A study of Mansonella perstans filariasis conducted in the Chaillu mountains, Southern Congo, showed that 108 of 134 Pygmies (80.6%) and 79 of 302 Bantus (26.2%) presented with microfilaraemia. The mean microfilarial densities were also significantly higher in the Pygmies (1213 ml-1 of blood) than in the Bantus (136 ml-1). Ninety eight per cent of the Culicoides taken which had bitten man in the daytime were C. grahamii, and 0.8% of these were infected with filarial larvae. Two other species of Culicoides (C. kumbaensis and C. rutshuruensis) might also play a role in the transmission of M. per…

One hundred and twelve inhabitants of an area endemic for M. ozzardi received 20 cmm capillary blood samples by finger prick before and 30 minutes after the ingestion of 100 mg of diethylcarbamazine citrate. No significant variations in microfilaremia were detected. Thus, a provocative dose of DEC would not appear to be helpful in sampling for mansonellosis in Haiti.

Experimental studies in Bayeux, Haiti showed that the biting midge, Leptoconops bequaerti, is capable of supporting the complete development of Mansonella ozzardi but only on a very limited scale. This suggests that the species may not be involved in the natural transmission cycle despite its abundance and pestiferous nature in certain areas of Haiti. A midge-holding container is described which markedly enhanced the survival of engorged L. bequaerti in the laboratory.

The colonized midge, Culicoides variipennis, is shown to be a competent laboratory host for the Amazon form of Mansonella ozzardi. It raises the possibility that certain Culicoides spp. in the Amazon region may support development of the parasite and possibly play a role in the natural transmission cycle.

The prevalence of Mansonella ozzardi in the Comisaría del Vaupés of eastern Colombia was investigated as part of an extensive study of human filariasis in Colombia. Three hundred and forty-seven individuals from various parts of Vaupés were examined. The overall infection rate for M. ozzardi was 49%, but the distribution of the parasite was not uniform throughout the Comisaría. Microfilariae were found in both venous blood samples and ski biopsies of infected individuals. The rate of infection was 55% for males and 42% for females, and infection ratenerally low, with 88% of the positive films …

Mansonellosis is endemic in several regions of Africa, the Caribbean, and Latin America. Mansonella ozzardi and Mansonella perstans have been reported in Latin America, including the Amazon region. A morphological and molecular microfilariae study was performed in Pauini (Brazil). Blood samples were collected from 40 individuals, and were analyzed by Giemsa-stained blood film and by two different nested polymerase chain reactions which detect internal transcribed spacer-1 and the major sperm protein gene. By microscopy, 14 of 40 were positive: 11 as M. ozzardi and three as M. perstans-like inf…

The long-term effect of a single oral dose of 150 microg/kg of body weight of ivermectin on Mansonella streptocerca microfilariae was studied in western Uganda. Before treatment, the geometric mean microfilaria density (mf) in 93 infected persons was 2.4 mf/mg of skin (range, 0.1-42.6). One year after treatment, 43 persons (46%) were microfilaria-negative, and the geometric mean in the remaining persons dropped significantly, to 0.7 mf/mg (range, 0.1-6.9). Thus, ivermectin is highly effective against M. streptocerca, and a single dose leads to a sustained suppression of microfilariae in skin. …

A case of co-infection with Plasmodium vivax and Mansonella ozzardi was detected in a blood sample from a person who had shown symptoms of malaria and lived in a city that was close to the Argentina/Bolivia border. The case was detected during a random revision of thick and thin smears from patients diagnosed with malaria from various towns and cities located in north-western Argentina between 1983 and 2001. Trophozoites of P. vivax were observed in the thin blood smear along with M. ozzardi microfilaria (larval form), which presented a long, slender, pointed anucleate tail and the absence of …

The discovery of obligatory intracellular bacteria of the genus Wolbachia in filariae infecting humans led to the use of antibiotics as a potent treatment option. Mansonella perstans is the cause of the second most prevalent filariasis in Gabon, but so far reports on the presence of Wolbachia in this nematode have been inconsistent. We report on the presence of Wolbachia in M. perstans in patients from Gabon, which we identified using polymerase chain reaction (PCR) with primer sets specific for 16S rDNA and ftsZ. Sequence analysis revealed a single consensus sequence, which could be phylogene…

Mansonella ozzardi is a tissue-dwelling parasitic nematode, the causative agent of mansonelliasis in almost all Latin American countries. It has been described along the Argentine Yungas region. The microscopic diagnosis can yield false-negative test results at low microfilaremia levels. The aim of this study was to optimize the molecular diagnostic technique and compare it with the Knott's method and standard blood smear procedures (thin blood films and thick smears) in 92 blood samples of individuals from an endemic area. The PCR technique followed by the sequencing of the amplified product …

INTRODUCTION: Mansonella ozzardi, a human filarial nematode transmitted by Diptera Simuliidae flies (black flies), is widely spread in the Amazon, while the occurrence of mansonelliasis in the State of Rondônia (Western Amazonia) has not been reported thus far. The existence of this parasitic disease in this state is investigated in this study.
METHODS: A total of 4,452 people who lived alongside the Madeira, Mamore, Guapore, Machado and Preto Rivers, in the State of Rondônia, were examined using the thick blood smear method. Alongside the Machado and Guapore rivers, 2,112 adult simuliid spec…

A search for filariae was performed on a wild male Gorilla g. gorilla from the Lopé Reserve in Gabon, which had died as a result of injuries inflicted by another male gorilla. A female worm of Loa loa and female worms of two species of Mansonella were recovered from the deep tissues of a wounded thigh. In order to analyze these Mansonella, specimens of M. (E.) perstans, M. (E.) vanhoofi and M. (E.) streptocerca from the Collections of the Museum National d'Histoire Naturelle de Paris have been studied and new important discriminative characters characterised, such as the body-swellings in M. Microfilariae were found in blood samples of a 32 year-old-student from Guinea-Bissau (Africa). Based on parasite morphology as well as some other features the microfilariae were identified as Mansonella perstans. The course of infection was asymptomatic (except for slight urticarial skin lesions). The microfilariae disappeared from peripheral blood after mebendazole treatment (200 mg daily for 1 month).

Patients harbouring Loa loa and/or Mansonella perstans infections, or presenting typical filarial symptomatology, were treated with mebendazole, a drug shown to be efficient in other parasitoses. Of the patients who took the drug during 21 days and provided regular blood samples, 84% showed a significant decrease in their L. loa microfilaraemia and 100% in their M. perstans microfilaraemia, with a concomitant improvement in the clinical manifestations and no side reaction to drug administration. Additionally, the long-lasting effect of the drug (up to day 200 in some cases) may suggest a posit…

Experimental infection with Mansonella ozzardi of common haematophagous Diptera collected at a Ticuna Indian village on the upper reaches of the Solimões river in the Brazilian Amazon, showed that Simulium amazonicum and Simulium n.sp. are capable of supporting full development of the parasite. Natural infections with this filaria were found in both species including infective larvae in Simulium n.sp. No development of M. ozzardi occurred in Mansonia amazonensis, Culicoides insinuatus or Lepiselaga crassipes (Tabanidae). The dimensions of developing larvae of M. ozzardi in both species of blac…
