[Immunochemical properties of the extracellular hydrolases (protease and alkaline phosphatase) of Coccidioides immitis].

Extracellular hydrolases (protease and alkaline phosphatase) of the coccidioidal fungus possessed antigenic properties and caused production of the corresponding antibodies. Phosphatase-antiphosphatase-substrate system apparently has future prospects for the elaboration of immunobiochemical methods for the diagnosis of coccidioidomycosis.

Secretory bile salt hydrolases regulate niche displacement and gut colonization of vancomycin-resistant Enterococcus.

Vancomycin-resistant enterococci (VRE) residing in the gut are major causes of hospital-acquired infections worldwide. Bacteria belonging to the Bacteroidales order have been linked to protection against VRE, but the mechanism remains unclear. Here, we identify an important role for Bacteroidales species in mediating resistance to VRE through a distinct subtype of bile salt hydrolases (BSHs) that retain an uncleaved N-terminal signal peptide. This subclass of BSHs harbored by Bacteroides species promotes VRE inhibition through deoxycholic acid (DCA) production in the presence of Lachnospiracea

Similar but Distinct-Biochemical Characterization of the  Staphylococcus aureus Serine Hydrolases FphH and FphI.

Staphylococcus aureus is a major cause of infections like bacteremia, pneumonia, and endocarditis. These infections are often linked to the ability of S. aureus to form biofilms. Several S. aureus serine hydrolases have previously been identified to be active during biofilm-forming conditions. Here, we present the biochemical characterization of two of these enzymes-fluorophosphonate binding hydrolase H and I (FphH, FphI). Cryogenic and room-temperature X-ray crystallography, enzymatic substrate profiling, small-angle X-ray scattering analysis, and molecular dynamics simulations provide new in

Cloning and Characterization of Drosophila melanogaster Juvenile Hormone Epoxide Hydrolases (JHEH) and Their Promoters.

Juvenile hormone epoxide hydrolase (JHEH) plays an important role in the metabolism of JH III in insects. To study the control of JHEH in female Drosophila melanogaster, JHEH 1, 2 and 3 cDNAs were cloned and sequenced. Northern blot analyses showed that the three transcripts are expressed in the head thorax, the gut, the ovaries and the fat body of females. Molecular modeling shows that the enzyme is a homodimer that binds juvenile hormone III acid (JH IIIA) at the catalytic groove better than JH III. Analyses of the three JHEH promoters and expressing short promoter sequences behind a reporte

Preclinical Evaluation of Recombinant Microbial Glycoside Hydrolases in the Prevention of Experimental Invasive Aspergillosis.

Aspergillus fumigatus is a ubiquitous mold that can cause invasive pulmonary infections in immunocompromised patients. Within the lung, A. fumigatus forms biofilms that can enhance resistance to antifungals and immune defenses. Aspergillus biofilm formation requires the production of a cationic matrix exopolysaccharide, galactosaminogalactan (GAG). In this study, recombinant glycoside hydrolases (GH)s that degrade GAG were evaluated as antifungal agents in a mouse model of invasive aspergillosis. Intratracheal GH administration was well tolerated by mice. Pharmacokinetic analysis revealed that

Role of epoxy-fatty acids and epoxide hydrolases in the pathology of neuro-inflammation.

Neuroinflammation is a physiologic response aimed at protecting the central nervous system during injury. However, unresolved and chronic neuroinflammation can lead to long term damage and eventually neurologic disease including Parkinson's disease, Alzheimer's disease and dementia. Recently, enhancing the concentration of epoxyeicosatrienoic acids (EETs) through blocking their hydrolytic degradation by soluble epoxide hydrolase (sEH) has been applied towards reducing the long-term damage associated with central neurologic insults. Evidence suggests this protective effect is mediated, at least

A machine learning approach to identify active polysorbate 20 degrading hydrolases in biopharmaceutical formulations.

Polysorbate degradation by host cell-derived hydrolases presents a critical challenge in biopharmaceutical formulations. It can lead to fatty acid release, particle formation and reduced product stability. Mass spectrometry-based host cell protein (HCP) analysis is widely used for HCP identification, but detection becomes challenging in formulations where monoclonal antibodies are present in large excess. In such cases, hydrolases can remain undetected, despite being enzymatically active at trace levels. In this study, we demonstrate that individual CHO-derived hydrolases generate distinct pol

Unlocking the molecular engineering of Geobacillus glycoside hydrolases as a source of industrial biocatalysts.

This review examines Geobacillus sensu stricto as a source of thermostable glycoside hydrolases (GH) for biomass conversion, food processing, and enzyme engineering. Recent peer-reviewed literature was assessed with emphasis on taxonomy, genome-based Carbohydrate-Active Enzymes (CAZyme) prediction, biochemical validation, structural data, and engineering case studies. Taxonomic boundaries were interpreted using current Anoxybacillaceae frameworks, with Parageobacillus treated as a related comparator rather than as Geobacillus. The strongest evidence supports GH13 alpha-amylases, xylan-active s

Activity-based protein profiling of serine hydrolases and penicillin-binding proteins in Enterococcus faecium.

Enterococcus faecium is a gut commensal bacterium which is gaining increasing relevance as an opportunistic, nosocomial pathogen. Its high level of intrinsic and acquired antimicrobial resistance is causing a lack of treatment options, particularly for infections with vancomycin-resistant strains, and prioritizes the identification and functional validation of novel druggable targets. Here, we use activity-based protein profiling (ABPP), a chemoproteomics approach using functionalized covalent inhibitors, to detect active serine hydrolases across 11 E. faecium and Enterococcus lactis strains.

Activity-Based Proteome Profiling of Serum Serine Hydrolases: Application in Pediatric Abusive Head Trauma.

PURPOSE: Traumatic brain injury (TBI), including pediatric abusive head trauma (AHT), is the leading cause of death and disability in children and young adults worldwide. The current understanding of trauma-induced molecular changes in the brain of human subjects with intracranial hemorrhage (ICH) remains inadequate and requires further investigation to improve the outcome and management of TBI in the clinic. Calcium-mediated damage at the site of brain injury has been shown to activate several catalytic enzymes. EXPERIMENTAL DESIGN: Serine hydrolases (SHs) are major catalytic enzymes involve

Engineering of Long-Circulating Peptidoglycan Hydrolases Enables Efficient Treatment of Systemic Staphylococcus aureus Infection.

Staphylococcus aureus is a human pathogen causing life-threatening diseases. The increasing prevalence of multidrug-resistant S. aureus infections is a global health concern, requiring development of novel therapeutic options. Peptidoglycan-degrading enzymes (peptidoglycan hydrolases, PGHs) have emerged as a highly effective class of antimicrobial proteins against S. aureus and other pathogens. When applied to Gram-positive bacteria, PGHs hydrolyze bonds within the peptidoglycan layer, leading to rapid bacterial death by lysis. This activity is highly specific and independent of the metabolic

Replication of Coxiella burnetii in a Lysosome-Like Vacuole Does Not Require Lysosomal Hydrolases.

Coxiella burnetii is an intracellular bacterium that causes query, or Q fever, a disease that typically manifests as a severe flu-like illness. The initial target of C. burnetii is the alveolar macrophage. Here, it regulates vesicle trafficking pathways and fusion events to establish a large replication vacuole called the Coxiella-containing vacuole (CCV). Similar to a phagolysosome, the CCV has an acidic pH and contains lysosomal hydrolases obtained via fusion with late endocytic vesicles. Lysosomal hydrolases break down various lipids, carbohydrates, and proteins; thus, it is assumed C. burn

Activity of selected hydrolases in excretion-secretion products and extracts of adult Contracaecum rudolphii.

BACKGROUND: Enzymes contained in excretion-secretion (ES) products of parasites released to the environment play multiple roles: they facilitate hatching and moulting of larvae, enable a parasite to migrate within tissues, inhibit blood coagulation, defend the parasite from host's immunological response, and enhance feeding and nutrition. The aim of the study was to determine hydrolase activity in ES products and extracts from adult Contracaecum rudolphii. MATERIALS AND METHODS: Adult nematodes were isolated from intestines of black cormorants nesting at Katy Rybackie (the Vistula Lagoon). Ne

[The enzyme-antienzyme interactions of a number of Burkholderia pseudomallei hydrolases with monoclonal immunoglobulins].

Phosphatase, phospholipase C and a proteolytic complex, including casein- and hemoglobin-hydrolases, have been isolated from cell-free extracts of B. pseudomallei cultivation medium. A set of monoclonal antibodies (McAb) to the antigenic complexes of this infective agent has been obtained. Conditions for the study of the interaction of the enzymes and McAb have been worked out, and most of the 14 immunoglobulins under study have been shown to neutralize 2-3 hydrolases each. Three types of McAb, selectively inhibiting only the proteolytic complex, only casein-hydrolase and only phospholipase C,
