Evidence for programmed odontoblast process retraction after dentine exposure in the rat incisor.

OBJECTIVE: To re-examine the morphology and potential functions of odontoblasts in intact rat incisors and after cavity preparation into dentine. DESIGN: Intact incisors were fixed, decalcified, snap frozen and sectioned (10μm), before staining with rhodamine phalloidin or antibodies for cyto-skeletal proteins: vimentin and actin, ion transporter: NaK-ATPase, and dendritic cell marker: OX6. Samples with cavity were processed similarly and stained for actin and vimentin before comparing the lengths of odontoblast processes (OP) at baseline, 3h and 24h (n=5 for each group). RESULTS: Actin was

Activation of human dental pulp progenitor/stem cells in response to odontoblast injury.

In restorative dentistry, whilst moderate carious lesion treatment does not significantly compromise odontoblast cell survival, deep cavity preparation may lead to a partial death of these cells. However, newly formed odontoblast-like cells can replace the necrotic odontoblasts and secrete a reparative dentine matrix. Although several lines of evidence strongly suggest the presence of resting progenitor or stem cells in the dental pulp, little is known about the activation and migration of these cells in response to injury. Human immature third molars extracted for orthodontic reasons were use

Direct capping with four different materials in humans: histological analysis of odontoblast activity.

Pulp inflammation in restored teeth is mainly due to the presence of bacteria or bacterial products introduced by microleakage around the restoration or to the material toxicity. Recent knowledge has permitted a precise identification of the risks for pulpal irritation associated with adhesive materials and procedures. The purpose of this work was to evaluate the cellular events that occur in direct pulp exposure capped using different materials. Twenty-four vital teeth without caries, scheduled for extraction for orthodontic reasons, were selected. After a control of the hemostasis, each pulp

Spondin-1 Inhibits Odontoblastic Differentiation of Human Dental Pulp Stem Cells.

Reparative dentin formation is a defensive response that restores a mineralized barrier to protect the dental pulp following various stimuli, such as bacterial invasion, tooth preparation, or restorative materials. However, reparative dentin is limited, and to avoid pathological calcification or pulp canal obliteration, mineral deposition must be restricted to the injured area and temporally restrained once the barrier is reestablished. This suggests the existence of negative regulators that can halt odontoblastic differentiation; however, such inhibitory regulators remain incompletely defined

Chromatin Remodeller BRD9 Orchestrates Odontoblastic Differentiation via Coordinating RUNX2-KLF4.

Dentinogenesis, a process essential for tooth function, relies on the precise differentiation of dental papilla cells into odontoblasts. This lineage commitment is governed by complex transcription factor networks. RUNX2 and KLF4, known key TFs co-expressed in this process, were proven to operate synergistically, but the specific cofactors coordinating their activity at the chromatin level remain unknown. Here, we identify the chromatin remodeller BRD9 as a novel interactor of both RUNX2 and KLF4 during odontoblastic differentiation. To probe their shared function, we generated neural crest-sp

FOXO3a promotes odontoblast differentiation in human dental pulp cells in coordination with RUNX2.

INTRODUCTION: Forkhead box O3a (FOXO3a) is a stress-responsive transcription factor involved in cellular differentiation across multiple tissues. However, its role in odontoblast differentiation and dentin matrix formation remains poorly defined. OBJECTIVE: This study investigated the functional involvement of FOXO3a in odontoblast differentiation and its association with key odontogenic regulators. METHODOLOGY: Human dental pulp cells (HDPCs) were induced to undergo odontoblast differentiation in vitro. FOXO3a gene and protein expression patterns were examined during differentiation and in

Potential role of dental pulp stem cells conditioned medium for odontoblastic differentiation.

BACKGROUND: Functional bioengineered tooth regeneration using autologous or allogeneic alternative differentiated cells sources are thought to have a great potential in replacing conventional dentures. This study investigated the potential of dental pulp stem cells (DPSCs) conditioned medium for odontoblastic differentiation of Wharton's jelly mesenchymal stem cells (WJMSCs). The DPSCs derived from healthy adult permanent first molars were cultured at high confluence prior to conditioned medium collection. The WJMSCs were cultured in six different treatments, with varying ratios of culture med

Dentinal mineralization is not limited in the mineralization front but occurs along with the entire odontoblast process.

The mineralization-front theory is historically rooted in mineralization research fields for many decades. This theory is widely used to describe mineralization events in both osteogenesis and dentinogenesis. However, this model does not provide enough evidence to explain how minerals are propagated from the pulp-end dentin to dentin-enamel junction (DEJ). To address this issue, we modified the current research approaches by a) extending the mineral deposition windows of time from minutes to hours, instead of limiting the mineralization assay on days and weeks only; b) switching a regular fluo

Odontoblast turnover in the impeded and unimpeded rat incisor derived from computerized histomorphometry.

A computerized histomorphometric method was devised to estimate the kinetics of odontoblast turnover and dentinogenesis in rat incisors. The method was applied to two groups of rats: one group with lower incisors in impeded eruption and another group with the left lower incisor in the unimpeded state. The teeth were divided into six equal segments, from which consecutive ground sections were obtained. The distance of each ground section from the posterior border of the alveolar bone was calculated. Each section was magnified, traced, and the tracings fed into a computer by a sonic digitizer. T

Metalloproteinase 14 (MMP-14) and hsa-miR-410-3p expression in human inflamed dental pulp and odontoblasts.

The objective of this study is to evaluate MMP-14 expression in odontoblasts and in the bulk of dental pulp of teeth with pulpitis; to determine the expression of microRNA-410 (miR-410) in pulp tissue, since sequence analysis suggests that miR-410 has potential binding site on MMP-14's 3'UTR, and hence, can regulate expression of the latter one. Tissue samples of dental pulp from teeth with pulpitis and healthy (control) were formalin fixed and paraffin embedded (FFPE). Samples were examined using immunohistochemical staining for MMP-14 and the expression of miR-410 was evaluated using qRT-PCR

Mx1 -labeled pulp progenitor cells are main contributors to postnatal odontoblasts and pulp cells in murine molars.

Regeneration of dentin and odontoblasts from dental pulp stem cells (DPSCs) is essential for permanent tooth maintenance. However, the in vivo identity and role of endogenous DPSCs in reparative dentinogenesis are elusive. Here, using pulp single-cell analysis before and after molar eruption, we revealed that endogenous DPSCs are enriched in Cxcl12- GFP + coronal papilla-like cells with Mx1- Cre labeling. These Mx1 + Cxcl12- GFP + cells are long-term repopulating cells that contribute to the majority of pulp cells and new odontoblasts after eruption. Upon molar injury, Mx1 + DPSCs localize int

Restoring Cell-Cell Junctions in DSPP-Deficient Odontoblasts Through Nanofibrous Topography and Wnt5a-Cdc42 Activation: A Laboratory Investigation.

AIM: Biomaterial-based strategies offer therapeutic potential for inherited disorders such as Dentinogenesis imperfecta type II (DGI-II). DGI-II, a hereditary dental disorder caused by dentine sialophosphoprotein (Dspp) gene mutations, results in fragile, discoloured teeth susceptible to wear and decay. This study investigates the structural alterations in the dentine matrix induced by mutant DSPP and the underlying molecular mechanisms. Furthermore, we evaluated a therapeutic strategy combining nanofibrous substrates with molecular activation of the Wnt5a-Cdc42 pathway, aiming to mitigate the

Myricetin Modulates Matrix Metalloproteinases Expression Induced by TEGDMA in Human Odontoblast-Like Cells.

The activity of matrix metalloproteinases (MMPs) plays a crucial role in the aging of the resin-dentin interface. The in situ action of MMP-2 and MMP-9 has been confirmed in the process of dentin-collagen degradation. However, the involvement of dental pulp cells in MMP secretion as a response to oxidative stress induced by contact with resin monomers has not been fully elucidated. Myricetin (MYR), like proanthocyanidin (PAC), has antioxidant properties and may help prevent extracellular matrix degradation. The objective was to evaluate the effect of MYR on the MMP expression and activity in r

EGCG and Taxifolin Modulate Secretory Activity and Expression of Dentinogenesis Markers in Odontoblast-Like Cells.

Odontoblasts are cells specialized in dentin matrix deposition and the first line of defense when the dentin-pulp complex is injured by pathological processes, such as dental caries and trauma. Natural compounds, such as flavonoids, could be useful to stimulate odontoblast activity and reparative dentinogenesis in vital pulp therapies, especially in immature permanent teeth. This study evaluated the effect of flavonoids on odontoblast secretory activity and the expression of dentinogenesis markers. The effect of flavonoids was evaluated on phenotypic mineralization markers (alkaline phosphatas

Melanoma-inhibiting activity promotes the migration and odontoblastic differentiation of stem cells of apical papilla.

OBJECTIVE: Melanoma Inhibitory Activity (MIA) has been predominantly studied in the context of melanoma and cartilage development. However, its role in dental pulp development and stem cell behavior remains largely unexplored. This study investigates the expression pattern of MIA in dental pulp tissues and its potential role in the proliferation, migration, and odontoblastic differentiation of stem cells from the apical papilla (SCAPs). DESIGN: MIA expression in human pulp tissue was demonstrated by immunohistochemistry. SCAPs were cultured in normal and mineralization induction media, with M

Identification of GPI-anchored protein LYPD1 as an essential factor for odontoblast differentiation in tooth development.

Lipid rafts are membrane microdomains rich in cholesterol, sphingolipids, glycosylphosphatidylinositol-anchored proteins (GPI-APs), and receptors. These lipid raft components are localized at the plasma membrane and are essential for signal transmission and organogenesis. However, few reports have been published on the specific effects of lipid rafts on tooth development. Using microarray and single-cell RNA sequencing methods, we found that a GPI-AP, lymphocyte antigen-6/Plaur domain-containing 1 (Lypd1), was specifically expressed in preodontoblasts. Depletion of Lypd1 in tooth germ using an

Odontoblast-like MDPC-23 cells function as odontoclasts with RANKL/M-CSF induction.

The identity of odontoclast precursors and the molecular mechanism by which odontoclasts resorb dentine remain unclear. MDPC-23 cells were derived from dental papilla cells and possessed odontoblast characteristics such as expressing dentine sialophosphoprotein (Dspp) and dentine matrix acidic phosphoprotein 1 (Dmp1). Here we induced MDPC-23 cells to have an odontoclast-like function with receptor activator of nuclear factor-κB ligand (RANKL) and macrophage colony-stimulating factor (M-CSF). We found that tartrate-resistant acid phosphatase (TRAP)-positive cells, TRAP-positive multinucleated c

The role of heme oxygenase-1 in the proliferation and odontoblastic differentiation of human dental pulp cells.

INTRODUCTION: It was recently reported that heme oxygenase-1 (HO-1) activity is related to stem cell differentiation; however, the involvement of HO-1 in pulp cell growth and differentiation has not been well explored. The purpose of this study was to investigate the role of HO-1 in the growth and differentiation of human dental pulp cells (HDPCs). METHODS: We evaluated cell growth by MTT assay, mineralization by alizarin red staining, and differentiation marker mRNA expression by reverse transcriptase polymerase chain reaction. RESULTS: HO-1 induction by cobaltic protoporphyrin IX (CoPP) in

Dentin sensitivity, odontoblasts and nerves under desiccated or infected experimental cavities. A clinical, light microscopic and ultrastructural investigation.

Cavities were prepared in 35 pairs of young human premolars. One tooth in each pair served as a control. In one Series the test cavity was desiccated with compressed air for one minute. LM and TEM examination of the dentin and the adjacent pulp revealed an extensive aspiration of odontoblasts. Nerve fibers had moved 0.1 to 0.2 mm outward in the tubules. It is suggested that a streching or disruption of the nerve had occurred during desiccation. In a second Series, the test cavity was infected for 1 to 2 weeks. No nerve fibers were seen in the dentinal tubules under the cavities, only cellular
