of death in people with cancer, yet its underlying mechanisms remain poorly understood. Here we show that a longitudinal full-body single-nuclei-resolution transcriptome analysis in a Drosophila model of cancer cachexia captures interorgan dysregulations. Our study reveals that the tumour-secreted interleukin-like cytokine Upd3 induces fat-body expression of Pepck1 and Pdk, key regulators of gluconeogenesis, disrupting glucose metabolism and contributing to cachexia. Similarly, in mo…

Role of small leucine zipper protein in hepatic gluconeogenesis and metabolic disorder.

Hepatic gluconeogenesis is the central pathway for glucose generation in the body. The imbalance between glucose synthesis and uptake leads to metabolic diseases such as obesity, diabetes, and cardiovascular diseases. Small leucine zipper protein (sLZIP) is an isoform of LZIP and it mainly functions as a transcription factor. Although sLZIP is known to regulate the transcription of genes involved in various cellular processes, the role of sLZIP in hepatic glucose metabolism is not known. In this study, we investigated the regulatory role of sLZIP in hepatic gluconeogenesis and its involvement …

Regulation of hepatic gluconeogenesis by nuclear factor Y transcription factor in mice.

Hepatic gluconeogenesis is essential to maintain blood glucose levels, and its abnormal activation leads to hyperglycemia and type 2 diabetes. However, the molecular mechanisms in the regulation of hepatic gluconeogenesis remain to be fully defined. In this study, using murine hepatocytes and a liver-specific knockout mouse model, we explored the physiological role of nuclear factor Y (NF-Y) in regulating hepatic glucose metabolism and the underlying mechanism. We found that NF-Y targets the gluconeogenesis pathway in the liver. Hepatic NF-Y expression was effectively induced by cAMP, glucagon…

Changes in liver gluconeogenesis during the development of Walker-256 tumour in rats.

Few studies have investigated liver gluconeogenesis in cancer and there is no agreement as to whether the activity of this pathway is increased or decreased in this disease. The aim of this study was to evaluate gluconeogenesis from alanine, pyruvate and glycerol, and related metabolic parameters in perfused liver from Walker-256 tumour-bearing rats on days 5 (WK5 group), 8 (WK8 group) and 12 (WK12 group) of tumour development. There was reduction (P < 0.05) of liver glucose production from alanine and pyruvate in WK5, WK8 and WK12 groups, which was accompanied by a decrease (P < 0.05) in oxyg…

Zonation of the action of glucagon on gluconeogenesis studied in the bivascularly perfused rat liver.

We have measured the action of glucagon, infused into the hepatic artery, on gluconeogenesis from lactate in the rat liver, bivascularly perfused in both the anterograde and retrograde modes. Concerning glucose production and oxygen uptake per unit cell space, the response of the periportal cells reached via the hepatic artery in retrograde perfusion to glucagon is superior to the response of the cells reached via the same vessel in anterograde perfusion. This phenomenon, however, most probably reflects zonation of gluconeogenesis rather than zonation of the hormonal action. The latter conclus…

[Characteristics of the normal gluconeogenesis effect of prostaglandin F2 alpha and in myocardial infarct].

The effect of PGF2 alpha on glucose synthesis de novo in a healthy rat organism and those with coronary occlusion-myocardial infarction was studied. There was observed prostaglandin's metabolic action simultaneously at one direction: there was increased the concentration of non-nitric precursors of gluconeogenesis in blood of animals of both groups, final disintegration product of tissue proteins, the gluconeogenic activity of key enzymes and therefore the concentration of newly formed glucose went up as so as glycogen in liver, cardiac and skeletal muscle. Seemingly, PGF2 alpha stimulates the…

FoxO transcription factors coordinate the urea cycle and gluconeogenesis by controlling Ass1.

Amino acid catabolism during fasting requires coordinated nitrogen disposal and glucose production, but the transcriptional logic linking the urea cycle to gluconeogenesis remains unclear. Forkhead box O (FoxO) transcription factors regulate fasting metabolism, and here we identify FoxOs as direct hepatic regulators of argininosuccinate synthase 1 (Ass1). Acute hepatic FoxO1/3a knockdown in fasted mice reduced Ass1 expression, lowered blood glucose, and altered urea-cycle amino acids, with decreased arginine and increased ornithine, even in Klf15-deficient livers. Ass1 silencing phenocopied th…

A genome-wide siRNA screen to identify modulators of insulin sensitivity and gluconeogenesis.

BACKGROUND: Hepatic insulin resistance impairs insulin's ability to suppress hepatic glucose production (HGP) and contributes to the development of type 2 diabetes (T2D). Although the interests to discover novel genes that modulate insulin sensitivity and HGP are high, it remains challenging to have a human cell based system to identify novel genes. METHODOLOGY/PRINCIPAL FINDINGS: To identify genes that modulate hepatic insulin signaling and HGP, we generated a human cell line stably expressing beta-lactamase under the control of the human glucose-6-phosphatase (G6PC) promoter (AH-G6PC cells)
