Understanding the beneficial and detrimental effects of donepezil and rivastigmine to improve their therapeutic value.

Cholinesterase enzymes metabolize acetylcholine (ACh). Inhibition of acetylcholinesterase (AChE) in damaged but functional cholinergic synap … regulatory role in controlling the reactivity and functional states of non-excitable cells, such as neuroglia. Current inhibitors of cholinesterases (ChEIs) have similar effects on intrasynaptic ACh, but differ markedly in abilities to upregulate extracellular AChE, inhibit BuChE, and influence the fibrilization of amyloid-β peptides. Importantly, ChEIs can …

Transcriptomic signatures of individual cell types in cerebral cavernous malformation.

Cerebral cavernous malformation (CCM) is a hemorrhagic neurovascular disease with no currently available therapeutics. Prior evidence suggests that different cell types may play a role in CCM pathogenesis. The contribution of each cell type to the dysfunctional cellular crosstalk remains unclear. Herein, RNA-seq was performed on fluorescence-activated cell sorted endothelial cells (ECs), pericytes, and neuroglia from CCM lesions and non-lesional brain tissue controls. Differentially Expressed Gene (DEG), pathway and Ligand-Receptor (LR) analyses were performed to characterize the dysfunctional…

The role of alarmins in neuroinflammation following spinal cord injury: A systematic review of the literature.

BACKGROUND: Alarmins, or damage-associated molecular patterns (DAMPs), are a diverse class of molecules essential for cellular homeostasis; … found to act as alarmins in animal models of SCI. The expression of these molecules in neurons and neuroglia at the SCI lesion site increased levels of TNF-α, IL-1β, and iNOS, contributing to neuroinflammation. Induction of the neurotoxic phenotypes of macrophages, microglia, and astrocytes by IL-1α, HMGB1, and IL-33 promoted cell death and reduction in ol …

Pericyte-endothelial cell interaction increases MMP-9 secretion at the blood-brain barrier in vitro.

The extracellular matrix (ECM) connecting brain capillary endothelial cells (BCEC) with the surrounding brain resident cells is an essential part of the blood-brain barrier (BBB). Represented by the basement membrane which joins BCEC with brain neuroglia (astrocytes and pericytes) it forms a neurovascular unit. Neuroglia-secreted matrix metalloproteinases (MMPs) control the ECM composition and are involved in the integrity and function of the BBB during cell diapedesis and BBB breakdown after ischemia and other CNS diseases. We examined the involvement of pericytes and astrocytes in endothelia…

Transient cerebral ischemia induces the neuroglial proliferative activity and the potential to redirect neuroglial differentiation.

Brain injury triggers a complex response involving morphological changes, cellular proliferation, and differentiation of newly formed neuroglial subpopulations. These processes have been extensively studied in animal stroke models with permanent large vessel occlusion. However, less is known about neuroglial response after transient cerebral ischemia. Herein, we aimed to determine an astrocytic and NG2 glial proliferative response, potential changes in expression of developmental neuroglial markers: vimentin, nestin, oligodendrocyte transcription marker (Olig2), and a role of neuroglial subpop…

Neuroanatomical distribution of galectin-3 in the adult rat brain.

Galectin-3 is a member of the lectin subfamily that enables the specific binding of β-galactosides. It is expressed in a broad spectrum of species and organs, and is known to have various functions related to cell adhesion, signal transduction, and proinflammatory responses. Although, expression of galectin-3 in some activated neuroglia under neuroinflammation has been well documented in the central nervous system, little is known about the neuronal expression and distribution of galectin-3 in normal brain. To describe the cellular and neuroanatomical expression map of galectin-3, we performed…

[Effect of morphine dependence on the hypothalamus of the mother and offspring during postnatal ontogenesis (experimental study)].

Electron microscopy was used to study the time-course of changes in the neuroglia of the hypothalamus. The offspring manifested deceleration of the time of glia maturation with partial destruction of the protein-synthesizing and oxidative-energy structures as well as more frequent occurrence of satellitosis. The mothers and offspring demonstrated membranous bodies in the nucleus and cytoplasm of astrocytes, pronounced osmiophilia of the nucleus of oligodendrocytes accompanied by the impairment of myelination in the function of the latter ones (demyelinization) and axon degeneration by the dark…

Electron microscopic studies on the cerebral lesions of rats in experimental chronic disulfiram poisoning.

Following chronic administration of disulfiram to rats, changes of the brain were examined electron-microscopically. Pathological findings were observed in the nerve cells of the cerebral cortex and hypothalamus at later stage and synaptic changes in the hypothalamus from initial stage. On the other hand, changes of myelinated fibers, neuroglias and capillaries were very slight. It was considered that neurons were affected more predominantly than other neuronal elements by the cytotoxic action of the drug, and that the synaptic changes of the hypothalamus might reveal chronic disturbance of no…

Construction of a rodent neural network-skeletal muscle assembloid that simulate the postnatal development of spinal cord motor neuronal network.

Neuromuscular diseases usually manifest as abnormalities involving motor neurons, neuromuscular junctions, and skeletal muscle (SkM) in postnatal stage. Present in vitro models of neuromuscular interactions require a long time and lack neuroglia involvement. Our study aimed to construct rodent bioengineered spinal cord neural network-skeletal muscle (NN-SkM) assembloids to elucidate the interactions between spinal cord neural stem cells (SC-NSCs) and SkM cells and their biological effects on the development and maturation of postnatal spinal cord motor neural circuits. After coculture with SkM…

Synergy assessment of fixed combinations of Herba Andrographidis and Radix Eleutherococci extracts by transcriptome-wide microarray profiling.

BACKGROUND: Generally accepted, but insufficiently proved, the concept of synergy is based on an assumption that combining of two biologically active substances is justified because the combination is more active and less harmful than the ingredients. HYPOTHESIS: Analysis of RNA microarray of isolated neuroglia cells and the comparison the number of genes deregulated by plant extracts and their fixed herbal formulation might be a useful tool/method for assessment of synergistic and antagonistic interactions of herbal extracts in human organism. AIM: The primary aim of this study was to exten…

[A painful occipital mass revealing a posterior encephalocele].

Encephalocele is a congenital malformation caused by a neural tube defect during embryonic development. We report a case of posterior encephalocele in a 7-month-old infant with a painful occipital mass known since birth. Pathological examination of the mass showed different mature tissues derived from the brain and its coverings (e.g., neuroglia, ependymal canals and clusters of meningothelial cells). A diagnosis of encephalocele was made. The different forms of neural tube defect will be briefly discussed, especially the "aborted" forms (e.g., non-specific midline mass lesion or angioma) that…

[Effects of prion protein on central nervous system and the mechanisms of action].

The following questions are discussed in this paper: (1) Can the bovine prion protein PrpRES infect a man? (2) What is the route of PrPRES infection? (3) In what condition the PrPRES may infect animals, (4) What are the normal functions of PrPc at the surface of neurons and neuroglia cells, (5) What is the structure of the prion protein, (6) What is possible mechanism of the PrPRES destroying neurons. Finally, the two possible mechanisms of the PrPRES to affect central nervous system are analysed. One is to destroy the normal function of the PrPC, the other is to raise the sensibility of the n…

Cytopathological changes in human and animals brains in prion diseases.

Studies of histological preparations of the brains of humans and animals with prion diseases showed that neuron vacuolization often starts and develops intensely in the distal segments of dendrites. Two types of neuron body death were noted - cytolysis and pyknosis, the latter usually being associated with vacuolization of dendrites. The involvement of the main types of neuroglia in the pathogenesis of prion diseases was established, which provides grounds for referring to gliosis rather than astrocytosis. Accumulation of phagocytes (microgliocytes, blood macrophages, pericytes) and release of…
