Mn(I)-Catalyzed Site-Selective C-H Activation: Unlocking Access to 3-Arylated Succinimides from 2-Arylpyridines and Maleimides.

An efficient and cost-effective Mn(I)-catalyzed site-selective C-H activation of 2-arylpyridines with maleimides has been described. This approach facilitates the synthesis of 3-arylated succinimide derivatives with high site selectivity, chemoselectivity, catalytic efficiency, and outstanding tolerance to numerous functional groups. The practicality of this approach is further evidenced by its successful application in large-scale reactions and the conversion of the synthesized succinimide derivatives into other valuable compounds.

Engineering Poly(ε-caprolactone)-Based Maleimide-Enediyne Micelles for Next-Generation Anticancer Therapies.

Inspired by the DNA-cleaving potency of natural enediynes such as calicheamicin, we report the design and synthesis of maleimide-enediyne conjugates integrated into poly(ε-caprolactone) (PCL) as a next-generation platform for anticancer drug delivery. Maleimide-based enediynes typically suffer from poor aqueous solubility and nonspecific toxicity, addressed here by covalently incorporating enediyne groups into amphiphilic PCL block copolymers via post-polymerization functionalization. The resulting polymers form ∼120 nm spherical micelles with a critical micelle concentration of 9.1 × 10-4 mg

Preparation of Targeted Lipid Nanoparticles by Insertion of Either Tetrazine-trans-Cyclooctene or Thiol-Maleimide Micelles.

Lipid nanoparticles (LNPs) have emerged as a promising nonviral nucleic acid delivery platform for clinical use. To expand LNPs as a treatment option in nonhepatic-based diseases, LNPs surface coated with targeting moieties produce a precise modular delivery method that can bind to and be internalized by specific receptors expressed on target cells. This study showcases the tetrazine-trans-cyclooctene inverse electron-demand Diels-Alder click reaction and directly compares its performance with the widely employed thiol-maleimide conjugation. We also compare direct mixing and micelle mixing ins

Surface modification of keratin films via thiol-maleimide chemistry toward enhanced anticoagulant response.

Protein-based biomaterials are widely used in tissue engineering and regenerative medicine; however, many conventional materials suffer from limitations including immunogenicity, low stability, and risks associated with animal-derived sources. In this context, keratin derived from human hair has attracted increasing attention due to its inherent biocompatibility and cysteine-rich structure, which provides reactive thiol (-SH) groups suitable for chemical modification. In the present study, reductively extracted keratin was utilized to preserve thiol functionality, enabling site-selective modif

Synthesis of polymeric monoliths via thiol-maleimide polymerization reaction for highly efficient chromatographic separation.

One-step thiol-maleimide polymerization reaction was firstly adopted for direct preparation of polymeric monoliths via alkaline-catalyzed reaction of 4,4'-bis(maleimidophenyl)methane (BMI) and trimethylolpropane tris(3-mercaptopropionate) (3SH)/pentaerythriol tetra(3-mercaptopropionate) (4SH) in the presence of a small amount of triethylamine (TEA). The polymerization could be performed within 3h, which was faster than thermal-initiated free radical polymerization. Two kinds of monoliths, poly(BMI-co-3SH) (marked as I) and poly(BMI-co-4SH) (marked as II), were characterized with scanning elect

Improved harmonization of eosin-5-maleimide binding test across different instruments and age groups.

BACKGROUND: The eosin-5'maleimide (EMA) binding test has been studied extensively for the detection of hereditary spherocytosis (HS). Its performance characteristics have been compared to NaCl-based or glycerol lysis-based red cell osmotic fragility tests and cryohemolysis. HS samples are also better identified when both mean channel fluorescence (MCF) of EMA relative to controls and the coefficient of variation (CV) are analyzed. METHODS: We looked at 65 normal controls including 30 adults 25-65 years old and 35 newborns and 12 HS cases. In addition to the MCF and the CV, we used a side scat

Cascade Construction of Cinnoline and Furanone Skeletons from Phenylhydrazine and Maleimide Featuring a Formal Oxygen Swap.

Both cinnoline and furanone are the pivotal skeletons of numerous natural products and pharmaceuticals. While a number of reliable methods for the respective synthesis of cinnoline or furanone derivatives are currently available, concurrent construction of these two privileged scaffolds in one pot has not been disclosed. We report herein a concise and convenient synthesis of cinnoline fused furanone from the easily obtainable and economically sustainable phenylhydrazine and maleimide. Mechanistically, the formation of product involves C-H bond activation-initiated cinnoline ring forming along

S-Nitrosothiols as Thiol-Protecting Groups for Controlled Thiol-Maleimide Crosslinking of Homogeneous Soft Hydrogels.

The thiol-maleimide Michael-type addition is used in bioconjugation and hydrogel crosslinking for its chemoselectivity and rapid kinetics under physiological conditions. However, this same reactivity limits its use for soft hydrogels, as gelation often proceeds faster than precursor mixing, leading to spatially heterogeneous networks. Here, S-nitrosothiols (RSNOs) are introduced as thiol-protecting groups that suppress premature thiol-maleimide coupling and enable homogeneous mixing of polymer precursors prior to on-demand crosslinking. Despite common assumptions about RSNO instability, RSNO-m

Mn(I)-Catalyzed Preferential Electrophilic C3-Maleimidation in Quinoxaline Leading to Spirocyclization and Dehydrogenation of Succinimides.

A Mn(I)-catalyzed site-selective nondirected C3-maleimidation of quinoxaline is established. Herein, the electrophilic C3-metalation precedes over the o-directed strategy to access diversely substituted quinoxaline-appended succinimides. The products undergo PIFA-promoted C(sp2)-C(sp3) spirocyclization via π-electrons drifting from aryls and Selectfluor-mediated dehydrogenation of succinimide at room temperature.

Toward the harmonization of result presentation for the eosin-5'-maleimide binding test in the diagnosis of hereditary spherocytosis.

BACKGROUND: The eosin-5'-maleimide (EMA) Binding test measures reduced mean channel fluorescence (MCF) reading of EMA-labeled red cells (EMA-RBCs) from patients with hereditary spherocytosis (HS). Reporting test results can be either in the actual MCF reading or as a ratio by normalization of the test MCF result to the mean MCF value of six normal controls. The latter format has potential for universal reporting. METHODS: We analyzed three years' archival MCF data from HS and non-HS patient groups for establishment of reference ranges of ratios for normal adults and HS. A prospective study us

Synthesis of Succinimide Spiro-Fused Sultams from the Reaction of N-(Phenylsulfonyl)acetamides with Maleimides via C(sp2)-H Activation.

Presented herein is an effective preparation of succinimide spiro-fused sultams through the coupling reaction of N-(phenylsulfonyl)acetamides with maleimides. It is deduced that this reaction should proceed through a cascade process including Rh(III)-catalyzed C(sp2)-H bond cleavage of N-(phenylsulfonyl)acetamide, maleimide double bond insertion into the C-Rh bond, β-hydride elimination, reductive elimination, and intramolecular aza-Michael addition. Notably, this cascade procedure features simultaneous annulation and spirocyclization through traceless fusion of the directing group into target…

Tools for the Design of Diels-Alder-Based Reversible Polymer Systems: Reactivity and Reversibility of Furan/Maleimide Adducts.

The equilibrium conversion of 16 model furan derivatives in Diels-Alder (DA) reactions with an aromatic bis-maleimide was investigated across the temperature range of 30-110 °C. The impact of furan substituents on the reactivity and reversibility of DA adducts was systematically investigated for establishing a useful toolbox of structure-thermal property relationships and clear guidelines for designing reversible furan-maleimide DA-based polymers. Two strategies were considered for furan integration in a polymer network. Monosubstituted furans mimicking grafted-onto moieties were first categor

RAFT Polymerization of Styrene and Maleimide in the Presence of Fluoroalcohol: Hydrogen Bonding Effects with Classical Alternating Copolymerization as Reference.

The impacts of hydrogen bonding on polymerization behavior has been of interest for a long time; however, universality and in-depth understanding are still lacking. For the first time, the effect of hydrogen bonding on the classical alternating-type copolymerization of styrene and maleimide was explored. N-phenylmaleimide (N-PMI)/styrene was chosen as a model monomer pair in the presence of hydrogen bonding donor solvent 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP), which interacted with N-PMI via hydrogen bonding. Reversible addition-fragmentation chain transfer polymerization (RAFT) technique wa

Stability of eosin-5'-maleimide dye used in flow cytometric analysis for red cell membrane disorders.

BACKGROUND: The eosin-5'-maleimide (EMA) binding test using flow cytometry is a common method to measure reduced mean channel fluorescence (MCF) of EMA-labeled red blood cells (RBCs) from patients with red cell membrane disorders. The basic principle of the EMA-RBC binding test involves the covalent binding of EMA to lysine-430 on the first extracellular loop of band 3 protein. METHODS: In the present study, the MCF of EMA was analyzed for samples derived from 12 healthy volunteers (controls) to determine the stability (i.e., the percentage decrease in fluorescence) of EMA over a period of 1
