In vivo expression of ephrinA5-Fc in mice results in cephalic neural crest agenesis and craniofacial abnormalities.

Eph receptors and their ligands ephrins have been implicated in guiding the directed migration of neural crest cells (NCCs). In this study, we found that Wnt1-Cre-mediated expression of ephrinA5-Fc along the dorsal midline of the dien- and mesencephalon resulted in severe craniofacial malformation of mouse embryo. Interestingly, expression of cephalic NCC markers decreased significantly in the frontonasal process and branchial arches 1 and 2, which are target areas for the migratory cephalic NCCs originating in the dien- and mesencephalon. In addition, these craniofacial tissues were much smal

Distribution of substance P-like immunoreactivity in the brain of the newt (Triturus cristatus).

The distribution of immunoreactive substance P (sP)-containing structures in the newt brain and spinal cord was explored with an indirect immunofluorescence method. Five sP-positive elements were detected: perikarya, dots, fibers, pericellular appositions, and pipe-shaped structures. Perikarya were seen at the levels of the spinal ganglia, spinal cord, raphe nucleus, interpeduncular nucleus, mesencephalon, preoptic area, infundibulum, dorsocaudal part of the ventral hypothalamus, habenula, and corpus striatum. Pericellular terminals were observed in periventricular areas, known to be rich in c

Artery of Percheron infarction presented with isolated downgaze paralysis: A case report.

Isolated downgaze paralysis is the most infrequent expression of vertical gaze abnormalities. Vertical eye movements are controlled by nuclei and circuits located in the thalamic-mesencephalon region, and more particularly the rostral interstitial nucleus of the medial longitudinal fasciculus (riMLF). The Artery of Percheron (AP) is a rare vascular anatomic variation that supplies the paramedian region of the thalami and the rostral portion of the mesencephalon. We present a unique case of isolated downgaze paralysis caused by AP ischemia.

Cardioembolic Artery of Percheron Infarction.

Occlusion of the artery of Percheron (AOP) is a rare yet potentially disabling form of ischemic stroke resulting in infarction of the bilateral paramedian thalami and mesencephalon with variable and often atypical presentation. Given the various patterns of thalamic blood supply, recognizing the presence of AOP infarction is crucial for the diagnosis and management of ischemic strokes involving these regions. Here, we report a case of acute hemorrhagic infarction involving the bilateral thalami and the rostral mesencephalon caused by a cardioembolic occlusion of the AOP.

Local and transient expression of E-cadherin involved in mouse embryonic brain morphogenesis.

We found that E-cadherin (uvomorulin) is transiently expressed in restricted regions of the metencephalon, mesencephalon and diencephalon of mouse embryonic brain. This expression first occurred in parts of the mesencephalon and diencephalon at around E9.5, and subsequently extended to the primordia of cerebellum, the dorsal midline of mesencephalon and some other regions of the embryonic brain. These E-cadherin expressions ceased by E15 except at the dorsal midline. Immunohistological analyses showed that E-cadherin-positive cells are radially arranged in the neural tube and the E-cadherin-po

The avian tectobulbar tract: development, explant culture, and effects of antibodies on the pattern of neurite outgrowth.

The tectobulbar tract is the first long-distance projecting fiber pathway to appear during the development of the avian optic tectum (dorsal half of the mesencephalon). Immunologically stained wholemounts of the E3 mesencephalon reveal that the early tectobulbar axons course in a dorsal-to-ventral direction and abruptly turn in a caudal direction shortly before reaching the ventral midline. During subsequent development, more tectobulbar axons are generated that form a parallel array of thick fascicles coursing ventrally within the mesencephalon. At this later stage of development, the tectobu

Hereditary diabetes insipidus in rats. Altered cerebral indolamine and catecholamine metabolism.

Compared to heterozygous Brattleboro animals, homozygous (diabetes insipidus) rats exhibited higher steady-state levels of serotonin in the mesencephalon, septum and striatum. These differences disappeared upon the administration of pargyline, suggesting accumulation of serotonin. The norepinephrine level was higher in the mesencephalon, while the disappearance rate (alpha-met hyl-p-tyrosine) was accelerated in the septum and decreased in the hypothalamus. The lower striatal dopamine level was associated with a decreased disappearance rate. The data suggest that the altered monoamine metabolis

Stimulation sites in periaqueductal gray, nucleus raphe magnus and adjacent regions effective in suppressing oral-facial reflexes.

Electrode penetrations were made in the mesencephalon and caudal brainstem at the levels of the periaqueductal gray matter (PAG) and nucleus raphe magnus (NRM) in chloralose-anaesthetized or decerebrate cats. In a systematic fashion, mesencephalic and brainstem loci at every 1 mm of vertical depth were electrically stimulated in a series of mediolateral or anteroposterior electrode penetrations to determine the lowest stimulation threshold at each locus for suppressing the digastric jaw-opening reflex evoked by tooth pulp or infraorbital nerve stimulation; at some loci, the threshold current r

Is MRPI 2.0 More Useful than MRPI and M/P Ratio in Differential Diagnosis of PSP-P with Other Atypical Parkinsonisms?

Differential diagnosis of progressive supranuclear palsy remains difficult, especially when it comes to the parkinsonism predominant type (PSP-P), which has a more favorable clinical course. In this entity, especially during the advanced stages, significant clinical overlaps with other tauopathic parkinsonian syndromes and multiple system atrophy (MSA) can be observed. Among the available additional diagnostic methods in every-day use, magnetic resonance imaging (MRI) focused specifically on the evaluation of the mesencephalon seems to be crucial as it is described as a parameter associated wi

Rhodanese in mouse brain: regional differences and their metabolic implications.

Our results concern the regional localization of rhodanese in the mouse brain. A histoenzymatic examination was undertaken in telencephalon, diencephalon, mesencephalon, and rhombencephalon. The sites of rhodanese activity are revealed as punctuate, granular, dark dots, small in some regions such as hippocampus or bigger in others, and as long, thread-like particles especially, abundant in the region of the telencephalon in the astroglia cells and in the region of the mesencephalon in the hippocampus. There were sites with a high density of the histochemical test products, for example, the epe

Dopaminergic neurons modulate GABA neuron migration in the embryonic midbrain.

Neuronal migration, a key event during brain development, remains largely unexplored in the mesencephalon, where dopaminergic (DA) and GABA neurons constitute two major neuronal populations. Here we study the migrational trajectories of DA and GABA neurons and show that they occupy ventral mesencephalic territory in a temporally and spatially specific manner. Our results from the Pitx3-deficient aphakia mouse suggest that pre-existing DA neurons modulate GABA neuronal migration to their final destination, providing novel insights and fresh perspectives concerning neuronal migration and connect

Severe second-trimester obstructive ventriculomegaly related to disorders of diencephalic, mesencephalic and rhombencephalic differentiation.

By review of a series of cases, we set out to identify sonographic features suggestive of an obstructive mechanism in second-trimester fetuses with ventriculomegaly and describe developmental disorders related to pathological differentiation of the diencephalon, mesencephalon and rhombencephalon that lead to obstruction of cerebrospinal fluid flow. We studied retrospectively 11 fetuses referred for severe second-trimester ventriculomegaly of undetermined origin. Neurosonography was performed with detailed analysis of the third ventricle, thalami, cerebral aqueduct and cerebellum. The cerebral

Effect of a unilateral cortical lesion on the monoamine content in the cat brain.

The serotonin and noradrenalin content in the cerebral cortex, hypothalamus, and mesencephalon was investigated spectrofluorometrically in 12 cats on the 5th-6th day after the production of a pathological focus in the region of the occipital cortex. Diffuse changes of brain activity were recorded on the EEG at this period: spike-like waves and slow waves of increased amplitude. A considerable increase in the serotonin concentration was observed in the cortex, with the effect predominant in the region immediately adjacent to the pathological focus. A tendency for the serotonin level to fall was

Deep Learning-Based Assessment of Brainstem Volume Changes in Spinocerebellar Ataxia Type 2 (SCA2): A Study on Patients and Preclinical Subjects.

Spinocerebellar ataxia type 2 (SCA2) is a neurodegenerative disorder marked by progressive brainstem and cerebellar atrophy, leading to gait ataxia. Quantifying this atrophy in magnetic resonance imaging (MRI) is critical for tracking disease progression in both symptomatic patients and preclinical subjects. However, manual segmentation of brainstem subregions (mesencephalon, pons, and medulla) is time-consuming and prone to human error. This work presents an automated deep learning framework to assess brainstem atrophy in SCA2. Using T1-weighted MRI scans from patients, preclinical carriers,

MRI topography of lesions related to internuclear ophthalmoplegia in patients with multiple sclerosis or ischemic stroke.

BACKGROUND AND PURPOSE: Internuclear ophthalmoplegia is a dysfunction of conjugate eye movements, caused by lesions affecting the medial longitudinal fasciculus (MLF). Multiple sclerosis (MS) and ischemic stroke represent the most common pathophysiologies. While magnetic resonance imaging (MRI) allows for localizing lesions affecting the MLF, comprehensive comparative studies exploring potential different spatial characteristics of lesions affecting the MLF are missing until now. METHODS: We retrospectively investigated MRI examinations of 82 patients (40 patients with MS and 42 patients with

Improvement of cerebral blood perfusion in certain cerebral regions after cranioplasty could be monitored via tympanic membrane temperature changes.

OBJECTIVE: Delayed neurological deficit was often observed in patients underwent craniectomy, which could be improved by cranioplasty. Little is known about hemodynamic improvement before and after cranioplasty. METHODS: Cerebral blood perfusion, tympanic membrane temperature (TMT), neuropsychological and cognitive function were assessed in eleven craniectomy patients before and after cranioplasty. RESULTS: Before cranioplasty, the cerebral blood volume (CBV) on the decompressed side was significantly lower than that of the contralateral side. The cranioplasty led to instant improvement (7 d

Otx2 homeobox gene induces photoreceptor-specific phenotypes in cells derived from adult iris and ciliary tissue.

PURPOSE: It remains unclear which gene induction effectively generates photoreceptor-specific phenotypes from nonretinal tissues. The purpose of this study was to determine whether Crx and Otx2--homeobox genes related to photoreceptor development--can induce the generation of these phenotypes in cells derived from adult ciliary and iris tissue and in mesencephalon-derived neural stem cells. METHODS: Crx and Otx2 were transferred into adult rat ciliary- and embryonic mesencephalon-derived neurospheres and adult rat iris-derived cells with the aid of a recombinant retrovirus.
