Neurotensin is a known inhibitor of gastrin-stimulated acid secretion in dogs and humans. In order to study the dose-related effect of neurotensin, we prepared pentobarbital-anesthetized rats by pyloric ligation and collected gastric secretions one hour after injection of saline (Basal), pentagastrin, 6 micrograms/Kg subcutaneously (PG Alone), or pentagastrin plus neurotensin by tail vein injection (PG + NT). Gastric secretion was investigated in 187 subjects under conditions of maximal stimulation with pentagastrin. Thirty subjects, out of them, were healthy, 30--with gastric ulcer, 110--with duodenal ulcer and 17--with double ulcer localization--in duodenum and stomach. The indices of gastric secretion in duodenal ulcer and ulcer disease with double localization are considerably higher than those on the healthy subjects and differ essentially. With single gastric ulcer they are significantly lower than those of the healthy controls.

Basal and pentagastrin-stimulated gastric acid secretion were determined in 122 patients with various types of gastritis and in 20 control subjects. Pentagastrin (0.1-1.0 microgram/kg) first depressed and then augmented the myoelectrical activity of the stomach and pylorus. Acetylcholine (500 mu k/kg) potentiated and atropine (0.2 mg/kg) inhibited the pentagastrin effects. The data suggest involvement of the cholinergic mechanism in the pentagastrin-induced myoelectrical gastroduodenal activity.

Junction potentials at the tip of a 0.15 M NaCl- and a 3 M KCl-flowing bridge dependent on the surrounding electrolyte solution were measured and calculated. An x-ray method modified by the authors was employed in 40 patients with peptic ulcer of the duodenum to examine the effect of pentagastrin and histamine on the rate of gastric evacuation of x-ray capsules 3, 5 and 10 mm in diameter taken with the trial breakfast. The beta 2 adrenergic agonist Salbutamol was infused intravenously in doses of 0.05, 0.1 and 0.2 microgram/kg-min in conscious dogs with gastric fistulae. Salbutamol inhibited meal-stimulated acid (measured with intragastric titration) by up to 82%. Pentagastrin, histalog, glucagon, secretin, and perchlorate were intravenously injected into mice in order to investigate the hormonal regulation of 99mTc pertechnetate uptake by gastric mucosa. Pentagastrin significantly increased uptake; the effect of histalog was weaker. Secretin did not alter the uptake. Glucagon produced some diminution, and perchlorate significantly inhibited gastric uptake. Hormones may play a role in improving the sensitivity and specificity of diagnostic tests for Meckel's diverticulum.

In chronic gastric fistula (GF) rats, HCl and a hyperosmolal solution of polyethylene glycol (PEG) in the upper intestine inhibit pentagastrin-stimulated gastric acid secretion by different mechanisms, but their anatomic sites have not yet been established. BACKGROUND/AIM: Medullary thyroid cancer (MTC) originates from parafollicular cells (C cell) and produces calcitonin (CT). Basal serum CT was used in the diagnosis and treatment of MTC. If basal CT level is 100 pg/mL or higher, it is likely to have MTC, but if basal CT level is below 10 pg/mL, the probability of developing thyroid disease is low. In cases with basal CT level between 10–100 pg/mL, pentagastrin-stimulated (PS) CT level is studied to evaluate MTC and C cell hyperplasia (CHH).
