One of the primary approaches in experimental brain research is to investigate the effects of specific destruction of its parts. Here, several neurotoxins are available which can be used to eliminate neurons of a certain neurochemical type or family. With respect to the study of dopamine neurons in the brain, especially within the basal ganglia, the neurotoxin 6-hydroxydopamine (6-OHDA) provides an important tool. The most common version of lesion induced with this toxin is the unilateral lesion placed in the area of mesencephalic dopamine somata or their ascending fibers, which leads to a lat

To clarify the behavioral characteristics of rats neonatally treated with 6-hydroxydopamine (6-OHDA), their performance on 4 aversive learning tasks, 3 active (shuttle, one-way, and rearing) avoidance tasks and one passive (step-through) avoidance task, was examined. On days 2 and 4 after birth, each rat of F344/Du strain received bilateral intraventricular injections of 6-OHDA (35 micrograms x 2) or vehicle solution following desmethylimipramine (20 mg/kg, s.c.) pretreatment. From day 90, each rat was trained in one of the 4 avoidance tasks. 6-OHDA-treated rats showed significantly less avoid

The effect of substance P and morphine on the 6-hydroxydopamine (6-OHDA) induced alteration of the postnatal development of central noradrenaline (NA) neurons in the rat has been investigated using neurochemical techniques. Neonatal administration of 6-OHDA systemically leads to permanent NA denervations of distant NA projections, while the projections close to the NA cell bodies are increased, leading to NA hyperinnervation. Intracisternal injection of substance P was found to counteract both the NA denervation and hyperinnervation induced by 6-OHDA. The effect of substance P disclosed a clea

Parkinson's disease (PD) is a neurodegenerative disorder caused by oxidative stress-dependent loss of dopaminergic neurons in the substantia nigra and elevated microglial inflammatory responses. Recent studies show that cell loss also occurs in the hypothalamus in PD. However, effective treatments for the disorder are lacking. Thioredoxin is the major protein disulfide reductase in vivo. We previously synthesized an albumin-thioredoxin fusion protein (Alb-Trx), which has a longer plasma half-life than thioredoxin, and reported its effectiveness in the treatment of respiratory and renal disease

Parkinson's disease (PD) is a chronic neurodegenerative disease characterized by a significant loss of dopaminergic neurons within the substantia nigra pars compacta (SNpc) and a subsequent loss of dopamine (DA) within the striatum. Despite advances in the development of pharmacological therapies that are effective at alleviating the symptoms of PD, the search for therapeutic treatments that halt or slow the underlying nigral degeneration remains a particular challenge. Activin A, a member of the transforming growth factor β superfamily, has been shown to play a role in the neuroprotection of

Gait disturbances similar to those of human Parkinson's disease (PD) can be observed in animals after administration of neurotoxin 6-hydroxydopamine (6-OHDA) to induce unilateral nigrostriatal dopamine depletion. However, the relationship between gait disturbances and dopamine depletion following 6-OHDA infusion has not been determined. The present study investigated the longitudinal changes of spatiotemporal gait patterns using a walkway system to acquire footprints and lateral limb images over a 6-week period following unilateral 6-OHDA injection into the medial forebrain bundle of rats. Our

Parkinson's disease (PD) is a neurodegenerative disorder characterized by progressive loss of dopaminergic (DA) neurons of the substantia nigra pars compacta. 6-Hydroxydopamine (6-OHDA) is specific to dopaminergic neurons in intrastriatal rodent models. It induces neuronal death either via uncoupling mitochondrial oxidative phosphorylation resulting in energy deprivation or alternatively, is associated with its ability to produce hydrogen peroxide, hydroxyl and superoxide radicals. Caffeic acid phenethyl ester (CAPE), an antioxidant flavanoid, has antiviral, anti-inflammatory, antioxidant, and

We investigated the effects of dopamine depletion on acoustically guided behavior of anurans by conducting phonotaxis experiments with female gray treefrogs (Hyla versicolor) before and 90 min after bilateral injections of 3, 6, or 12 microg 6-hydroxydopamine (6-OHDA) into the telencephalic ventricles. In experiments with one loudspeaker playing back a standard artificial mating call, we analyzed the effects of 6-OHDA on phonotactic response time. In choice tests we measured the degree of distraction from the standard call (20 pulses/s) by three different variants with altered pulse-rate (30/s

Apomorphine is a potent radical scavenger and iron chelator. In vitro apomorphine acts as a potent iron chelator and radical scavenger with IC50 of 0.3 microM for iron (2.5 microM) induced lipid peroxidation in rat brain mitochondrial preparation, and it inhibits mice striatal MAO-A and MAO-B activities with IC50 values of 93 microM and 241 microM. Apomorphine (1-10 microM) protects rat pheochromocytoma (PC12) cells from 6-hydroxydopamine (150 microM) and H2O2 (0.6 mM) induced cytotoxicity and cell death. The neuroprotective property of (R)-apomorphine, a dopamine D1-D2 receptor agonist, has b

The response to administration of the specific monoamine oxidase A (MAO-A) blocker clorgyline was investigated in adult male Sprague-Dawley rats which were adrenalectomized four days prior to treatment or were additionally sympathectomized as newborns by injection of 6-hydroxydopamine. In both groups, the contents of pineal indoles melatonin and N-acetylserotonin were augmented, and the contents of 5-hydroxyindoleacetic acid and 5-hydroxyindoletryptophol decreased 90 min following clorgyline injections when compared to rats receiving saline. The observed responses were less pronounced in rats

This investigation revealed that the contents of A II and NE in plasma, heart and aorta and E content in plasma were significantly increased in SHR at the age of 12th week. At the same time the systolic blood pressure of SHR was much higher than that of the age-matched WKY. The intracerebroventricular administration of 6-OHDA in SHR at the age of 8th week not only prevented the development of hypertension, but also reduced the contents of NE and E in the brain regions, heart, aorta and plasma. Simultaneously A II content in heart, aorta and plasma was decreased. These results suggested that: 1

Amine accumulation is observed in the lateral hypothalamus (LH) after nigrostriatal neurons degenerate. It has been proposed that this accumulation is a source of amines which are released into the hypothalamus thereby affecting the function of adjacent aminergic receptors. To approximate this condition of continuous exposure of LH receptors to endogenous amines, dopamine (DA) was injected into the LH of rats once daily for 5 consecutive days. A control group received 4 daily injections of tartaric acid vehicle and then DA on day 5. Rats pretreated with DA showed severe impairment of open fiel

Rats in which central noradrenergic terminals have been damaged by injecting 6-hydroxydopamine (6-OHDA) into the cerebral ventricular system have been used to study the possible role of noradrenaline (NA) in the effect of trauma on thermoregulation. In non-injured rats the 6-OHDA treatment did not affect the threshold environmental temperature (Ta) for the onset of shivering or the slope of the regression lines relating intensity of shivering to Ta. It raised the core temperature (Tc) threshold for the opening of the A-V anastomoses in the tail. Whereas in normal rats ischaemic limb injury dep

The effects of the catecholamine neurotoxin 6-hydroxydopamine on phenylethanolamine-N-methyl transferase (PNMT) in rat brain has been investigated by biochemical and immunohistochemical analysis. 6-Hydroxydopamine was administerered either intracisternally to adult rats or systemically to newborn rats, treatments known to affect markedly central noradrenaline neurons. None of these treatments had any significant effect on the PNMT activity in the hypothalamus, pons-medulla and the spinal cord, as measured in vitro using a radiometric assay. The neonatal 6-hydroxydopamine treatment, which produ
