Innate Immune Recognition of Helminths: TLRs and Beyond.

Helminth parasites are complex metazoans belonging to diverse taxonomic families, each exhibiting distinct host tissue tropisms as their larval stages migrate through various tissues to complete their life cycles. Migrating larvae through the host trigger tissue disruption/damage, leading to the release of damage-associated molecular patterns (DAMPs). In addition, helminth larvae and adults release excretory/secretory factors, including proteins, lipids, nucleic acids or carry bacterial symbionts, that act as pathogen-associated molecular patterns (PAMPs), which similar to DAMPs, are sensed by

Helminths in alternative therapeutics of inflammatory bowel disease.

Inflammatory bowel disease (IBD), which includes Crohn's disease and ulcerative colitis, is a nonspecific chronic inflammation of the gastrointestinal tract. Despite recent advances in therapeutics and newer management strategies, IBD largely remains untreatable. Helminth therapy is a promising alternative therapeutic for IBD that has gained some attention in the last two decades. Helminths have immunomodulatory effects and can alter the gut microbiota. The immunomodulatory effects include a strong Th2 immune response, T-regulatory cell response, and the production of regulatory cytokines. Alt…

Helminths as an alternative therapy for intestinal diseases.

Animal models and clinical studies have shown that helminth infections exert immunomodulatory activity, altering intestinal permeability and … e for a significant rise in expenses. Some questions remain to be investigated regarding the use of helminths in intestinal disease, such as the importance of the specific species of helminths used, appropriate dosing regimens, optimal timing of treatment, the role of host genetics, diet, environment, and the elucidation of the exact mechanisms of action. Why does infection with some helminths cause cancer?

Infections with Opisthorchis viverrini, Clonorchis sinensis and Schistosoma haematobium are classified as Group 1 biological carcinogens: definitive causes of cancer. These worms are metazoan eukaryotes, unlike the other Group 1 carcinogens including human papilloma virus, hepatitis C virus, and Helicobacter pylori. By contrast, infections with phylogenetic relatives of these helminths, also trematodes of the phylum Platyhelminthes and major human pathogens, are not carcinogenic. These inconsistencies prompt several questions, including how might these infections cause cancer? And why is infec…

[Helminths of domestic pork in Ituri, Upper Zaire].

In Ituri (Upper-Zaïre) pig farming is extensively and traditionally managed, animals wandering about in the villages and neighbouring fields. Helminths of the gastrointestinal tract most commonly encountered are Oesophagostomum sp. Hyostrongylus rubidus, Globocephalus urosubulatus and Ascarops strongylina. Parasitic burdens are low and gross lesions not frequently seen except nodules of oesophagostomiasis. Infections with Metastrongylus salmi and Metastrongylus pudendoctus have a very high prevalence (between 64 and 86%) causing gross lesions of pneumonia on the diaphragmatic lobes. The presen…

The clinical significance of gastrointestinal helminths--a review.

Gastrointestinal helminths (nematodes, trematodes and cestodes) constitute some of the most common and important infective agents of mankind and are responsible for much morbidity and some mortality. Whereas many symptoms and signs are confined to the intestine and less often the associated digestive organs, systemic manifestations are also numerous; this applies especially to indigenous populations of developing 'Third World' countries. Using a clinical classification these organisms can be broadly separated into those involving the small-intestine and those which have a colo-rectal distribut…

Bacterial communities in the gastrointestinal tract segments of helminth-resistant and helminth-susceptible sheep.

BACKGROUND: Helminth parasitism is a world-wide problem in livestock industries, with major impacts on health, welfare and productivity. The role of the gut microbiota in host-helminth interactions in ruminants has been extensively examined and the present study added to this body of knowledge by assessing the effects of resistance and susceptibility to helminth infection in the gastro-intestinal tract (GIT). Australian Sheep Breeding Values (ASBVs) for faecal egg count (FEC) were used to select the 10 highly helminth-susceptible (High-FEC) and 10 highly helminth-resistant (Low-FEC) sheep. FEC

Helminth Mediated Attenuation of Systemic Inflammation and Microbial Translocation in Helminth-Diabetes Comorbidity.

Type 2 diabetes mellitus (T2DM) is characterized by heightened systemic inflammation and microbial translocation. Whether concomitant helminth infections can modulate this systemic response is unclear. We examined the presence of markers of systemic inflammation (levels of acute phase proteins) and of microbial translocation [levels of lipopolysaccharide (LPS) and its associated products] in T2DM individuals with (Ss+) or without (Ss-) Strongyloides stercoralis (Ss) infection. We also analyzed these parameters at 6 months following anthelmintic treatment in Ss+ individuals. Ss+ individuals exh

Intestinal helminth infections among inmates in Bedele prison with emphasis on soil-transmitted helminths.

BACKGROUND: Intestinal helminths infect more than two billion people worldwide. They are common in developing countries where sanitary facilities are inadequate. There is scarcity of documented data on the magnitude of intestinal helminths among inmates in Ethiopia. The aim of this study was to determine prevalence of intestinal helminth infections among inmates in Bedele prison, south-western Ethiopia. METHODS: A cross-sectional study involving 234 inmates in Bedele prison was conducted in April 2012. Socio-demographic data was collected from each study participant using semi-structured ques…

Recovery of helminths postmortem from equines. II. Helminths and larvae of gasterophilus in the gastro-intestinal tract and oestrids from the sinuses.

The tongue, pharynx, oesophagus and gums are examined for larvae of gasterophilus spp., and the nose and sinuses for oestrid larvae. The gastro-intestinal tract is divided into separate specimens-stomach, small intestine, caecum, ventral colon, dorsal colon, descending colon and rectum- and each is examined separately. Aliquots of ingesta of 1/4 by mass of the stomach and 1/10 by mass of the small intestine, caecum, ventral colon and dorsal colon are collected for microscopic examination. Each part of the wall of the caecum, ventral and dorsal colon is washed and specimens are collected for su

Intestinal helminths skew dendritic cell development to counter the anti-helminth immune response.

Intestinal dendritic cells (DC) orchestrate the adaptive immune response that balances protection against pathogens and tolerance towards harmless antigens. Yet, it remains largely unknown what molecular mechanisms underly this functional duality. Heligmosomoides polygyrus bakeri (Hpb) is a naturally occurring parasite in mice characterised by unique immune-evasion strategies that counter host protective immunity. Here, we utilised this unique infection environment to study mechanisms by which DC maintain the immune balance. In-depth characterisation of intestinal DC during Hpb infection revea

Hunting for Helminths: short- and long-read shotgun metagenomics for helminth detection in faecal samples.

Soil-transmitted helminths (STHs) pose significant challenges to public health in endemic areas, necessitating reliable methods for their detection. Shotgun metagenomics enables simultaneous detection of STHs and microbes in a sample without prior knowledge of what is present. However, validation of shotgun metagenomics with known infection intensity or across different sequencing platforms has not been carried out for eukaryote parasites including STHs, and false positives remain a pervasive issue. We validated shotgun metagenomics as a method of STH detection in faecal samples. Using the Str…

Molecular screening of mosquitoes for filarioid helminths in Slovenia.

BACKGROUND: Dirofilaria repens infections have already been reported in both dogs and humans in Slovenia. Xenomonitoring of mosquitoes for filarioid helminths has been used in many studies across Europe to analyze the autochthonous occurrence of filarioids. The aim of this study was to identify potential mosquito vectors of D. repens and other filarioid helminths in Slovenia, to shed light on the significance and extent of possible autochthonous transmission of filarioids in the country. METHODS: This study was a part of the Slovenian nationwide screening program of mosquitoes for filarioid h…

Anthelmintic resistance in soil-transmitted helminths: One-Health considerations.

The One-Health approach recognizes the intricate connection between human, animal, and environmental health, and that cooperative effort from various professionals provides comprehensive awareness and potential solutions for issues relating to the health of people, animals, and the environment. This approach has increasingly gained appeal as the standard strategy for tackling emerging infectious diseases, most of which are zoonoses. Treatment with anthelmintics (AHs) without a doubt minimizes the severe consequences of soil-transmitted helminths (STHs); however, evidence of anthelmintic resist…

Mites and helminths infestation in Lebanese broiler farming systems.

This study aims at the identification of endo- and ectoparasites in broiler farms while assessing their prevalence in semi-open and closed rearing systems in Lebanon. Seventeen semi-open and six closed broiler farms were investigated in various Lebanese provinces between March and September 2020 and 2021. Intestinal and composite litter samples were collected and observed for morphological identification of prevalent parasites. Results notably revealed the prevalence of mites in all of the surveyed farms. Moreover, mixed mite-helminth infestation was recorded in 100% of the semi-open farms and
