Enzyme immunoassay (EIA) is often the initial test performed for diagnosing coccidioidomycosis by serology. When EIA immunoglobulin G (IgG) optical density (OD) positive results were compared to immunodiffusion complement-fixation (IDCF) titer among 339 unique patient samples, there was only moderate correlation (R2 = 0.5345; P < 0.001). Coccidioides immitis produces two unrelated heat-stable antigens which are diagnostically useful in immunodiffusion tests. One, the tube precipitin antigen, is valuable for specifically detecting antibody and diagnosing early primary cases of coccidioidomycosis. Immunoelectroporesis (IEF) and immunoelectroosmophoresis-immunodiffusion (IEOP-ID) (1) were comparatively used in the diagnosis of 16 patients with mycologically proved paracocidioidomycosis. The value of various serological tests in the diagnosis of paracoccidioidomycosis was studied. Quantitative agar-gel immunodiffusion and indirect immunofluorescent tests were performed, and the results were compared with those of complement fixation and qualitative agar-gel procedures. The quantitative immunodiffusion procedure was found to serve as the simplest and safest quantitative test that could be performed for evaluation purposes, whereas the indirect fluorescent-antibody test gave nonspecific reactions and, as such, proved unsuitable.

The antibodies in sera of patients affected by Chromoblastomycosis are detected using the technique of double immunodiffusion and the mycelial somatic antigens and the culture filtrates antigens of Cladosporium carrionii. From the 13 sera tested 8 have given positive results. The fresh serum from a patient under treatment gives 2 bands, while fresh serum from a non-treated patient gives 3 bands. The titre of antibodies was also determined for the two fresh sera, having found 1/4 for the patient under treatment and 1/32 for the non-treated one.

Five commercial test kits for the serodiagnosis of coccidioidomycosis and histoplasmosis based upon immunodiffusion were evaluated. The correlation of results with the test kits in the Clinical Laboratory varied from 71 to 100% for coccidioidomycosis. Synovial fluids (83.3%) from patients with rheumatoid arthritis gave a positive reaction when examined by immunodiffusion against a Lancefield extract of Group A Type 12 streptococci.
