Defects in cardiac conduction system lineages and malignant arrhythmias: developmental pathways and disease.

To unravel the complex disease phenotype of heart failure, we are utilizing an integrative approach employing genomics, physiology, and mouse genetics to identify nodal pathways for specific physiological end points such as myocyte stretch activation responses, contractility and electrical conduction. A new class of genetic pathways for cardiac sudden death and associated arrhythmias has been based on transcription factors that control conduction system lineages, including HF1b/SP4 and NKX2.5. Previous studies have established that HF1b plays a critical role in conduction system lineage format

Conduction of sensory action potentials across the posterior fossa in infratentorial space-occupying lesions in man.

Central conduction time following median nerve stimulation has been recorded in 28 patients with infratentorial space occupying lesions. Fourteen of these (50%) showed a prolonged central conduction time, which invariably involved that segment of the conduction time between the dorsal column nuclei generators and the generation site for P15, which we believe to be the upper brain stem or thalamus. Three other cases showed prolongation of the N14-P15 segments though total central conduction time was within normal limits. One case with marked hydrocephalus showed prolongation both before and aft

Visual Pathways Involvement in Friedreich's Ataxia Patients Without Macular Impairment.

Objectives: Visual Evoked Potential (VEP) abnormalities in Friedreich Ataxia (FA) may result from dysfunction of retinal ganglion cells (RGC), post-retinal visual pathways, or both. Therefore, we aimed to clarify whether in Friedreich Ataxia (FA), abnormal neural conduction is due to dysfunctional RGC, impaired conduction along the post-retinal visual pathways, or both. The secondary aim of the study was to clarify the relationship between a possible RGC and/or post-retinal dysfunction and the patient's age, age at disease onset, and disease duration. Method: Fourteen FA patients (14 eyes; mea…

Auditory brainstem response.

The auditory brainstem response (ABR), consisting of five to six vertex-positive peaks with separation of about 0.8ms, is very sensitive to … ucher disease, and Krabbe disease, the (longer latency) ABR peaks are reduced or absent. Effects on neural conduction, resulting in increased ABR interwave latencies, are found in vestibular schwannomas, Bell's palsy, Duane retraction syndrome, Marcus Gunn ptosis, and various encephalomyopathies. These measures allow an assessment of the parts of the brainstem that …

Autonomic innervation of the human cardiac conduction system: changes from infancy to senility--an immunohistochemical and histochemical analysis.

In order to study the changes in the pattern of autonomic innervation of the human cardiac conduction system in relation to age, the innervation of the conduction system of 24 human hearts (the age of the individuals ranged from newborn to 80 years), freshly obtained at autopsy, was evaluated by a combination of immunofluorescence and histochemical techniques. The pattern of distribution and density of nerves exhibiting immunoreactivity against protein gene product 9.5 (PGP), a general neural marker, dopamine beta-hydroxylase (DBH) and tyrosine hydroxylase (TH), indicators for presumptive symp

A Computational Framework for Myelin as a Regulator of Temporal Integration, Neuronal Excitability, and Synaptic Plasticity.

Myelin regulates neural signaling not only by enabling fast conduction but also by dynamically tuning axonal conduction velocity (CV) and thereby shaping spike timing. While activity-dependent myelination is thought to optimize neural communication, how changes in myelin translate into altered single-cell dynamics and plasticity remains unclear. Using a computational modeling approach, we show that disruptions of axonal CV reorganize presynaptic input correlations, leading to systematic changes in postsynaptic excitability, excitation-inhibition balance, and spike-timing-dependent plasticity.

Human PSC-derived sinoatrial node-cardiac plexus assembloids model innervation-associated maturation of pacemaker systems.

Heart rhythm and contraction are initiated by electrical impulses generated by the sinoatrial node (SAN) and modulated by intrinsic cardiac neural inputs. Despite its physiological importance, human in vitro systems that recapitulate neural-SAN interactions are lacking. Here, we develop SAN-plexus assembloids by integrating human pluripotent stem cell-derived SAN organoids (SANOs) with cardiac ganglionated plexus organoids (CGPOs), together with atrial-like cardiac organoids, to model pacemaker-to-atrial conduction in a tri-assembloid system. This platform exhibits molecular, structural, and e

[Active control of neural circuit synchrony via oligodendroglial lipid synthesis].

Myelin is increasingly recognized as a dynamic regulator of neural circuits, shifting the paradigm from a purely synaptic-centric view of brain plasticity. This report details how activity-dependent changes in myelin lipid composition optimize neural circuit synchrony during motor learning. Using a mouse model with subtle myelin structural deficits, we demonstrate that impaired myelin regulation leads to increased temporal dispersion of axonal conduction. This asynchrony reduces the signal-to-noise ratio of neural activity in the primary motor cortex, thereby hindering motor skill acquisition.

Isolated Phrenic and Vagus Nerve Stimulation and Conduction Suppression Induced by Electroporation Pulses.

BACKGROUND: Pulsed field ablation (PFA) is a novel non-thermal ablation modality with high myocardial selectivity. However, stimulation and transient dysfunction of adjacent neural structures remain important off-target effects. OBJECTIVE: To define pulse-parameter effects on excitation, conduction suppression and recovery of phrenic and vagus nerves exposed to single biphasic electroporation pulses. METHODS: Ex vivo swine phrenic and vagus nerves were studied using a multi-electrode chamber enabling four-channel compound action potential (CAP) recordings. Pulse amplitude, pulse width (0.5-1

Microfluidics-guided localized low-temperature modulation of axonal signal propagation.

Low-temperature stimulation is recognized as a promising approach for neuromodulation, with the potential to suppress or slow neural activity. However, its impact on the spatial and electrophysiological properties of axonal conduction remains poorly understood. Conventional methods have lacked the spatial resolution necessary to isolate axon-specific responses to localized cooling. To overcome these limitations, we developed a microfluidic platform that integrates a microelectrode array (MEA) with a rapid and spatially confined cooling module. This platform enables real-time, phase-resolved mo

Role of myelin plasticity in oscillations and synchrony of neuronal activity.

Conduction time is typically ignored in computational models of neural network function. Here we consider the effects of conduction delays on the synchrony of neuronal activity and neural oscillators, and evaluate the consequences of allowing conduction velocity (CV) to be regulated adaptively. We propose that CV variation, mediated by myelin, could provide an important mechanism of activity-dependent nervous system plasticity. Even small changes in CV, resulting from small changes in myelin thickness or nodal structure, could have profound effects on neuronal network function in terms of spik

Modeling the response of small myelinated axons in a compound nerve to kilohertz frequency signals.

OBJECTIVE: There is growing interest in electrical neuromodulation of peripheral nerves, particularly autonomic nerves, to treat various diseases. Electrical signals in the kilohertz frequency (KHF) range can produce different responses, including conduction block. For example, EnteroMedics' vBloc® therapy for obesity delivers 5 kHz stimulation to block the abdominal vagus nerves, but the mechanisms of action are unclear. APPROACH: We developed a two-part computational model, coupling a 3D finite element model of a cuff electrode around the human abdominal vagus nerve with biophysically-reali

Segmental median nerve conduction measurements discriminate carpal tunnel syndrome from diabetic polyneuropathy.

Median nerve conduction has been compared in CTS, with or without diabetes, and diabetic polyneuropathy. Approximately 90% of hands were correctly predicted as CTS or diabetic polyneuropathy by a comparison including the median antidromic sensory nerve conduction velocities in the elbow-to-wrist segment, wrist-to-palm segment, palm-to-finger segment, and the amplitude of the sensory nerve action potential. CTS with diabetes could not be distinguished from CTS without diabetes. The association between proximal and distal nerve conduction velocities was similar in CTS and diabetic polyneuropathy

Mechanisms of analgesic action of Gln49-PLA(2) from Gloydius ussurensis snake venom.

Gln49-PLA(2) is a phospholipase A(2) isolated from the Gloydius ussurensis snake venom. In this paper, we studied its effect on the function of neural conduction. Electrophysiological studies demonstrated that Gln49-PLA(2) reduced the amplitude of the action potential and the velocity of nerve conduction on isolated mouse sciatic nerve. Patch clamp recordings confirmed that Gln49-PLA(2) significantly decreased neural excitability by the potentiation of sodium channels and the blockade of potassium channels in nerve terminal. In freshly isolated hippocampal pyramidal neurons, 54.25% of potassiu…

[Tumor-induced function disorders of the acoustic nerve and the lower portions of the auditory pathway].

The manyfold features of acoustic neuromas, respectively tumors of the cerebello pontine angle, are tried to explain by pathophysiologic phenomena of the neural conduction. They are the pathologic fatigue of hearing, the disturbed excitability of the acustico facial stapedial reflex, the impaired velocity of nerve conduction, a poor synchronization of conduction as well as the excessively reduced monosyllabic and dichotic discrimination. With respect to the original tissue, localization, size as well as regional spread and growth pressure there result different constellations of symptoms, even…

The role of neural precursor cells and self assembling peptides in nerve regeneration.

OBJECTIVE: Cranial nerve injury involves loss of central neural cells in the brain stem and surrounding support matrix, leading to severe functional impairment. Therapeutically targeting cellular replacement and enhancing structural support may promote neural regeneration. We examined the combinatorial effect of neural precursor cells (NPC) and self assembling peptide (SAP) administration on nerve regeneration. METHODS: Nerve injury was induced by clip compression of the rodent spinal cord. SAPs were injected immediately into the injured cord and NPCs at 2 weeks post-injury. Behavioral analys
