Mitochondrial loss, dysfunction and altered dynamics in Huntington's disease.

Although a direct causative pathway from the gene mutation to the selective neostriatal neurodegeneration remains unclear in Huntington's di … rial biogenesis, were also significantly reduced with increasing disease severity. Abnormalities in mitochondrial dynamics were observed, showing a significant increase in the fission protein Drp1 and a reduction in the expression of the fusion protein mitofusin 1. Lastly, mitochondrial PCR array profiling in HD caudate nucleus specimens showed increased mRNA expression …

Mitochondrial dynamics: A promising tool for personalized TNBC management.

Triple-negative breast cancer (TNBC) is an aggressive and highly metastatic form of breast cancer and is associated with poor prognosis due to the lack of targeted therapies. Mitochondrial dysfunction is a crucial factor contributing to tumor growth and chemoresistance in TNBC. Dysregulation in mitochondrial dynamics leads to disruption of the normal process of oxidative phosphorylation, elevated reactive oxygen species generation, and resistance to apoptosis, contributing to TNBC aggressiveness. Moreover, alterations in mitochondrial energetics, including elevated glycolysis and glutamine add…

Systems modelling of mitochondrial dynamics in different exercise regimes.

Exercise stimulates skeletal muscle signalling and mitochondrial metabolism. Emerging evidence shows that mitochondrial dynamics (i.e. fission and fusion) could be regulated by exercise. Yet, key gaps remain in identifying (i) the signals that drive fission vs. fusion; (ii) how energy status and reactive oxygen species (ROS) shift control between dynamin-related protein 1 (DRP1) and mitofusin (MFN)/optic atrophy 1 (OPA1); and (iii) which intensity-duration combinations yield similar cytosolic signals but different mitochondrial remodelling. Therefore, we developed an integrative computational …

UCP2 mediates mitochondrial dynamics to induce AgRP neuronal activity.

OBJECTIVES: The hypothalamic agouti-related protein (AgRP)- expressing neurons regulate feeding and whole-body energy homeostasis. A growing body of evidence indicates that changes in mitochondrial dynamics, such as fission and fusion, play a crucial role in regulating AgRP neuronal activity. However, the mechanisms underlying this process remain to be elucidated. Here, we showed a role of mitochondrial UCP2-mediated mitochondrial dynamics in AgRP neurons in regulating AgRP neuronal activity and fasting-induced feeding behavior. METHODS: We analyzed mitochondrial morphology, expression of act…

HIF-1 signaling: an emerging mechanism for mitochondrial dynamics.

A growing emphasis has been paid to the function of mitochondria in tumors, neurodegenerative disorders (NDs), and cardiovascular diseases. Mitochondria are oxygen-sensitive organelles whose function depends on their structural basis. Mitochondrial dynamics are critical in regulating the structure. Mitochondrial dynamics include fission, fusion, motility, cristae remodeling, and mitophagy. These processes could alter mitochondrial morphology, number, as well as distribution, to regulate complicated cellular signaling processes like metabolism. Meanwhile, they also could modulate cell prolifera…

Caspase-Cleaved Tau Impairs Mitochondrial Dynamics in Alzheimer's Disease.

Alzheimer's disease (AD) is characterized by the presence of aggregates of tau protein. Tau truncated by caspase-3 (D421) or tau hyperphosphorylated at Ser396/S404 might play a role in the pathogenesis of AD. Mitochondria are dynamic organelles that modify their size and function through mitochondrial dynamics. Recent studies have shown that alterations of mitochondrial dynamics affect synaptic communication. Therefore, we studied the effects of pathological forms of tau on the regulation of mitochondrial dynamics. We used primary cortical neurons from tau(-/-) knockout mice and immortalized c…

Light-evoked activity and BDNF regulate mitochondrial dynamics and mitochondrial localized translation in CNS axons.

Mitochondria coordinate well-described maintenance functions within neuronal axons and dendrites. However, less is known about how mitochond … calized and translated at mitochondria within RGC axons in vivo, concomitant with the regulation of mitochondrial dynamics. These data highlight the previously undescribed regulation of mitochondrial dynamics in axonal maturation, dependent on mechanisms involving neuronal activity and neurotrophic factor signaling, coordinated with mitochondrial-localized translation.

Mitochondrial SIRT3 confers neuroprotection in Huntington's disease by regulation of oxidative challenges and mitochondrial dynamics.

SIRT3 is a major regulator of mitochondrial acetylome. Here we show that SIRT3 is neuroprotective in Huntington's disease (HD), a motor neurodegenerative disorder caused by an abnormal expansion of polyglutamines in the huntingtin protein (HTT). Protein and enzymatic analysis revealed that increased SIRT3 is a signature in several HD models, including human HD brain, which is regulated by oxidative species. While loss of SIRT3 further aggravated the oxidative phenotype, antioxidant treatment regularized SIRT3 levels. SIRT3 overexpression promoted the antioxidant effect in cells expressing muta

Bioenergetic flux, mitochondrial mass and mitochondrial morphology dynamics in AD and MCI cybrid cell lines.

Bioenergetic dysfunction occurs in Alzheimer's disease (AD) and mild cognitive impairment (MCI), a clinical syndrome that frequently precedes symptomatic AD. In this study, we modeled AD and MCI bioenergetic dysfunction by transferring mitochondria from MCI, AD and control subject platelets to mtDNA-depleted SH-SY5Y cells. Bioenergetic fluxes and bioenergetics-related infrastructures were characterized in the resulting cytoplasmic hybrid (cybrid) cell lines. Relative to control cybrids, AD and MCI cybrids showed changes in oxygen consumption, respiratory coupling and glucose utilization. AD an
