Rhodnius prolixus smells repellents: Behavioural evidence and test of present and potential compounds inducing repellency in Chagas disease vectors.

Insect repellents are known since many decades ago and constitute a major tool for personal protection against the biting of mosquitoes. Despite their wide use, the understanding of why and how repellents repel is relatively recent. In particular, the question about to what extent insects other than mosquitoes are repulsed by repellents remains open. We developed a series of bioassays aimed to test the performance of well established as well as potential repellent molecules on the Chagas disease vector Rhodnius prolixus. Besides testing their ability to prevent biting, we tested the way in whi…

Passive prophylaxis with permethrin-treated tents reduces mosquito bites among North American summer campers.

OBJECTIVE: Over 2 million adolescents participate in summer-camp experiences, placing themselves at risk for mosquito-borne illness. Insect repellent is recommended but often not used. It is unknown whether permethrin treatment of a location, such as a campsite, provides passive prophylaxis reducing mosquito bites among all persons in the immediate vicinity. METHODS: This randomized, double-blind controlled trial used 0.4% permethrin (2.5% permethrin diluted by 7 parts water) sprayed once onto the external surface of canvas tents at a Boy Scout summer camp. During a 9-week period, subjects (n

Does Zika virus infection affect mosquito response to repellents?

The World Health Organization (WHO) recommends that people travelling to or living in areas with Zika virus (ZIKV) outbreaks or epidemics adopt prophylactic measures to reduce or eliminate mosquito bites, including the use of insect repellents. It is, however, unknown whether repellents are effective against ZIKV-infected mosquitoes, in part because of the ethical concerns related to exposing a human subject's arm to infected mosquitoes in the standard arm-in-cage assay. We used a previously developed, human subject-free behavioural assay, which mimics a human subject to evaluate the top two r…

[Individual protection against insect vectors].

Many diseases for which no vaccine is available are transmitted by insect and arthropod vectors, the main exceptions being yellow fever and Japanese encephalitis B. Treatment is less and less effective due to the development of chemoresistance to therapeutic and prophylactic drugs as is well-illustrated by malaria. One of the best methods of preventing these diseases is personal protection against insect bites. Personal protection measures can be divided into three categories which can be used separately or in combination : application of repellents to the skin, wearing clothes impregnated wit

Health risk assessment of trace elements in plant-based cosmetics and personal care products using inductively coupled plasma-mass spectrometry (ICP-MS).

The increasing demand for plant-based cosmetics and personal care products (CPCPs) has raised concerns regarding trace element contamination stemming from agricultural practices and industrial pollution. In this study, 39 plant-based CPCPs, including deodorants, sunscreens, face creams, lip moisturizers, body lotions, and insect repellents, were analyzed to determine the concentration of 14 trace elements (Al, V, Cr, Mn, Fe, Co, Ni, Zn, As, Se, Mo, Cd, Hg, and Pb) using microwave-assisted digestion and inductively coupled plasma-mass spectrometry (ICP-MS). The method demonstrated excellent lin…

Ethnobotanical study of some of mosquito repellent plants in north-eastern Tanzania.

BACKGROUND: The use of plant repellents against nuisance biting insects is common and its potential for malaria vector control requires evaluation in areas with different level of malaria endemicity. The essential oils of Ocimum suave and Ocimum kilimandscharicum were evaluated against malaria vectors in north-eastern Tanzania. METHODOLOGY: An ethnobotanical study was conducted at Moshi in Kilimanjaro region north-eastern Tanzania, through interviews, to investigate the range of species of plants used as insect repellents. Also, bioassays were used to evaluate the protective potential of sele…

[Antihistaminic effect of a hydrogel with the components diphenhydramine, N,N-diethyl-m-toluamide and phthalic acid dimethyl ester].

The combination of the antihistamine diphenhydramine as well as the repellents N,N-diethyl-m-toluamide and dimethylphthalate as the active constituents of an insect repellent (Pellit) guarantees a remarkable antihistaminic effect. Surprisingly the repellent N,N-diethyl-m-toluamide proved itself to be an antihistamine. The UV-erythema model was used on the dorsal skin of male volunteers.

Design, Synthesis, Antifeedant Activity, and Mechanistic Research of Aconitine Derivatives.

For decades, natural product-derived insect repellents have been a major focus of research. Aconitine, a diterpene alkaloid isolated from Aconitum plants, exhibits not only significant antifeedant activity but also potent toxicological effects against certain insects. In this study, 48 novel aconitine derivatives were synthesized via esterification of the C3-hydroxyl group. Among them, compound A4 demonstrated optimum antifeedant activity with an AFC50 value of 4.91 µg/cm2, surpassing that of the parent compound aconitine (AFC50 = 11.65 µg/cm2) and the positive control Azadirachtin (AFC50 = 5.…

[Prevention with repellent in children].

Use of topical insect repellent is an important component in prophylaxis of arthropod bite vector borne diseases. Topical insect repellent are used in a three part management regimen, along with impregnated clothing and mosquito netting. Parental training for efficacious and secure use of repellents for their children is essential part of a successful strategy to combat Lyme borreliosis, dengue fever, Chikungunya, West Nile virus infection and malaria, amongst children, according to local epidemiological risks. Rational repellent prescription for a child must take into account age, active subs

Multiple channels of DEET repellency in Drosophila.

BACKGROUND: N,N-Diethyl-meta-toluamide (DEET) is the prophylactic insect repellent used most widely to inhibit insect bites. Despite its use since 1944, the mechanism of DEET repellency remains controversial. Here, we revisited the role of smell and taste in DEET repellence using Drosophila as a model. RESULTS: Analysis of the responses of individual olfactory receptor neuron (ORN) classes to DEET reveals that 11 ORNs are activated and two are inhibited by this compound. Blocking individual ORN classes in the antenna does not block DEET repellence. This argues against the existence of a singl

Geminiviral C2 proteins promote virus transmission by subverting plant defenses upon infestation of non-vector herbivores.

BACKGROUND: Insect vectors are responsible for transmitting the majority of plant viruses. While the interplays between insect vectors and viruses have been extensively investigated, how other organisms associated with the shared host plants of the insect and virus impact virus transmission dynamics, and the corresponding viral countermeasures, remain largely unknown. RESULTS: We explored the modulation of whitefly-mediated transmission of geminiviruses by two non-vector herbivores and the role of viral C2 proteins. The infestation of cotton bollworm induced the accumulation of jasmonic acid

From Molecular Recognition to Behavioral Control: Dual-Target Aphid Repellent Discovery of Novel Ionone Derivatives.

Odorant-binding proteins (OBPs) are promising targets for insect behavior regulators. We established an original target-to-behavior strategy for developing dual-target aphid repellents. Structure-based virtual screening initially identified β-ionone as a lead compound, from which 24 novel ionone derivatives were designed and synthesized targeting Acyrthosiphon pisum OBP3 and OBP7. Most ionone derivatives exhibited enhanced binding affinities to both ApisOBP3 (5.14-20.70 μM) and ApisOBP7 (3.64-28.68 μM). Molecular dynamics simulations and alanine site-directed mutagenesis revealed that mutation

What can we do about the Zika virus in the United States?

Control measures include using insect repellents, aerial spraying of insecticides, eliminating mosquito breeding sites, covering water tanks, and using mosquito nets or door and window screens. Infection during pregnancy is the greatest concern because of congenital anomalies (including microcephaly) that negatively affect brain development.

Pruritic Horse: Approach to Allergic Skin Diseases in Horses.

Allergy to insects is the most common skin allergy in horses. Pruritus in affected patients can be extreme. Face, ears, mane, and tail area are commonly affected areas. Diagnosis of insect bite hypersensitivity (IBH) is clinical and is based on history, clinical signs, and response to repellents. Allergy tests are not to be used for diagnostic purposes. Currently, there is no specific treatment for IBH other than insect avoidance, treatment of secondary infections, and symptomatic relief of pruritus. Many allergic horses become also sensitized to pollens. For these patients, allergen specific

Safe chemical repellents to prevent the spread of invasive ants.

BACKGROUND: The red imported fire ant, Solenopsis invicta, is one of the most dangerous invasive species in the world. Fire ants can spread by hiding among plant material and soil that are transported from infested areas in vehicles and vessels. Therefore, efficient repellents may be used to prevent fire ants from infesting transported goods. Although some fire ant repellants have been identified, novel, cost-effective and environmentally friendly materials for fire ant control are still needed. Recent studies with other model insects have suggested readily available, non-toxic alternatives (e

Why is it crucial to test anti-lice repellents?

It is difficult to stop lice propagation just by treating infested heads, since reinfections are possible just a few hours after a successful elimination of all lice from a child's head by application of an active anti-louse product. Therefore, several products have been developed that claim to have a louse repellent activity; however, definite proofs are scarce. The present study involving two louse repellents (Linicin® Preventive Spray, Picksan® NoLice) and three substances (at 10% dilution) known for their general repellency activity shows that there are much more difficulties to repel lice

Odorant receptor-based discovery of natural repellents of human lice.

The body louse, Pediculus humanus humanus, is an obligate blood-feeding ectoparasite and an important insect vector that mediates the transmission of diseases to humans. The analysis of the body louse genome revealed a drastic reduction of the chemosensory gene repertoires when compared to other insects, suggesting specific olfactory adaptations to host specialization and permanent parasitic lifestyle. Here, we present for the first time functional evidence for the role of odorant receptors (ORs) in this insect, with the objective to gain insight into the chemical ecology of this vector. We id
