Mercury isotope dynamics in marine fish during bioaccumulation and excretion: Temporal responses across tissues.

Mercury (Hg) is a potent neurotoxin that bioaccumulates through aquatic food webs. In fish, Hg excretion regulates Hg burden, yet whether it induces tissue-specific Hg isotope fractionation remains unclear. To address this question, we conducted a controlled feeding experiment exposing starry flounders (Platichthys stellatus) to diets amended with inorganic mercury (IHg; 4 and 25 μg/g) or methylmercury (MeHg; 1 μg/g) for 4 weeks (bioaccumulation), followed by 16 weeks of Hg-unamended feeding (excretion). Total Hg and MeHg concentrations, together with Hg isotope ratios (δ202Hg, Δ199Hg) were me

Evidence of Mercury Methylation and Demethylation by the Estuarine Microbial Communities Obtained in Stable Hg Isotope Studies.

Microbial activity is a critical factor controlling methylmercury formation in aquatic environments. Microbial communities were isolated from sediments of two highly mercury-polluted areas of the Tagus Estuary (Barreiro and Cala do Norte) and differentiated according to their dependence on oxygen into three groups: aerobic, anaerobic, and sulphate-reducing microbial communities. Their potential to methylate mercury and demethylate methylmercury was evaluated through incubation with isotope-enriched Hg species (199HgCl and CH₃201HgCl). The results showed that the isolated microbial communities

Tracing maternal transfer of methylmercury in the sheepshead minnow (Cyprinodon variegatus) with an enriched mercury stable isotope.

Maternal transfer of methylmercury (MeHg) to eggs is an important exposure pathway for developing offspring. However, our understanding of this process is limited, particularly in estuarine fish. We conducted a 91-day chronic laboratory exposure of Cyprinodon variegatus to four concentrations of dietary MeHg ranging from 0.04 to 9.90 μg g(-1) dw. Juvenile fish were fed a preoogenesis MeHg diet for 28 days, after which the diet was switched to a diet enriched with Me(199)Hg, spanning the period of oogenesis, allowing us to differentiate between mercury stored in female tissues and mercury assim

Isotope fractionation of mercury during its photochemical reduction by low-molecular-weight organic compounds.

Photochemical reduction of Hg(II) by various low-molecular-weight organic compounds (LMWOC) was investigated to evaluate the effect of specific functional groups that are typically encountered in natural dissolved organic matters (DOM) on the photoreactivity and isotope fractionation of Hg. LMWOC with reduced sulfur functional groups (e.g., cysteine, glutathione) resulted in slower photochemical reduction of Hg(II) than those without reduced sulfur groups (e.g., serine, oxalic acid). Reduction rate constants were specifically determined for two contrasting LMWOC: dl-serine (0.640 h(-1)) and l-

Equilibrium mercury isotope fractionation between dissolved Hg(II) species and thiol-bound Hg.

Stable Hg isotope ratios provide a new tool to trace environmental Hg cycling. Thiols (-SH) are the dominant Hg-binding groups in natural organic matter. Here, we report experimental and computational results on equilibrium Hg isotope fractionation between dissolved Hg(II) species and thiol-bound Hg. Hg(II) chloride and nitrate solutions were equilibrated in parallel batches with varying amounts of thiol resin resulting in different fractions of thiol-bound and free Hg. Mercury isotope ratios in both fractions were analyzed by multiple collector inductively coupled plasma mass spectrometry (MC

Unveiling the Sources and Transfer of Mercury in Forest Bird Food Chains Using Techniques of Vivo-Nest Video Recording and Stable Isotopes.

Knowledge gaps in mercury (Hg) biomagnification in forest birds, especially in the most species-rich tropical and subtropical forests, limit our understanding of the ecological risks of Hg deposition to forest birds. This study aimed to quantify Hg bioaccumulation and transfer in the food chains of forest birds in a subtropical montane forest using a bird diet recorded by video and stable Hg isotope signals of biological and environmental samples. Results show that inorganic mercury (IHg) does not biomagnify along food chains, whereas methylmercury (MeHg) has trophic magnification factors of 7

Evaluating total mercury and methylmercury biomagnification using stable isotopes of carbon and nitrogen in fish from the Madeira River basin, Brazilian Amazon.

The isotopic signatures of carbon in primary consumer fish species in the Madeira River basin (Western Amazon) follow a pattern, with basal species feeding mainly on C3 (e.g., vascular plants and phytoplankton). In this study with 196 specimens of 19 fish species and six trophic guilds, significant differences were found between the two main groups of primary consumers (herbivores and detritivores) in the Madeira River basin. The mercury and δ15N data indicated that the fish specimens collected in Puruzinho and Cuniã Lakes provided greater trophic magnification factors (TMFs) for this metal th

Development of a quadruple isotope spiking methodology to study mercury methylation and methylmercury demethylation in environmental and biological media.

A quadruple isotope spiking methodology has been developed and applied for the study of Hg-species-specific transformations in incubation experiments. The method enables for the first time discrimination between interconversions reactions occurring during the incubation step from those occurring during the analytical procedure. Two different mathematical models for quadruple spiking were developed and applied here in model incubation experiments to track inorganic Hg(II) methylation and methylmercury demethylation. The results showed identical Hg compounds concentrations and interconversion re

New insights into the biomineralization of mercury selenide nanoparticles through stable isotope analysis in giant petrel tissues.

Tiemannite (HgSe) is considered the end-product of methylmercury (MeHg) demethylation in vertebrates. The biomineralization of HgSe nanoparticles (NPs) is understood to be an efficient MeHg detoxification mechanism; however, the process has not yet been fully elucidated. In order to contribute to the understanding of complex Hg metabolism and HgSe NPs formation, the Hg isotopic signatures of 40 samples of 11 giant petrels were measured. This seabird species is one of the largest avian scavengers in the Southern Ocean, highly exposed to MeHg through their diet, reaching Hg concentrations in the

Mercury Stable Isotope Fractionation during Abiotic Dark Oxidation in the Presence of Thiols and Natural Organic Matter.

Mercury (Hg) stable isotope fractionation has been widely used to trace Hg sources and transformations in the environment, although many important fractionation processes remain unknown. Here, we describe Hg isotope fractionation during the abiotic dark oxidation of dissolved elemental Hg(0) in the presence of thiol compounds and natural humic acid. We observe equilibrium mass-dependent fractionation (MDF) with enrichment of heavier isotopes in the oxidized Hg(II) and a small negative mass-independent fractionation (MIF) owing to nuclear volume effects. The measured enrichment factors for MDF

Investigating uptake and translocation of mercury species by sawgrass ( Cladium jamaicense ) using a stable isotope tracer technique.

The role of macrophytes in mercury (Hg) cycling in the Florida Everglades ecosystem has not been fully understood. In this study, a stable isotope ((199)Hg(2+)) addition technique was used to trace the methylation, uptake, and translocation of Hg by sawgrass ( Cladium jamaicense ) and quantitatively evaluate the contribution of atmospheric and soil Hg to Hg in sawgrass leaves and below-ground biomass. The results showed that spiked (199)Hg(2+) could be rapidly methylated to monomethylmercury (Me(199)Hg) in the soil of the sawgrass pots. Only small portions of total Hg (THg) and monomethylmercu

Biomolecular and isotopic evidence reveals key cellular controls on methylmercury fate during bacterial mercury methylation.

Microbial mercury (Hg) methylation drives the formation of methylmercury (MeHg) hotspots in natural environments, but the cellular pathways that determine MeHg fate and isotopic signatures remain largely unresolved. In this study, we cultured the model sulfate-reducing bacterium Pseudodesulfovibrio hydrargyri BerOc1 anaerobically under fumarate respiration for 30 h and quantified Hg speciation and fraction- and species-specific Hg isotopic compositions. The results showed that intracellular MeHg was rapidly exported and dominated the extracellular pool by 30 h (> 80%). Exported MeHg bound to s

Mass spectrometry measurements of mercury isotope ratios support geochemical sourcing of archaeological cinnabar in the Andean region.

Geochemical methods can identify the long-distance exchange of resources in the archaeological record. Cinnabar is a mineral with a limited number of geological sources; however, methods for determining the geological origin of cinnabar are constricted by the limited availability of comparative geological source materials. This study applies a multi-method approach to compare isotopic ratios of mercury and sulfur in archaeological specimens of cinnabar from museum collections and scientifically excavated materials from the Andes region of South America. We demonstrate that the δ202Hg to Δ199Hg

Tracing anthropogenic mercury in soils from Fe-Hg mining/smelting area: Isotopic and speciation insights.

Mercury (Hg) stable isotope ratios supplemented by Hg solid speciation data were determined in soils in a former Fe-Hg mining/smelting area (Jedová hora, Czech Republic, Central Europe). The dominant Hg phase in the studied soils was found to be cinnabar (HgS). A secondary form of soil Hg(II) was represented by Hg weakly and strongly bound to mineral (micro)particles, as revealed by thermo-desorption analysis. These Hg species probably play a key role in local soil Hg processes and biogeochemical cycling. The Hg isotopic data generally showed small differences between HgS (-1.1 to -0.8‰; δ202H

Increased Contribution of Circumpolar Deep Water Upwelling to Methylmercury in the Upper Ocean around Antarctica: Evidence from Mercury Isotopes in the Ornithogenic Sediments.

Upwelling plays a pivotal role in supplying methylmercury (MeHg) to the upper oceans, contributing to the bioaccumulation of MeHg in the marine food web. However, the influence of the upwelling of Circumpolar Deep Water (CDW), the most voluminous water mass in the Southern Ocean, on the MeHg cycle in the surrounding oceans and marine biota of Antarctica remains unclear. Here, we study the mercury (Hg) isotopes in an ornithogenic sedimentary profile strongly influenced by penguin activity on Ross Island, Antarctica. Results indicate that penguin guano is the primary source of Hg in the sediment

Reference measurements for total mercury and methyl mercury content in marine biota samples using direct or species-specific isotope dilution inductively coupled plasma mass spectrometry.

The analytical procedures for reference measurements of the total Hg and methyl mercury (MeHg) mass fractions at various concentration levels in marine biota samples, candidates for certified reference materials (oyster and clam Gafrarium tumidum), were evaluated. Two modes of application of isotope dilution inductively coupled plasma mass spectrometry method (ID ICP-MS), namely direct isotope dilution and species-specific isotope dilution analysis with the use of two different quantification mass spectrometry techniques were compared. The entire ID ICP-MS measurement procedure was described b

Dissociation of Mercuric Oxides Drives Anomalous Isotope Fractionation during Net Photo-oxidation of Mercury Vapor in Air.

The atmosphere is the primary medium for long-distance transport and transformation of elemental mercury (Hg), a potent neurotoxin. The recent discovery of mass-independent fractionation (MIF) of even-mass Hg isotopes (even-MIF, measured as Δ200Hg and Δ204Hg) in the atmosphere is surprising and can potentially serve as a powerful tracer in understanding Hg biogeochemistry. Far-ultraviolet (UVC) light-induced gas-phase reactions have been suspected as a likely cause for even-MIF, yet the mechanism remains unknown. Here, we present the first experimental evidence of large-scale even-MIF caused b

Species-specific isotope tracers to study the accumulation and biotransformation of mixtures of inorganic and methyl mercury by the microalga Chlamydomonas reinhardtii.

The present study demonstrates that species-specific isotope tracing is an useful tool to precisely measure Hg accumulation and transformations capabilities of living organisms at concentrations naturally encountered in the environment. To that end, a phytoplanktonic green alga Chlamydomonas reinhardtii Dangeard (Chlamydomonadales, Chlorophyceae) was exposed to mixtures of (199)-isotopically enriched inorganic mercury ((199)IHg) and of (201)-isotopically enriched monomethylmercury ((201)CH3Hg) at a concentration range between less than 1 pM to 4 nM.
