Ochratoxin A (OTA) is a chemically stable and ubiquitous mycotoxin that poses persistent threats to environmental safety and public health due to its potent nephrotoxicity and carcinogenicity. Given the limitations of conventional physical and chemical remediation, biological detoxification has emerged as a sustainable and high-efficiency alternative. This review systematically evaluates three core biological strategies: biological inhibition via nutrient competition and metabolic interference; biosorption through non-covalent binding by microbial cell wall components; and biodegradation, focu

Cyanides are highly toxic chemicals in several edible plants that threaten food safety and human health. A phenotypically distinct Yarrowia lipolytica strain that efficiently detoxifies multiple cyanogenic glycosides from edible plants was constructed using a family 1 glycosyl-hydrolase (GH1). The strain displayed higher growth rates and metabolic activities when exposed to high concentrations of cyanides than the wild-type. It overexpressed genes that promoted the binding of molecular oxygen to the cytochrome iv complex. The engineered strain repressed fatty acid production to optimize energy

Ammonia is a neurotoxic agent that is primarily generated in the intestine and detoxified in the liver. Toxic increases in systemic ammonia levels predominantly result from an inherited or acquired impairment in hepatic detoxification and lead to potentially life-threatening neuropsychiatric symptoms. Inborn deficiencies in ammonia detoxification mainly affect the urea cycle, an endogenous metabolic removal system in the liver. Hepatic encephalopathy, on the other hand, is a hyperammonemia-related complication secondary to acquired liver function impairment. A range of therapeutic options is a

Individuals who have a diminished capacity to detoxify cyanide to thiocyanate because of either a genetic predisposition (e.g. as occurs in those with Leber's Optic Atrophy) or a diet low in sulfur containing amino acids are predicted to be at increased risk to developing adverse side effects from laetrile, a cyanide containing substance used in cancer treatment.

Hypothalamic pathology, metabolic dysfunction and psychiatric symptoms are part of Huntington disease (HD), which is caused by an expanded CAG repeat in the huntingtin (HTT) gene. Inactivation of mutant HTT selectively in the hypothalamus prevents the development of metabolic dysfunction and depressive-like behavior in the BACHD mouse model. The hypothalamic paraventricular nucleus (PVN) is implicated in metabolic and emotional control, therefore we here tested whether inactivation of mutant HTT in the PVN affects metabolic and psychiatric manifestations of HD in BACHD mice. BACHD mice were cr

The review focuses on genetic variants of human flavin-containing monooxygenase 3 (FMO3) and their impact on enzyme activity, drug metabolism and disease. The majority of FMO-mediated metabolism in adult human liver is catalyzed by FMO3. Some drugs are metabolized in human liver predominantly by FMO3, but most drug substrates of FMO3 are metabolized also by other enzymes, particularly cytochromes P-450, and the FMO3-catalyzed reaction is not the major route of metabolism.Rare variants that severely affect production or activity of FMO3 cause the disorder trimethylaminuria and impair metabolism

It has been understood for nearly a century that most of the cyanide entering mammals by either ingestion or inhalation of sublethal quantities is detoxified by combination with cyanide-reactive sulphur. The detoxifying reaction is primarily enzymic, and the enzymes that catalyse it, as well as those that catalyse reactions which generate the cyanide-reactive sulphane sulphur, have been isolated and studied extensively. However, analytical methods suitable for quantitative characterization of the physiological pool of sulphur available for this detoxification have only recently come under deve

Icotinib (ICT) is an antitumor drug approved by China National Medical Products Administration and is found to be effective against non-small cell lung cancer. The present study aimed at the interaction of ICT with CYP3A. ICT exhibited time-, concentration-, and NADPH-dependent inhibitory effect on recombinant human CYP3A4/5. About 60% of CYP3A activity was suppressed by ICT at 50 μM after 30 minutes. The observed enzyme inhibition could not be recovered by dialysis. Nifedipine protected CYP3A from the inactivation by ICT. The inhibitory effects of ICT on CYP3A were influenced neither by gluta

Effective Brucella inactivation is essential for safe vaccine development, diagnostics, and sample handling, particularly in resource-limited regions without high-level biosafety facilities. This study evaluated heat (80°C/95°C, 10-20 min) and formaldehyde (0.4%/0.6%, 48-72 h) inactivation using the Rev.1 vaccine strain and three Brucella melitensis field isolates from Gansu (GS-XG1, GS-SN2, GS-MQ3). All field isolates were collected from a single geographic region (Gansu, China), which should be considered when interpreting the broader geographic generalizability of the findings. Complete ina

Cardiometabolic disorders encompass a broad range of cardiovascular complications associated with metabolic dysfunction. These conditions have an increasing share in the health burden worldwide due to worsening endemic of hypertension, obesity, and diabetes. Previous studies have identified Tumor Protein p53-inducible Nuclear Protein 2 (Trp53inp2) as a molecular link between hyperglycemia and cardiac hypertrophy. However, its role in cardiac pathology has never been determined in vivo. In this study, we generated a cardiac specific knockout model of Trp53inp2 (Trp53inp2-cKO) and investigated t

Inherited metabolic disorders (IMDs) are a growing group of genetic diseases caused by defects in enzymes that mediate cellular metabolism, often resulting in the accumulation of toxic substrates. The liver is a highly metabolically active organ that hosts several thousands of chemical reactions. As such, it is an organ frequently affected in IMDs. In this article, we review current approaches for liver-directed gene-based therapy aimed at metabolite detoxification in a variety of IMDs. Moreover, we discuss current unresolved challenges in gene-based therapies for IMDs.

OBJECTIVES: To evaluate zolpidem as a mechanism-based inactivator of human CYP3A in vitro, and to assess its metabolic interaction potential with CYP3A drugs (in vitro-in vivo extrapolation; IV-IVE).
METHODS: A co- vs. pre-incubation strategy was used to quantify time-dependent inhibition of human liver microsomal (HLM) and recombinant CYP3A4 (rCYP3A4) by zolpidem. Experiments involving a 10-fold dilution step were employed to determine the kinetic constants of inactivation (K (I) and k (inact)) and to assess the in vitro mechanism-based inactivation (MBI) criteria. Inactivation data were ent

The average worldwide human life expectancy is 70 years, with a significantly higher value in Western societies. Many modern diseases are not associated with premature mortality but with a decreased quality of life in aged patients and an excessive accumulation of various toxic compounds in the human body during life. Today, scientists are especially interested in finding compounds that can help increase a healthy lifespan by detoxifying the body. Phytotherapy with specific approaches is used in alternative medicine to remove toxins from the body. Worldwide, research is conducted to identify m

In this review, novel aspects of the role of esterases, which contribute to the metabolism of 10% of therapeutic drugs, are described. Esterases hydrolyze the compounds that contain ester, amide, and thioester bonds, which cause prodrug activation or detoxification. Among esterases, carboxylesterases are well known to be involved in the hydrolysis of a variety of drugs. Additionally, other esterases have recently received attention for their pharmacological and toxicological roles. Arylacetamide deacetylase (AADAC) is involved in the hydrolysis of flutamide, phenacetin, and rifamycins. AADAC i

Five healthy subjects were exposed to known concentrations of trichloroethylene (T.R.I.) for five hours. The amount retained was calculated. The excretion of the metabolites monochloroacetic acid (M.C.A.), trichloroacetic acid (T.C.A.), and trichloroethanol (T.C.E.) in the urine was measured over the next seven to 14 days. Metabolites excreted represented an average of 73% of the dose of T.R.I. retained (M.C.A. 4%, T.C.A. 19%, T.C.E. 50%). The amount and speed of excretion of metabolites in one experiment was increased by giving glucose and insulin. The diurnal variation in the excretion of T.

Mosquito olfaction regulates host-seeking, ecological adaptation, and vectorial capacity. Beyond the classical receptor-centric paradigm, we argue that xenobiotic-metabolizing enzymes form a dynamic perireceptor layer that actively sculpts odor identity, signal intensity, and duration. This metabolism gain control mechanism may integrate olfactory sensitivity and behavioral plasticity within a unified sensory framework.
