Chlamydial infection induces pathobiotype-specific protein tyrosine phosphorylation in epithelial cells.

Members of the genus Chlamydia are strict obligate intracellular pathogens that exhibit marked differences in host range and tissue tropism despite sharing a remarkable level of genomic synteny. These pathobiotype differences among chlamydiae are also mirrored in their early interactions with cultured mammalian host cells. Chlamydial attachment and entry is known to trigger protein tyrosine phosphorylation. In this study, we examined the kinetics and pattern of protein tyrosine phosphorylation induced by infection with a comprehensive collection of chlamydial strains exhibiting diversity in ho

Altering the tropism of lentiviral vectors through pseudotyping.

The host range of retroviral vectors including lentiviral vectors can be expanded or altered by a process known as pseudotyping. Pseudotyped lentiviral vectors consist of vector particles bearing glycoproteins (GPs) derived from other enveloped viruses. Such particles possess the tropism of the virus from which the GP was derived. For example, to exploit the natural neural tropism of rabies virus, vectors designed to target the central nervous system have been pseudotyped using rabies virus-derived GPs. Among the first and still most widely used GPs for pseudotyping lentiviral vectors is the v

Comparative Infection Pathways of Listeria monocytogenes in Humans Versus Ruminants.

Severe infection caused by L. monocytogenes in humans is primarily linked to the pathogen's ability to cross the intestinal barrier and disseminate within the host. Once internalized, L. monocytogenes exhibits intracellular motility that enables colonization of secondary target organs, including the brain and placenta. This review aims to critically compare the infection pathways of L. monocytogenes in humans and ruminant hosts, highlighting the molecular mechanisms that enable bacterial survival, organ tropism, and adaptation to host-specific stressors. By focusing on these comparative pathwa

Differences in Tissue and Species Tropism of Reptarenavirus Species Studied by Vesicular Stomatitis Virus Pseudotypes.

Reptarenaviruses cause Boid Inclusion Body Disease (BIBD), and co-infections by several reptarenaviruses are common in affected snakes. Reptarenaviruses have only been found in captive snakes, and their reservoir hosts remain unknown. In affected animals, reptarenaviruses appear to replicate in most cell types, but their complete host range, as well as tissue and cell tropism are unknown. As with other enveloped viruses, the glycoproteins (GPs) present on the virion's surface mediate reptarenavirus cell entry, and therefore, the GPs play a critical role in the virus cell and tissue tropism. The evolution of in vitro culture systems for the study of hepatitis E virus infection.

Hepatitis E virus (HEV), a major cause of acute viral hepatitis worldwide, is increasingly recognized as an important zoonotic pathogen capable of causing chronic infection and diverse extrahepatic manifestations. However, progress in understanding the biology and pathogenesis of HEV has historically been constrained by the limited availability of robust and reproducible in vitro culture systems. Early work relied heavily on hepatoma-derived cell lines, which, while experimentally tractable, often support inefficient viral replication and incompletely reflect hepatic physiology. Recently, subs

The contribution of viral toxins to infection and pathogenesis.

The process by which viruses cause disease, viral pathogenesis, is the result of both infection of cells and the host immune response. A less studied but equally important contributor to viral pathogenesis is viral dissemination, the capacity of a virus to move from the primary site of infection, traverse physiological barriers, and gain access to secondary sites of infection. This dictates viral tropism and pathogenesis, but the mechanisms governing barrier crossing are incompletely understood. While the presence of viral receptors on cells is a major determinant of viral tropism and a prereq

Erythroid cells and malaria parasites: it's a match!

PURPOSE OF REVIEW: The current review outlines recent discoveries on the infection of erythroid cells by Plasmodium parasites, focusing on the molecular interactions governing the tropism of parasites for their host cell and the implications of this tropism for parasite biology and erythroid cell maturation. RECENT FINDINGS: Although most studies about the interactions of Plasmodium parasites and their host cell focused on the deadliest human malaria parasite, Plasmodium falciparum, and the erythrocyte, there is increasing evidence that several Plasmodium species, including P. falciparum, als

Organs infected with Trypanosoma cruzi and DTU identification in the naturally infected rodent Octodon degus.

Chagas disease is a public health problem in America. Its parasite, Trypanosoma cruzi, presents different discrete typing units (DTUs), colonizes organs of mammalian hosts in chronic infections, and presents tropism for particular organs in experimental infections. We evaluated T. cruzi tropism towards organs on the naturally infected rodent Octodon degus, identifying the parasites' DTUs, by means of conventional PCR and hybridization. Almost all the analyzed organs presented T. cruzi. More than 42% of the tested oesophagus, skin, skeletal muscle, brain and intestine showed T. cruzi DNA. Other

Beyond hepatitis D virus: the discovery of animal deltaviruses, their diversity, helpers and their position in an expanding world of circular RNA replicons.

Hepatitis D virus (HDV) was long considered a singular human satellite virus that depends on hepatitis B virus (HBV) for envelopment and transmission. Recent metatranscriptomic mining has uncovered a growing diversity of HDV-like viruses, we term deltaviruses, across vertebrates, invertebrates, and environmental samples, overturning assumptions about strict hepatic tropism and obligatory hepadnavirus helpers. Here, we synthesize shared structural and replication principles of deltaviruses: rolling-circle amplification by host polymerases and ribozyme-driven RNA processing. We then discuss emer

Update on enterovirus 71 infection.

Human enterovirus type 71 (EV71) has emerged as a major cause of viral encephalitis in children worldwide. The identified EV71 receptors provide useful information for understanding EV71replication and tissue tropism. Host factors interact with the internal ribosome entry site (IRES) of EV71 to regulate viral translation. However, the specific molecular features of the EV71 genome that determine virulence remain unclear. The EV71 capsid protein VP1 region might contribute to virulence and neurotropism. Transgenic mice expressing the EV71 receptor that were infected with the virus exhibited a d

Infectious bronchitis-virus-like QX strain transmission, pathogenesis, replication, and host miRNA biogenesis pathway hijacking mechanism.

The infectious bronchitis virus (IBV) is an acute, highly contagious, single-stranded RNA (ssRNA) gammacoronavirus mainly transmitted to chickens through the intranasal route. Positive-sense ssRNA viruses primarily act on mRNA and enhance the replication of viral copies. We identified the nasal entry site of the IBV-QX strain and provided insights into the minimal viral replication in systemic organs. Here is an overview of its entry mechanisms and tropism in systemic organ tissues. It enters the host cells via spike proteins, which bind to highly expressed receptors in respiratory, renal, and

Pathogenicity and virulence of Candida auris.

Candida auris (Candidozyma auris) has emerged as a multidrug‑resistant human fungal pathogen that causes infections of high morbidity and mortality. Notably, it exhibits a unique ability to grow and persist on human skin, thus leading to efficient transmission through skin-to-skin contact. As a result, C. auris poses a significant risk of outbreaks in healthcare settings, especially in nursing homes that care for elderly patients. Most concerning, C. auris clinical isolates demonstrate widespread and, in some cases, untreatable resistance to all antifungal drug classes, including azoles, polye

Differential Interactions of Tissue-Restricted Host Proteins SPLUNC1 and VAMP8 with VP3 of Human Bocaviruses 1 and 2.

Background: Four genotypes of human bocaviruses (HBoVs) have been identified, with only HBoV1 being detected in respiratory specimens, and with HBoV2 being the predominant human bocavirus in fecal specimens, which implies different tissue tropisms for HBoV1 and HBoV2. It is vital to determine the factors that influence the tissue tropisms. Methods: The major capsid proteins VP3 of HBoV1 and HBoV2 were expressed in eukaryotic cells. Then co-immunoprecipitation (Co-IP) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) (IP-MS) was employed, along with Gene Ontology (GO) and Kyoto Ency

A permanent host shift of rabies virus from Chiroptera to Carnivora associated with recombination.

Bat virus host shifts can result in the spread of diseases with significant effects. The rabies virus (RABV) is able to infect almost all mammals and is therefore a useful model for the study of host shift mechanisms. Carnivore RABVs originated from two historical host shifts from bat viruses. To reveal the genetic pathways by which bat RABVs changed their host tropism from bats to carnivores, we investigated the second permanent bat-to-carnivore shift resulting in two carnivore variants, known as raccoon RABV (RRV) and south-central skunk RABV (SCSKV). We found that their glycoprotein (G) gen

A Cas9 Ribonucleoprotein Platform for Functional Genetic Studies of HIV-Host Interactions in Primary Human T Cells.

New genetic tools are needed to understand the functional interactions between HIV and human host factors in primary cells. We recently developed a method to edit the genome of primary CD4+ T cells by electroporation of CRISPR/Cas9 ribonucleoproteins (RNPs). Here, we adapted this methodology to a high-throughput platform for the efficient, arrayed editing of candidate host factors. CXCR4 or CCR5 knockout cells generated with this method are resistant to HIV infection in a tropism-dependent manner, whereas knockout of LEDGF or TNPO3 results in a tropism-independent reduction in infection. [Molecular mechanism of tissue-specific infection of poliovirus].

Poliovirus is the causative agent of an acute disease of the central nervous system, poliomyelitis. Poliovirus will be eradicated in the near future by a world-wide vaccination program. Poliovirus is a neurotropic virus that produces severe lesions selectively in the CNS. However, a basic question why poliovirus exhibits neurotropic property has not been elucidated. Poliovirus receptor and host factors involved in the translation initiation of viral protein, which are required for virus replication, play important roles in determining tissue tropism. We found that type I interferon response is

Influenza B Virus Receptor Specificity: Closing the Gap between Binding and Tropism.

Influenza A and influenza B viruses (FLUAV and FLUBV, respectively) cause significant respiratory disease, hospitalization, and mortality each year. Despite causing at least 25% of the annual disease burden, FLUBV is historically understudied. Unlike FLUAVs, which possess pandemic potential due to their many subtypes and broad host range, FLUBVs are thought to be restricted to only humans and are limited to two lineages. The hemagglutinins (HA) of both influenza types bind glycans terminating in α2,6- or α2,3-sialic acids. For FLUAV, the tropism of human- and avian-origin viruses is well-defin

Insight into the Life Cycle of Enterovirus-A71.

Human enterovirus 71 (EV-A71), a member of the Picornaviridae family, is predominantly associated with hand, foot, and mouth disease in infants and young children. Additionally, EV-A71 can cause severe neurological complications, including aseptic meningitis, brainstem encephalitis, and fatalities. The molecular mechanisms underlying these symptoms are complex and involve the viral tissue tropism, evasion from the host immune responses, induction of the programmed cell death, and cytokine storms. This review article delves into the EV-A71 life cycle, with a particular emphasis on recent advanc

A Masquerading Case of Cryptococcus neoformans.

Cryptococcus is a common infection in immunocompromised hosts. Ordinarily, Cryptococcus tends to have a strong tropism for the central nervous system (CNS). Other organs or systems of the body are less commonly involved. Here we describe a patient who had cutaneous tropism. The predominant presentation of this patient was painful thigh lesions. This case highlights several practical difficulties in identifying such infections, and also demonstrates the clinical utility of some of the commonly followed practices in infectious diseases and medicine.

Airway organoids reveal patterns of influenza A tropism and adaptation in wildlife species.

Identifying animal species that are susceptible to the plethora of existing and emerging viruses is critical for predicting and containing disease outbreaks. Current efforts to assess viral tropism largely rely on experimental infection models, but such experiments are logistically and ethically infeasible for many wildlife species. To tackle this challenge, we developed a panel of airway organoids from ten taxonomically diverse wildlife and livestock species and evaluated their susceptibility to influenza viruses of mammalian (pH1N1) and avian (H5N1) origin. Our analyses revealed large host-s
