Characterization of Three Different Endolysins Effective against Gram-Negative Bacteria.

Genes encoding endolysins were identified and cloned from three different Escherichia coli bacteriophages, 10-24(13), PBEC30, and PBEC56. Putative antimicrobial peptide (AMP)-like C-terminal alpha helix structures with amphipathic natures were predicted from the three endolysins. Each gene was cloned and expressed as hexahistidine-tagged forms, and the products were purified and characterized. The purified endolysins exhibited antibacterial activities against a variety of Gram-negative bacteria including Escherichia coli, Pseudomonas aeruginosa, Acinetobacter baumannii, and Klebsiella pneumoni…

Nasal antibodies against gram-negative bacteria in cotton-mill workers.

Antibody activity to cotton dust bacteria in the nasal secretions of cotton-mill cardroom workers was studied using the ELISA technique. Secretion samples were collected by inserting a roll of filter paper into the nasal cavities of the subjects. The amount of secretion was significantly less among cotton-mill workers than workers in a wood factory. No significant differences were seen between byssinotic and nonbyssinotic cotton-mill workers. No significant differences were observed between cotton and wood workers regarding antibody activity against Pseudomonas syringae and Enterobacter agglom

Moroccan Hospital Cockroaches: Carriers of Multidrug-Resistant Gram-Negative Bacteria.

Antimicrobial resistance in Gram-negative bacteria (GNB) is a growing global health concern, particularly in hospital environments, where cockroaches act as vectors for resistant strains. This study aimed to analyze antimicrobial resistance and biofilm formation in GNB isolated from cockroaches collected in the hospital environment. Cockroaches were collected, and bacterial isolation was performed from their gut contents and external surfaces. GNB strains were tested for antibiotic susceptibility using the disk diffusion method and examined for Extended-spectrum β-lactamases (ESBLs) and carbap…

Aminoglycoside-Peptide Nucleic Acid (PNA) Conjugates against Gram-Negative Bacteria.

Aminoglycosides (AGs) are among the earliest known classes of antibiotics. Despite decades of clinical utility, they have become largely ineffective due to the spread of antimicrobial resistance. In an effort to improve the activity of AGs against resistant strains, we conjugated them with antisense oligonucleotides, specifically peptide nucleic acids (PNAs). We report the synthesis and biological evaluation of novel neomycin (NEO) and amikacin (AMK) conjugates with 10-mer PNA oligomers targeting the essential bacterial gene encoding the acyl carrier protein. The conjugates were prepared via c

The respiratory threat posed by multidrug resistant Gram-negative bacteria.

Respiratory infections are a major cause of global mortality and morbidity. In recent years, an increased incidence of multidrug-resistant (MDR) Gram-negative bacteria (GNB) has been described. Microorganisms such as Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae or Acinetobacter baumannii have been identified as causative pathogens of different respiratory tract infections. Several studies have detected MDR-GNB in patients with community-acquired and nosocomial pneumonia. Furthermore, MDR-GNB have also been isolated in patients with chronic obstructive pulmonary disease and b…

[Recent progress in development of antibiotics against Gram-negative bacteria].

Multidrug-resistant (MDR) bacterial infections, especially those caused by Gram-negative pathogens, have emerged to be one of the world's greatest health threats. However, not only have recent decades shown a steady decline in the number of approved antimicrobial agents but a disappointing discovery also void. The development of novel antibiotics to treat MDR Gram-negative bacteria has been stagnated over the last half century. Though few compounds have shown activities in vitro, in animal models or even in clinical studies, the global antibiotic pipeline is not encouraging. There are a pletho…

Antibiotic resistance of gram-negative bacteria in rivers, United States.

Bacteria with intrinsic resistance to antibiotics are found in nature. Such organisms may acquire additional resistance genes from bacteria introduced into soil or water, and the resident bacteria may be the reservoir or source of widespread resistant organisms found in many environments. We isolated antibiotic-resistant bacteria in freshwater samples from 16 U.S. rivers at 22 sites and measured the prevalence of organisms resistant to beta-lactam and non-beta-lactam antibiotics. Over 40% of the bacteria resistant to more than one antibiotic had at least one plasmid. Ampicillin resistance gene

Tailoring the morphology and crystallinity of MoS2 for bacterial inhibition of Gram-negative and Gram-positive bacteria.

Antimicrobial resistance causes millions of infections and deaths each year, necessitating new approaches to minimise these effects. Here, we present a direct comparison of crystalline, MoS2 nanosheets (MNS) and amorphous MoS2 nanoflowers (MNF) against Escherichia coli MG1655 (Gram-negative bacteria) and Staphylococcus aureus RN4220 (Gram-positive bacteria). The crystalline MNS sheets of different sizes were obtained using exfoliation of bulk MoS2, while the amorphous MNF was formed using hydrothermal synthesis. Using XPS, the MNF was found to contain MoO3 in addition to MoS2, while the MNS ha…

Molecular mechanisms of antibiotic resistance in Gram-positive and Gram-negative bacteria - a narrative review.

Antimicrobial resistance (AMR) represents one of the most pressing global public health challenges of the 21st century, driven by the widespread inappropriate use of antibiotics and the remarkable adaptive capacity of pathogenic bacteria This narrative review provides an integrated comparative analysis of AMR mechanisms across Gram-positive and Gram-negative bacteria, with particular emphasis on clinically significant ESKAPE pathogens. The four principal resistance mechanisms: enzymatic drug inactivation, reduced drug uptake, target site modification, and active efflux, are examined comparativ…

Target affinities of faropenem to and its impact on the morphology of gram-positive and gram-negative bacteria.

Faropenem is a new oral beta-lactam antibiotic unique from carbapenems and other available beta-lactams. Determinants of the in vitro activity of beta-lactam antibiotics include affinity to penicillin-binding proteins (PBPs) and beta-lactamase stability. In this study, the binding affinity of faropenem to various PBPs and its impact on the morphology of Staphylococcus aureus and Escherichia coli were evaluated. In general, faropenem demonstrated high binding affinity to high-molecular-weight PBPs but low affinity to low-molecular-weight PBPs. In S. aureus and Streptococcus pneumoniae, faropene

Green synthesis of capped gold nanoparticles and their effect on Gram-positive and Gram-negative bacteria.

AIM: We report synthesis of capped gold nanoparticles (C-AuNPs) of ≈20-30 nm by reducing HAuCl4 with flower and leaf extracts of Ocimum tenuiflorum, leaves of Azadirachta indica and Mentha spicata and peel of Citrus sinensis plants. METHODS: Atomic force microscopy (AFM) and transmission electron microscopy (TEM) determined their size, shape and topographical structures. The C-AuNPs with UV-Vis spectrophotometer produced a maximum absorption within 530-535 nm wavelengths. Their Fourier transform IR stretching frequencies, from 450 to 4000 cm-1, have inferred HAuCl4 reduction to Au. RESULTS:

Thrombin-Derived Peptides Potentiate the Activity of Gram-Positive-Specific Antibiotics against Gram-Negative Bacteria.

The continued rise of antibiotic resistance threatens to undermine the utility of the world's current antibiotic arsenal. This problem is particularly troubling when it comes to Gram-negative pathogens for which there are inherently fewer antibiotics available. To address this challenge, recent attention has been focused on finding compounds capable of disrupting the Gram-negative outer membrane as a means of potentiating otherwise Gram-positive-specific antibiotics. In this regard, agents capable of binding to the lipopolysaccharide (LPS) present in the Gram-negative outer membrane are of par

Diagnosis, management, and outcomes of brain abscess due to gram-negative versus gram-positive bacteria.

OBJECTIVES: Differences in management and outcomes of brain abscesses due to gram-positive (GPB) versus gram-negative bacteria (GNB) are not well defined. METHODS: A retrospective review of adult patients with brain abscesses due to monomicrobial infection from 2009 through 2020 was performed. RESULTS: A total 177 patients had a monomicrobial brain abscess; 143 (80.8%) caused by GPB and 34 (19.2%) by GNB. Patients with GNB had more history of head/neck surgery than those with GPB (58.8% vs 36.4%; P = 0.02). Pathogens in the GNB group included Pseudomonas aeruginosa (29.4%), Klebsiella spp (2…

[Importance and prevalence of multidrug-resistant gram-negative bacteria in Germany].

Infections with multidrug-resistant gram-negative bacterial species are a great concern in clinics in Germany. By limiting therapeutic options dramatically, these bacteria pose a significant threat to patient health and cause extensive pressure on hygiene systems and patient management. In Germany, the recommendations on how to deal with these bacteria are called MRGN classification, using the terms 3MRGN and 4MRGN for bacteria resistant to three or four major classes of antibiotics. To be resistant to this large number of antibiotics and become classified as 3MRGN or 4MRGN, bacterial strains …
