Emerging Mosquito Resistance to Piperonyl Butoxide-Synergized Pyrethroid Insecticide and Its Mechanism.

Piperonyl butoxide (PBO)-synergized pyrethroid products are widely available for the control of pyrethroid-resistant mosquitoes. To date, no study has examined mosquito resistance after pre-exposure to PBO and subsequent enzymatic activity when exposed to PBO-synergized insecticides. We used Culex quinquefasciatus Say (Diptera: Culicidae), an important vector of arboviruses and lymphatic filariasis, as a model to examine the insecticide resistance mechanisms of mosquitoes to PBO-synergized pyrethroid using modified World Health Organization tube bioassays and biochemical analysis of metabolic …

Genetic structure and insecticide resistance characteristics of fall armyworm populations invading China.

The rapid wide-scale spread of fall armyworm (Spodoptera frugiperda) has caused serious crop losses globally. However, differences in the genetic background of subpopulations and the mechanisms of rapid adaptation behind the invasion are still not well understood. Here we report the assembly of a 390.38-Mb chromosome-level genome of fall armyworm derived from south-central Africa using Pacific Bioscience (PacBio) and Hi-C sequencing technologies, with scaffold N50 of 12.9 Mb and containing 22,260 annotated protein-coding genes. Genome-wide resequencing of 103 samples and strain identification

Insecticide resistance genes affect Culex quinquefasciatus vector competence for West Nile virus.

Insecticide resistance has been reported to impact the interactions between mosquitoes and the pathogens they transmit. However, the effect on vector competence for arboviruses still remained to be investigated. We examined the influence of two insecticide resistance mechanisms on vector competence of the mosquito Culex quinquefasciatus for two arboviruses, Rift Valley Fever virus (RVFV) and West Nile virus (WNV). Three Cx. quinquefasciatus lines sharing a common genetic background were used: two insecticide-resistant lines, one homozygous for amplification of the Ester2 locus (SA2), the other…

RDL mutations predict multiple insecticide resistance in Anopheles sinensis in Guangxi, China.

BACKGROUND: Anopheles sinensis is a major vector of malaria in China. The gamma-aminobutyric acid (GABA)-gated chloride channel, encoded by the RDL (Resistant to dieldrin) gene, is the important target for insecticides of widely varied structures. The use of various insecticides in agriculture and vector control has inevitably led to the development of insecticide resistance, which may reduce the control effectiveness. Therefore, it is important to investigate the presence and distribution frequency of the resistance related mutation(s) in An. sinensis RDL to predict resistance to both the wit…

Changes in insecticide resistance of the rice striped stem borer (Lepidoptera: Crambidae).

Application of insecticides is the most important method to control Chilo suppressalis (Walker) (Lepidoptera: Crambidae), and continuous use of individual insecticides has driven the rapid development of insecticide resistance in C. suppressalis during the past 30 yr. Monitoring insecticide resistance provides information essential for integrated pest management. Insecticide resistance of field populations to monosultap, triazophos, chlorpyrifos, and abamectin in China was examined in 2010 and 2011. The results indicated that the resistance levels of 14 field populations to four insecticides w…

Insecticide resistance in Culex quinquefasciatus from Zanzibar: implications for vector control programmes.

BACKGROUND: Zanzibar has a long history of lymphatic filariasis (LF) caused by the filarial parasite Wuchereria bancrofti, and transmitted by the mosquito Culex quinquefasciatus Say. The LF Programme in Zanzibar has successfully implemented mass drug administration (MDA) to interrupt transmission, and is now in the elimination phase. Monitoring infections in mosquitoes, and assessing the potential role of interventions such as vector control, is important in case the disease re-emerges as a public health problem. Here, we examine Culex mosquito species from the two main islands to detect W. ba

[Insecticide resistance mechanisms of Aedes aegypti (Diptera: Culicidae) from two Peruvian provinces].

Insecticide resistance of Aedes aegypti larvae and adults from two Peruvian provinces, that is, Trujillo and Tumbes provinces, was conducted. High infestation indexes and extensive use of insecticides based on the Vector Surveillance and Control Strategy of the Ministry of Public Health prevailed in these places. Larval bioassays revealed susceptibility to organophosphorate insecticide called malathion in TRUJILLO strain, it being moderate to fention and fenitrotion and high to chlorpyriphos and temephos;however, TUMBES strain was susceptible to the evaluated organophosphorate compounds, excep…

Investigating insecticide resistance in eggplant fruit and shoot borer: Multi-class insecticides and detoxification gene expression.

Eggplant fruit and shoot borer, Leucinodes orbonalis (Lepidoptera: Crambidae), is a destructive borer pest that infests eggplant crop from nursery stage to fruiting. This study aimed to evaluate the current insecticide resistance status of L. orbonalis collected from ten different locations of Tamil Nadu, India, using a fruit dip bioassay. Late second instar larvae were exposed to spinosad, spinetoram, thiacloprid and emamectin benzoate. Median lethal concentration (LC50) and resistance ratios (RR) were estimated for each population using a susceptible NBAIR population as a reference. The resu…

The gut microbiota of insecticide-resistant insects houses insecticide-degrading bacteria: A potential source for biotechnological exploitation.

The exploration of new niches for microorganisms capable of degrading recalcitrant molecules is still required. We hypothesized the gut microbiota associated with insect-resistant lines carry pesticide degrading bacteria, and predicted they carry bacteria selected to degrade pesticides they were resistant to. We isolated and accessed the pesticide-degrading capacity of gut bacteria from the gut of fifth instars of Spodoptera frugiperda strains resistant to lambda-cyhalothrin, deltamethrin, chlorpyrifos ethyl, spinosad and lufenuron, using insecticide-selective media. Sixteen isolates belonging

Role of cytochrome P450s in insecticide resistance: impact on the control of mosquito-borne diseases and use of insecticides on Earth.

The fight against diseases spread by mosquitoes and other insects has enormous environmental, economic and social consequences. Chemical insecticides remain the first line of defence but the control of diseases, especially malaria and dengue fever, is being increasingly undermined by insecticide resistance. Mosquitoes have a large repertoire of P450s (over 100 genes). By pinpointing the key enzymes associated with insecticide resistance we can begin to develop new tools to aid the implementation of control interventions and reduce their environmental impact on Earth. Recent technological advan…

Neuroactive insecticides: targets, selectivity, resistance, and secondary effects.

Neuroactive insecticides are the principal means of protecting crops, people, livestock, and pets from pest insect attack and disease transmission. Currently, the four major nerve targets are acetylcholinesterase for organophosphates and methylcarbamates, the nicotinic acetylcholine receptor for neonicotinoids, the γ-aminobutyric acid receptor/chloride channel for polychlorocyclohexanes and fiproles, and the voltage-gated sodium channel for pyrethroids and dichlorodiphenyltrichloroethane. Species selectivity and acquired resistance are attributable in part to structural differences in binding

Laboratory wash resistance of long-lasting insecticidal nets.

Long-lasting insecticidal nets (LLINs) may eliminate the need for retreatment of mosquito nets used for the control of malaria and other vector-borne diseases. The efficacy of LLINs after repeated washing under laboratory conditions has been used to predict long-lasting efficacy under field conditions. We evaluated under laboratory conditions the wash resistance of two LLINs (PermaNet 1.0, Vestergaard-Frandsen, Denmark; Olyset, Sumitomo Chemical Co., Japan), two candidate LLINs (Dawa, Siamdutch Mosquito Netting Co., Thailand; Insector, Athanor, France) and a net treated with a process designed

Metabolic resistance drives cross-resistance in Aedes albopictus to pyriproxyfen and pyrethroid insecticides.

Aedes albopictus is a major global vector of arboviruses. Pyriproxyfen, an insect growth regulator, is increasingly used for mosquito control, but the potential for resistance and cross-resistance with pyrethroids poses a serious threat. Through 13 generations of laboratory selection, we established a highly pyriproxyfen-resistant Ae. albopictus strain (Lab-R, RR50 = 10.12). This strain developed moderate to high cross-resistance to the permethrin, deltamethrin, and beta-cypermethrin (RR50 = 8.11-10.30). Resistance was associated with significant fitness costs, including prolonged larval devel

Cross-resistance to insecticides in a malathion-resistant strain of Ceratitis capitata (Diptera: Tephritidae).

Resistance to malathion has been reported in field populations of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), in areas of Spain where an intensive use of this insecticide was maintained for several years. The main goal of this study was to determine whether resistance to malathion confers cross-resistance to different types of insecticides. Susceptibility bioassays showed that the malathion-resistant W-4Km strain (176-fold more resistant to malathion than the susceptible C strain) has moderate levels of cross-resistance (three- to 16-fold) to other organ

Biochemical and insecticidal effects of plant essential oils on insecticide resistant and susceptible populations of Musca domestica L. point to a potential cross-resistance risk.

Essential oils (EOs) can provide important alternatives to chemical insecticides in the control of pests. In this study, 12 EOs of native plant species from Iran were evaluated for their adulticidal activity against the house fly. In addition, we examined the insecticidal activity of Zataria multiflora and Rosmarinus officinalis EOs on adult female house flies from pyrethroid and organophosphate resistant and susceptible populations, using both fumigant and topical bioassays. The involvement of detoxification enzymes in susceptibility was investigated with synergism experiments in vivo, while

Dengue fever and insecticide resistance in Aedes mosquitoes in Southeast Asia: a review.

Dengue fever is the most important mosquito-borne viral disease in Southeast Asia. Insecticides remain the most effective vector control approach for Aedes mosquitoes. Four main classes of insecticides are widely used for mosquito control: organochlorines, organophosphates, pyrethroids and carbamates. Here, we review the distribution of dengue fever from 2000 to 2020 and its associated mortality in Southeast Asian countries, and we gather evidence on the trend of insecticide resistance and its distribution in these countries since 2000, summarising the mechanisms involved. The prevalence of re…
