Papua New Guinean schoolchildren in the highlands were randomly assigned to treatment groups in order to verify the effectiveness of mebendazole (Nordia) and compare it with mebendazole (Janssen) in both extended and single-dose therapy in a double-blind controlled study. Only the Janssen product given twice daily for three days was of value in 'curing' hookworm. Single-dose treatment with the same product was highly effective in treating roundworm but not hookworm or whipworm. Chemotherapy was administered to 53 patients with hydatid disease; to 27 of them it was administered to prevent recurrences. Mebendazole was prescribed by courses lasting for 14 to 30 days with intervals between the courses of 1 to 3 months, in a daily dose of 50 mg/kg to be taken 4 times. The plasma levels of mebendazole in children with hydatid cyst disease were measured with high pressure liquid chromatography. In 24 children who received mebendazole orally in a dose of 50 mg/kg, the mean (SD) level 4 hours later was 25.76(9.81) ng/ml (87.6(33) nanomole/l). This result was similar to those in most adult series. During more prolonged treatment, the plasma level 4 hours after the dose of the drug rose significantly with respect to the initial level (p < 0.05).

From 1984-1987, mass application of mebendazole-medicated salt was studied for the control of hookworm infection in 5 pilot areas (Wuming, Shanlin, Bobai, Guilin and Rongxian Counties) in Guangxi Zhuang Autonomous Region. The dosages of mebendazole (mixed with salt) administered were 15mg, 25mg, 30mg, 40mg, 50mg, 30mg, 70mg, 80mg, 100mg or 200mg per person per day for 15, 20 or 30 days. The use of mebendazole-treated diet (60 ppm) effectively controlled Hymenolepis nana and Aspiculuris tetraptera in a large mouse breeding colony. In a 3 generation pilot study using a medicated diet, there was some reduction in litter size and in female growth rate and an overall 2.07% incidence of kinky tails in the offspring. When the whole mouse colony was fed mebendazole-treated diet, a high incidence of kinky tails (maximum 46% of weaned offspring) occurred.

We report a case of invasive human Fasciola hepatica infection in which, for the first time, a successful treatment with high doses of mebendazole (4 g/day for 3 weeks) was achieved. This therapy resulted in a prompt and prolonged normalization of eosinophil counts, liver enzymes abnormalities and specific serological tests together with the disappearance of liver necrosis indicating the eradication of liver flukes. The efficacy of mebendazole in the treatment of fascioliasis must be confirmed by further studies.

The therapeutic efficacy of ketoconazole and mebendazole was studied in ICR/CD-1 mice infected with Trichinella spiralis for 17 to 20 weeks. Efficacy of both drugs was over 70% when compared with results in control mice. This study indicates that both ketoconazole and mebendazole should be considered in the treatment of trichinosis in humans.

Mebendazole was given prophylactically to a patient with a ruptured pulmonary hydatid cyst before surgical excision of the cyst. The cyst size was reduced by half and there was no radiological or immunological evidence of the presence of active secondary disease 15 months after rupture. The primary cyst was invaded by fungal hyphae, probably Aspergillus, and the danger of abscess or mycetoma formation if surgery is not carried out after chemotherapy is emphasized.

Oral administration of either Mebendazole or Fenbendazole at 50 mg/kg body weight was effective in killing a 7-hr Trichinella spiralis infection in mice. Adult worms, 72 hr postingection, were unaffected by the same dose of Fenbendazole, while Mebendazole showed partial activity. Both Mebendazole and Fenbendazole were active against developing muscle larvae. Mebendazole was effective against encysted muscle larvae while 50 mg of Fenbendazole per kg per day for 7 days from 28 days postinfection failed to have any effect on this stage of the life-cycle.

In a single-blind clinical study of mebendazole, a new, broadspectrum anthelmintic, 70 patients with light to severe infections of Trichuris trichiura were treated. Ten subjects received placebos, and 60 received 100 mg mebendazole twice a day for three and four days. The drug was well tolerated by the patients, and no toxic side effects were observed.
