Preacetylation of Kraft Lignin Facilitates Acid-Catalyzed Depolymerization in Acidic Deep Eutectic Solvents to Obtain Lignin Oil With High Antioxidant Activity.

Kraft lignin, an abundant renewable aromatic polymer, has attracted increasing interest for acid-catalyzed depolymerization to obtain lignin oil with high antioxidant activity. However, this process is plagued by severe condensation of reactive intermediates, leading to low lignin oil yields. To address this critical issue, we performed preacetylation modification on hardwood kraft lignin (HKL) and investigated its depolymerization behavior in acidic deep eutectic solvents (DESs). Furthermore, the underlying mechanism by which acetylation promotes depolymerization was clarified using lignin mo

Development and structural comparison of alkaline and organosolv coconut husks lignin as an eco-friendly lignin-phenol-glyoxal (LPG) wood adhesives.

The development of eco-friendly wood adhesives have gained more interest among adhesives industries due to the concerns about using carcinogenic formaldehyde and petroleum-based phenol in commercially available adhesives. Therefore, many studies have been done by using lignin to partially replace phenol and completely substitute formaldehyde with non-toxic glyoxal in a wood adhesive formulation. This study focused on using different percentages of lignin substitution (10 %, 30 % and 50 wt%) of alkaline and organosolv coconut husk lignin into soda lignin-phenol-glyoxal (SLPG), Kraft lignin-phen

Role of in situ surfactant modification of lignin structure and surface properties during glycerol pretreatment in modulating cellulase-lignin binding affinities.

Surfactants are effective agents for enhancing lignocellulosic pretreatment, synergistically modifying lignin with polyols to improve substrate hydrolyzability while achieving comparable delignification. However, the mechanisms underlying the multiple modifications from dual in situ surfactant/polyols grafting that passivate lignin-cellulase interactions and their core affecting factors remain unclear. Following the previously developed polyethylene glycol (PEG) and Triton-assisted pretreatment, the intrinsic correlation among lignin structures, physical barriers, and cellulase interactions wa

Co-upgrading of ethanol-assisted depolymerized lignin: A new biological lignin valorization approach for the production of protocatechuic acid and polyhydroxyalkanoic acid.

This study presents a promising biological co-upgrading of ethanol-assisted depolymerized lignin (EDL) into protocatechuic acid (PCA) and polyhydroxyalkanoic acid (PHA) without any separation process. A depolymerized alkali lignin containing various G-lignin-type monomers at a concentration of 77 mg/mL was used for co-upgrading. An engineered Pseudomonas putida KT2440 strain was constructed by knocking out the protocatechuate 3, 4-dioxygenase, expression of the formaldehyde utilization pathway, and the expression of aldehyde dehydrogenase to enhance the efficiency of the ethanol utilization pa

A knockout mutation in the lignin biosynthesis gene CCR1 explains a major QTL for acid detergent lignin content in Brassica napus seeds.

Seed coat phenolic compounds represent important antinutritive fibre components that cause a considerable reduction in value of seed meals from oilseed rape (Brassica napus). The nutritionally most important fibre compound is acid detergent lignin (ADL), to which a significant contribution is made by phenylpropanoid-derived lignin precursors. In this study, we used bulked-segregant analysis in a population of recombinant inbred lines (RILs) from a cross of the Chinese oilseed rape lines GH06 (yellow seed, low ADL) and P174 (black seed, high ADL) to identify markers with tight linkage to a majo

Effect of Dilute Acid Pretreatment and Lignin Extraction Conditions on Lignin Properties and Suitability as a Phenol Replacement in Phenol-Formaldehyde Wood Adhesives.

Corn stover was subjected to dilute sulfuric acid pretreatment to assess the impact of pretreatment conditions on lignin extractability, properties, and utility as a phenol replacement in wood phenol-formaldehyde (PF) adhesives. It was identified that both formic acid and NaOH could extract and recover 60-70% of the lignin remaining after pretreatment and enzymatic hydrolysis under the mildest pretreatment conditions while simultaneously achieving reasonable enzymatic hydrolysis yields (>60%). The availability of reaction sites for the incorporation of lignins into the PF polymer matrix (i.e.,

Brucella ligniniphila sp. nov., a Novel Lignin-Degrading Bacterium from Decaying Bamboo, and Synergistic Lignin Biodegradation by Consortium with White-Rot Fungi.

A lignin-degrading bacterium, strain BE17T, was isolated from decaying bamboo and identified as a novel species within the genus Brucella, proposed as Brucella ligniniphila, sp. nov. (type strain = MCCC 1K08845T = JCM 36600T). BE17T exhibited a lignin degradation rate of 7.9% with lignin as the sole carbon source, which increased to 17.2% upon glucose supplementation. Structural analysis indicated that BE17T degraded lignin by cleaving β-O-4 bonds, oxidizing aliphatic side chains and disrupting aromatic rings. Low-molecular-weight lignin fractions were more readily degraded. Genome analysis re

Influence of Lignin Content and Pressing Time on Plywood Properties Bonded with Cold-Setting Adhesive Based on Poly (Vinyl Alcohol), Lignin, and Hexamine.

The sustainability, performance, and cost of production in the plywood industry depend on wood adhesives and the hot-pressing process. In this study, a cold-setting plywood adhesive was developed based on polyvinyl alcohol (PVOH), high-purity lignin, and hexamine. The influence of lignin content (10%, 15%, and 20%) and cold-pressing time (3, 6, 12, and 24 h) on cohesion, adhesion, and formaldehyde emission of plywood were investigated through physical, chemical, thermal, and mechanical analyses. The increased lignin addition level lowered the solids content, which resulted in reduced average v

Novel catalytic potential of a hyperthermostable mono‑copper oxidase (LPMO-AOAA17) for the oxidation of lignin monomers and depolymerisation of lignin dimer in aqueous media.

Lytic polysaccharide monooxygenase (LPMO) are mono‑copper enzymes known for the oxidative cleavage of recalcitrant polysaccharides with their intriguing and unique catalytic chemistry. Such impeccable oxidation potential has made them highly valuable in the enzymatic consortia for the degradation of ligno-cellulosic biomass. Bioinformatic analysis has revealed an unannotated LPMO gene in the genome of A. oryzae. Multiple sequence alignment showed the presence of conserved "histidine brace" of LPMO in the amino acid sequence of the enzyme. The enzyme, named as LPMO-AOAA17 was recombinantly expr

Engineering lignin-tannin nanoassemblies for multifunctional and sustainable skincare.

Lignin is increasingly recognized as a sustainable raw material for the development of safe and multifunctional cosmetic ingredients. Nanoengineering strategies enable its transformation into uniform colloidal particles with improved dispersibility and functionality. However, reducing its intense color without compromising its intrinsic bioactivity remains a major challenge for cosmetic applications. In this work, a function-preserving particle design strategy was developed by synergistically combining lignin with tannins at the nanoscale to create fully biobased phenolic colloidal assemblies.

Lignin-Based Copper Nanoparticles for Green and Flexible Electronics.

Copper nanoparticles offer a cost-effective and sustainable alternative to silver-based materials for conductive inks in printed and flexible electronics. However, their practical application is often hindered by the need for inert atmospheres and environmentally hazardous reducing agents, but most critically by their rapid postsynthesis oxidation. In this work, we report a green, scalable synthesis of oxidation-resistant metallic Cu nanoparticles via a microwave-assisted polyol method using sodium hypophosphite as a nontoxic reducing agent and lignin as a capping and stabilizing agent. Polyphenol Oxidase Activity on Guaiacyl and Syringyl Lignin Units.

The natural heterogeneity of guaiacyl (G) and syringyl (S) compounds resulting from lignin processing hampers their direct use as plant-based chemicals and materials. Herein, we explore six short polyphenol oxidases (PPOs) from lignocellulose-degrading ascomycetes for their capacity to react with G-type and S-type phenolic compounds. All six PPOs catalyze the ortho-hydroxylation of G-type compounds (guaiacol, vanillic acid, and ferulic acid), forming the corresponding methoxy-ortho-diphenols. Remarkably, a subset of these PPOs is also active towards S-compounds (syringol, syringic acid, and si

Characterization and properties of phenolic resin doped modified lignin.

Phenolic resins occupy an important position in industrial applications, but phenol, one of the raw materials for synthesis, is a non-renewable resource. Lignin, as a natural polymer containing phenolic hydroxyl groups, alcohol hydroxyl groups and other reactive groups, can replace some of the phenol in the synthesis of phenolic resins, which can reduce the amount of phenol, thus reducing the cost of phenolic resins, while effectively promoting the high value-added use of renewable biomass resources. Due to its low reactivity, alkaline lignin is usually discharged as production waste, unaware

High-value utilization of hydroxymethylated lignin in polyurethane adhesives.

Lignin is a good candidate for the polymerization and chemical modification to prepare sustainable chemicals and materials, but a relatively low hydroxyl content becomes an obstacle for the preparation of lignin-based polyurethane (PU) adhesives. In order to improve its reactivity, the acetic acid lignin (AAL) was hydroxymethylated before copolymerized with isocyanate during the preparation of PU adhesives. The hydroxymethylation was carried out in an alkaline formaldehyde solution and it was found that 85 °C is the optimal temperature. On that condition, the free formaldehyde content of the c

Promoting Lignin Valorization by Coping with Toxic C1 Byproducts.

The toxic C1 compounds methanol and formaldehyde are generated during bioconversion of lignin into value-added chemicals. These toxins can be detoxified and assimilated by methylotrophs to synthesize useful metabolites and cell biomass. We discuss the promising future of constructing integrated biosystems to use toxic C1 byproducts and promote lignin valorization.

Linking phenylpropanoid metabolism, lignin deposition, and plant growth inhibition.

Lignin, a polymer found in the plant secondary cell wall, is a major contributor to biomass' recalcitrance toward saccharification. Because of this negative impact toward the value of lignocellulosic crops, there is a special interest in modifying the content and composition of this important plant biopolymer. For many years this endeavor has been hindered by the plant growth inhibition that is often associated with manipulations to phenylpropanoid metabolism. Although the actual mechanism by which dwarfism arises remains unknown, recent advances in tissue-specific lignin complementation and b

From Byproduct to Platform: Can Kraft Lignin Outcompete Lignosulfonates?

Lignosulfonates (LS) have long set the standard of dispersants for construction, agrochemical, oilfield, and animal feed industries, due to their amphiphilic structurearomatic domains linked with sulfonated functionalities, which result in efficient and cost-effective colloidal stabilization. Nevertheless, LS production is intrinsically limited by the falloff in sulfite pulping, which is today only a minor percentage of the world's pulp manufacturing. On the other hand, kraft lignin (KL) is produced at multimillion-tonne levels annually and is now being acknowledged as a renewable aromatic po

STRONG3 promotes lignin biosynthesis and culm strength in rice.

Lodging severely reduces rice yield and quality. Culm strength, mainly determined by lignin biosynthesis and secondary cell wall (SCW) formation, is core to lodging resistance. We identified an ethylene response factor (ERF) transcription factor STRONG3 via genome-wide association study (GWAS) of 405 rice accessions based on lignin content. Functional validation demonstrated that STRONG3 positively regulates lignin accumulation, culm diameter, and mechanical strength by promoting sclerenchyma tissue development, vascular bundle enlargement, and SCW thickening. Molecular analyses revealed that
