Microarray analysis of mitogenic effects of T3 on the rat liver.

BACKGROUND AND AIM: A single dose of the thyroid hormone tri-iodothyronine, T3, can enhance both size and function of normal rodent liver, w … esis. CONCLUSIONS: We compared the results with published results of the response to other primary mitogens, and identified the Gadd45beta/MyD118 gene as a common early factor upregulated during proliferation.

Hepatoma derived growth factor is a nuclear targeted mitogen.

Hepatoma Derived Growth Factor (HDGF) was originally identified as a secreted mitogen from the human hepatoma cell line Huh-7. Although initially thought to be a cytoplasmic protein, it became clear that HDGF is a nuclear targeted protein containing a canonical bipartite nuclear localization sequence. In the developing vasculature HDGF is highly expressed in the nucleus of smooth muscle and endothelial cells. HDGF expression in smooth muscle in particular, declines after birth in larger arteries only to be re-expressed with vascular injury in cells of the highly proliferative neointima. In add

Lymphocyte subsets and mitogen stimulation of blood lymphocytes in preeclampsia.

PROBLEM: The question of whether there are differences in systemic immune reactivity in severe preeclampsia compared with normal pregnancy w … he function of peripheral blood mononuclear cells (PBMCs) was studied as proliferative responses to mitogens alone and in combination with immunomodulating drugs. RESULTS: An increased number of B lymphocytes (CD19+) (P < 0.05) and natural killer (NK) cells (P < 0.05) was noticed in severe preeclampsia compared with normal pregnancy. The proliferative res …

PHA for aplastic anemias: the alpha but not the omega of mitogen therapies.

This manuscript updates an analysis of the foremost clinical investigation of PHA for aplastic anemias and other disorders associated with i … dequacy of dosages applied, yet much better results were observed than might have been anticipated. Mitogens such as the L4 isolectin of PHA might be applied to treat aplastic anemias in three different ways. (1) For milder direct cytotoxicity types, a stimulating regimen should be applied to increase the production of growth factors and thereby hasten rec …

Mitogenic signal transduction in human breast cancer cells.

1. Signal transduction pathways activated during growth of human breast cancer cells in tissue culture are reviewed. 2. Steroid hormones and growth factors stimulate similar mitogenic pathways and frequently modulate each other's activity. 3. A response common to estrogen, progestins and most polypeptide mitogens is induction of the nuclear transcription factors myc, fos and jun in early G1 phase of the cell cycle. 4. Some growth factors also stimulate cyclin D1, a regulatory protein responsible for the activation of cell cycle-dependent kinases in G1. 5. In addition, insulin, IGF-I and EGF ac…

Suppression of T lymphocyte mitogen response in patients with mastocytosis.

Immunological functions were studied in 22 patients with mastocytosis. Lymphocyte stimulation with concanavalin A (Con A) and phytohemagglutinin showed that the patients responded with lower mitogenic activity than healthy controls. Furthermore, the lymphocytes of patients with the most extensive mast cell disease had a significantly lower Con A mitogen response than the lymphocytes of the rest of the patients. The effect of histamine and its specific metabolites, tele-methylhistamine and tele-methylimidazoleacetic acid (MeImAA), on the Con A lymphocyte mitogen response was also studied in hea

Suppression of mitogen-induced lymphocyte transformation in cigarette smokers.

Mitogen- and antigen-induced lymphocyte transformation was determined in peripheral blood lymphocytes from tobacco smokers and nonsmokers. Each group was evaluated by race, sex, and age subgroups. Male smokers as a group had depressed responses to phytohemagglutinin (PHA), concanavalin A, and pokeweed mitogen. The effect was most pronounced in all subjects on PHA stimulation. There was no apparent relationship to age or race. Antigenic stimulation of lymphocytes with mumps skin test antigen was not affected in any of the groups studied.

Local mitogenic effect of tissue mast cell secretion.

The effect of drug-induced mast cell secretion on proliferation was studied in fibroblast-like and mesothelial-like cells in organ-cultured rat mesentery. Mast cell degranulation achieved by Compound 48/80 was followed by a marked mitogenic reaciton in the surrounding tissue cells. The drug itself lacked mitogenic effect on cultured guinea-pig mesentery, the mast cells of which are unresponsive to the drug, and on a human normal fibroblast-like cell line. In contrast, histamine at about 10-(10) M, a major mast cell component, induced marked mitogenesis in guinea-pig mesentery without causing d

Release of prostaglandin by mitogen- and antigen-stimulated leukocytes in culture.

The prostaglandin (PG) content of mitogen- and antigen-stimulated leukocyte cultures was examined by a radioimmunoassay procedure empolying … cal enterotoxin B. Since iPG levels rose only in the culture supernates and not in cells exposed to mitogens for 48 h, increases reflected extracellular release of PG. The validity of the radioimmunoassay determinations of PGE in spleen cell cultures was supported by the results of concomitant assessment of the PGE(2) content of basal and enterotoxin-stimu …

Phosphothreonine Lyase Promotes p65 Degradation in a Mitogen-Activated Protein Kinase/Mitogen- and Stress-Activated Protein Kinase 1-Dependent Manner.

Bacterial phosphothreonine lyases have been identified to be type III secretion system (T3SS) effectors that irreversibly dephosphorylate host mitogen-activated protein kinase (MAPK) signaling to promote infection. However, the effects of phosphothreonine lyase on nuclear factor κB (NF-κB) signaling remain largely unknown. In this study, we detected significant phosphothreonine lyase-dependent p65 degradation during Edwardsiella piscicida infection in macrophages, and this degradative effect was blocked by the protease inhibitor MG132. Further analysis revealed that phosphothreonine lyase prom

Arterial territory-specific phosphorylated retinoblastoma protein species and CDK2 promote differences in the vascular smooth muscle cell response to mitogens.

Despite recent advances in medical procedures, cardiovascular disease remains a clinical challenge and the leading cause of mortality in the western world. The condition causes progressive smooth muscle cell (SMC) dedifferentiation, proliferation, and migration that contribute to vascular restenosis. The incidence of disease of the internal mammary artery (IMA), however, is much lower than in nearly all other arteries. The etiology of this IMA disease resistance is not well understood. Here, using paired primary IMA and coronary artery SMCs, serum stimulation, siRNA knockdowns, and verificatio

Modulation of host immune responses by Fasciola hepatica: responses by peripheral lymphocytes to mitogens during liver fluke infections of sheep.

Fasciola hepatica infections of lambs (250 or 500 metacercariae) were shown to alter the proliferative responses of peripheral blood lymphocytes (whole blood culture) to mitogens at specific times postinfection (PI). Responses to concanavalin A (Con A) were significantly suppressed at weeks 4, 8, 10, and 11 PI whereas suppressed responses to phytohemagglutinin (PHA) occurred at weeks 4, 10, 11, and 16 PI. Only on weeks 4 and 6 PI were responses to pokeweed mitogen (PWM) suppressed. The fluke-induced modulation of responses appeared to be related more to specific phases of infection rather than…

Studying Mitogen-Independent Proliferation in Murine B Cells.

The prevailing dogma of the cell cycle posits that a mitogenic signal is required at the end of each mitosis for cells to progress through the next G1. This textbook model is based mostly on experiments with fibroblast cultures. Burnet's clonal selection theory states that selected B and T cells expand rapidly to initiate immune response. To achieve this, it is plausible that lymphocytes do not follow the canonical rules of cell cycle progression. One such cell cycle anomaly lymphocytes demonstrate is non-dependency on mitogenic signals during proliferative phases. Such modified cell cycle reg

Roles of Mitogen-Activated Protein Kinases in Osteoclast Biology.

Bone undergoes continuous remodeling, which is homeostatically regulated by concerted communication between bone-forming osteoblasts and bone-degrading osteoclasts. Multinucleated giant osteoclasts are the only specialized cells that degrade or resorb the organic and inorganic bone components. They secrete proteases (e.g., cathepsin K) that degrade the organic collagenous matrix and establish localized acidosis at the bone-resorbing site through proton-pumping to facilitate the dissolution of inorganic mineral. Osteoporosis, the most common bone disease, is caused by excessive bone resorption,

Expression of GADS enhances FLT3-induced mitogenic signaling.

Solar System bodies have similar abundances of non-volatile elements. Asteroids are categorized as unmelted chondritic bodies or as differentiated bodies formed by extensive to global melting of chondritic progenitors. Reflectance spectra show asteroids are compositionally similar to meteorites (which are composed of non-exotic materials-mainly silicates, metallic Fe-Ni, sulfides, oxides, and organic matter). Dense refractory siderophile elements (e.g., Re, Os, Ir, Pt) are present in iron meteorites in total concentrations < 0.05 wt%. The upper limit on the density of an asteroid is ~ 8 g cm-3

Mitogen-Activated Protein Kinases and Hypoxic/Ischemic Nephropathy.

Tissue hypoxia/ischemia is a pathological feature of many human disorders including stroke, myocardial infarction, hypoxic/ischemic nephropathy, as well as cancer. In the kidney, the combination of limited oxygen supply to the tissues and high oxygen demand is considered the main reason for the susceptibility of the kidney to hypoxic/ischemic injury. In recent years, increasing evidence has indicated that a reduction in renal oxygen tension/blood supply plays an important role in acute kidney injury, chronic kidney disease, and renal tumorigenesis. However, the underlying signaling mechanisms,
