Implementation of action sequences by a neostriatal site: a lesion mapping study of grooming syntax.

The neostriatum and its connections control the sequential organization of action ("action syntax") as well as simpler aspects of movement. This study focused on sequential organization of rodent grooming. Grooming syntax provides an opportunity to study how neural systems coordinate natural patterns of serial order. The most stereotyped of these grooming patterns, a "syntactic chain," has a particularly stereotyped order that recurs thousands of times more often than could occur by chance. The purpose of the present study was to identify the crucial site within the striatopallidal system wher…

Neural transplantation in Huntington disease: long-term grafts in two patients.

OBJECTIVE: Clinical trials of fetal neural tissue transplantation for Huntington disease (HD) have been conducted with variable clinical results. However, no long-term analysis of graft survival and integration has been published. Here, we report the pathologic findings in two patients with HD who died 74 and 79 months after transplantation. METHODS: Methods used were pathologic examination, histochemistry, and immunohistochemistry. RESULTS: Neostriatum from both patients showed typical neuropathologic changes of advanced HD. Surviving grafts were identified in both patients (6/6 sites and 7…

[The blocade of glutamate metabotropic 5-th tipe receptors prevents the locomotor behavior changes produced by intrastriatal picrotoxin microinjections in rats].

It was demonstrated in chronic experiments in Wistar rats that only the first of daily multiple microinjections of glutamate metabotropic 5-th type receptor antagonist MTEP into the rostral region of neostriatum impaired the avoidance conditioning in a shuttle box. Within the next two weeks, MTEP was ineffective but being injected into the neostriatum simultaneously with picrotoxin prevented the impairment of avoidance conditioning in a shuttle-box and decreased the hyperactivity (open-field locomotor hyperactivity and choreic hyperkinesis) produced by this GABA-A receptor antagonist. The resu…

[The participation of the neostriatal cholinergic system in the differentiation of acoustic signals in dogs].

In chronic experiments on six dogs the influence was studied of micro-injections of choline agonist carbocholine (0.05-0.2 mkg) and of blocker of choline receptors atropine (40 mkg) in the caudate nucleus head of the left and right hemispheres on realization of instrumental defensive reflexes, connected with the maintenance of definite posture and on differentiation of signals in defensive situation. It has been shown that the cholinergic system of the neostriatum participated in realization of both the motor and sensory mechanisms in connection with the realization of motor responses to defen…

Regional distribution of cholecystokinin binding sites in macaque basal ganglia determined by in vitro receptor autoradiography.

Cholecystokinin binding sites were labeled with [3H]cholecystokinin-8, [125I]cholecystokinin-33, and [125I]cholecystokinin-8 in major structures of macaque basal ganglia by in vitro receptor autoradiography. Analysis of autoradiograms revealed areas of heavy cholecystokinin binding in the neostriatum and substantia nigra that were set off, often quite sharply, from the adjacent globus pallidus and subthalamic nucleus where labeling was, by contrast, very light. Heavy label characterized the ventromedial and posterior parts of the caudate nucleus and adjacent putamen, binding was of moderate in…

Cholinergic synaptic input to different parts of spiny striatonigral neurons in the rat.

The postsynaptic targets of cholinergic boutons in the rat neostriatum were assessed by examination in the electron microscope of boutons that were immunoreactive for choline acetyltransferase, the synthetic enzyme for acetylcholine. These boutons formed symmetrical synaptic specializations with neostriatal neurons. Of 209 immunoreactive synaptic boutons observed in random searches of the neostriatum, 45% made contact with dendritic shafts, 34% with dendritic spines, and 20% with neuronal perikarya. Many of the postsynaptic structures had ultrastructural characteristics of the most common type…

Monoamine changes in the brain of BALB/c mice following sub-lethal infection with Nocardia asteroides (GUH-2).

BALB/c mice injected intravenously with a single, sub-lethal dose of Nocardia asteroides GUH-2 develop several levodopa responsive movement disorders. These included headshake, stooped posture, bradykinesia, and hesitation to forward movement. The changes in monoamine levels in the brain of these mice were determined. There was a significant loss of dopamine with greatly increased dopamine turnover in the neostriatum 7 to 29 days after infection. These effects were specific for dopaminergic neurons since minimal changes were found in neostriatal norepinephrine and serotonin even though seroton…

D2 dopamine receptor localization on striatonigral neurons.

Two major pharmacological classes of dopamine receptors exist in the central nervous system. These receptors have been designated as D1 or D2 based upon their differing pharmacology and influence on the cyclic AMP second messenger system. Different genes for the D1 and D2 dopamine receptors have been isolated and are found to be expressed in high abundance. Within the neostriatum, however the cellular distribution of the dopamine receptors is equivocal. Dopamine receptors are the targets for drugs used to treat neurological dysfunctions such as Parkinson's disease and schizophrenia, and thus k…

Serotonin neural adaptations to ontogenetic loss of dopamine neurons in rat brain.

In rat, the neonatal destruction of nigrostriatal dopamine (DA) neurons by intracerebral administration of 6-hydroxydopamine entails dramatic changes in serotonin (5-hydroxytryptamine, 5-HT) as well as DA function. Most striking is the 5-HT hyperinnervation of the adult neostriatum, associated with increases in density of various 5-HT receptor subtypes and enhanced neuronal responsiveness to the iontophoretic application of 5-HT and its 5-HT(1B/2C) and 5-HT(2A/2C) receptor agonists, m-chlorophenylpiperazine and iododimethoxyphenylaminopropane. The topographical distribution of these changes is…

Changes in reactive oxygen species (ROS) production in rat brain during global perinatal asphyxia: an ESR study.

A large body of evidence suggests that the production of reactive oxygen species (ROS) can play an important role in ischemic neuronal injury. However any studies has been performed in hypoxic conditions. In the present experiments we studied using electron spin resonance (ESR) techniques the ROS release in neostriatum of newborn rats subjected to acute perinatal asphyxia (PA) followed by various periods of reoxygenation. Pregnant rats' uteri still containing foetuses were taken out and subjected to PA by immersion in a 37 degrees C water bath during the following periods of time: 5, 10, 15, 1…

Pallidonigral pigmentation and spheroid formation with multiple striatal lacunar infarcts.

A 72-year-old hypertensive man had focal seizures and mild memory loss. At autopsy, there were many lacunar infarcts, particularly in the neostriatum. Rust-brown discoloration and spheroid formation within the globus pallidus and pars reticulata of the substantia nigra resembled Hallervorden-Spatz disease, in which more detailed analysis of the striatum may be worthwhile.

Frontal cortex stimulation evoked neostriatal potentials in rats: intracellular and extracellular analysis.

Evoked potentials, action potentials and intracellular events were recorded in the neostriatum of urethane anesthetized rats to electrical stimulation of frontal cortex white matter, motor cortex and pre-limbic cortex. Five major waves of the evoked potential were identified. Wave N1 (3.9 msec latency) was small, preceded cellular events and probably represents activation of corticostriate terminals. Wave P1 (10.8 msec latency to peak following white matter stimulation) coincided with an EPSP and neuronal firing. Both wave N2 (38.0 msec latency to peak) and P2 (approximately 110 msec duration)…

Neostriatal evoked inhibition and effects of dopamine on globus pallidal neurons in rat slice preparations.

Spontaneous unit discharges were recorded extracellularly from globus pallidal (GP) neurons in rat slice preparations. The firing rates of GP neurons ranged from 2.0 to 24.0 spikes/s and their firing patterns were predominantly of two types: regular and irregular. Stimulation of the neostriatum evoked two distinct types of inhibition which were dependent on GP neuronal firing patterns, a brief inhibition (about 75 ms) followed by resetting rhythmic neuronal activities and a relatively long-term inhibition (about 100 ms). These inhibitions evoked by neostriatal stimulation were attenuated or co…
