Reaction between peroxynitrite and boronates: EPR spin-trapping, HPLC Analyses, and quantum mechanical study of the free radical pathway.

Recently, we showed that peroxynitrite (ONOO(-)) reacts directly and rapidly with aromatic and aliphatic boronic acids (k ≈ 10(6) M(-1)s(-1)). Product analyses and substrate consumption data indicated that ONOO(-) reacts stoichiometrically with boronates, yielding the corresponding phenols as the major product (∼85-90%), and the remaining products (10-15%) were proposed to originate from free radical intermediates (phenyl and phenoxyl radicals). Here, we investigated in detail the minor, free radical pathway of boronate reaction with ONOO(-). The electron paramagnetic resonance (EPR) spin-trap

NOx regime-dependent effects of biogenic emissions on toluene degradation and product formation.

Interactions between anthropogenic and biogenic emissions play a critical role in shaping atmospheric chemistry, yet their combined effects remain insufficiently understood. To elucidate these interactions, we selected toluene and isoprene as representative anthropogenic and biogenic volatile organic compound (AVOC and BVOC), respectively. A series of photooxidation experiments were conducted under both low and high NOx regimes to investigate how isoprene influences the photooxidation of toluene and the formation of reaction products. In toluene-only experiments, the fractions of toluene consu

Rational Design of Hemicyanine Platform for Engineering High-Contrast Activatable NIR-II Probes.

Activatable NIR-II hemicyanine probes are crucial tools for high-contrast deep-tissue biomarker imaging. However, their development is constrained by an inherent dilemma: extending π-conjugation to narrow the energy gap inevitably compromises the fluorescence brightness in the activated state (dim on), while the strong intramolecular charge transfer (ICT) effect makes effective quenching in the locked state difficult to achieve (leaky off), leading to poor response sensitivity and a low on/off ratio. Here, we rationally designed a xanthene-cyanine hybrid NIR-II activatable platform (RC-OH), wh

A highly selective near-infrared fluorescent probe for imaging peroxynitrite in the brains of epileptic mice.

A ratiometric near-infrared probe, CCy7-DP, was developed for peroxynitrite detection, utilizing a highly selective addition-elimination reaction between ONOO- and diphenyl phosphinate. This reaction triggered an electron rearrangement of the cyanine skeleton and a blue shift in fluorescence, enabling high-contrast imaging of peroxynitrite in the brains of epileptic mice.

A Novel Colon-Targeting Ratiometric Probe with Large Emission Shift for Imaging Peroxynitrite in Ulcerative Colitis.

Ulcerative colitis (UC) is an inflammatory disease that leads to the overexpression of peroxynitrite (ONOO-). Up to now, it has been a challenge to design a colon-targeted fluorescent probe with a large emission shift that can detect and image ONOO- in UC mice. Here, a fluorophore (pyran-coumarin) is linked with cholic acid to develop a colon-targeted fluorescent probe (CPC) for the detection of ONOO-. The fluorescent probe showed strong near-infrared emission at 725 nm. After reacting with ONOO-, it can be observed that the fluorescence intensity at 725 nm decreases obviously, and the signal

BODIPY-Based Ratiometric Fluorescent Probe for Sensing Peroxynitrite in Inflammatory Cells and Tissues.

Peroxynitrite (ONOO-) plays an important role in many physiological and pathological processes. Excessive ONOO- in cells leads to oxidative stress and inflammation. However, precise monitoring of ONOO- levels in specific organelles (e.g., mitochondria) is still lacking and urgently needed. Herein, we rationally designed a mitochondria-targeted ratiometric fluorescent probe, MOBDP-I, for imaging of ONOO- in the mitochondria of inflammatory cells and model mice. This probe, MOBDP-I, was synthesized by conjugating a BODIPY fluorophore to a mitochondria-targeting moiety-indole-salt group by a carb

De Novo Design of Near-Infrared Fluorescent Agents Activated by Peroxynitrite and Glutathione-Responsive Imaging for Diabetic Liver Disease.

Diabetes and its complications, such as diabetes liver disease, is a major problem puzzling people's health. The detection of redox states in its pathological process can effectively help us gain a deeper understanding of the disease. The pair of oxidation-reduction substances peroxynitrite (ONOO- ) and glutathione (GSH) is considered to be closely related to their occurrence and development. Thus, direct visualization of ONOO- and GSH in diabetes liver disease is critical to evaluate the disease at the molecular level. Herein, two activatable agents NTCF-ONOO- and NTCF-GSH are prepared for se

FeTMPyP a peroxynitrite decomposition catalyst ameliorated functional and behavioral deficits in chronic constriction injury induced neuropathic pain in rats.

Neuropathic pain is a maladaptive pain phenotype that results from injury or damage to the somatosensory nervous system and is proposed to be linked to a cascade of events including excitotoxicity, oxidative stress, mitochondrial dysfunction, neuroinflammation and apoptosis. Oxidative/nitrosative stress is a critical link between neuroinflammation and neurodegeneration through poly (ADP) ribose polymerase (PARP) overactivation. Hence, the present study investigated the antioxidant and anti-inflammatory effects of peroxynitrite decomposition catalyst; FeTMPyP in chronic constriction injury (CCI

Pheomelanin Effect on UVB Radiation-Induced Oxidation/Nitration of l-Tyrosine.

Pheomelanin is a natural yellow-reddish sulfur-containing pigment derived from tyrosinase-catalyzed oxidation of tyrosine in presence of cysteine. Generally, the formation of melanin pigments is a protective response against the damaging effects of UV radiation in skin. However, pheomelanin, like other photosensitizing substances, can trigger, following exposure to UV radiation, photochemical reactions capable of modifying and damaging cellular components. The photoproperties of this natural pigment have been studied by analyzing pheomelanin effect on oxidation/nitration of tyrosine induced by

Nutraceuticals Targeting Generation and Oxidant Activity of Peroxynitrite May Aid Prevention and Control of Parkinson's Disease.

Parkinson's disease (PD) is a chronic low-grade inflammatory process in which activated microglia generate cytotoxic factors-most prominently peroxynitrite-which induce the death and dysfunction of neighboring dopaminergic neurons. Dying neurons then release damage-associated molecular pattern proteins such as high mobility group box 1 which act on microglia via a range of receptors to amplify microglial activation. Since peroxynitrite is a key mediator in this process, it is proposed that nutraceutical measures which either suppress microglial production of peroxynitrite, or which promote the

Direct Resonance Raman Characterization of a Peroxynitrito Copper Complex Generated from O2 and NO and Mechanistic Insights into Metal-Mediated Peroxynitrite Decomposition.

We report the formation of a new copper peroxynitrite (PN) complex [CuII (TMG3 tren)(κ1 -OONO)]+ (PN1) from the reaction of [CuII (TMG3 tren)(O2.- )]+ (1) with NO.(g) at -125 °C. The first resonance Raman spectroscopic characterization of such a metal-bound PN moiety supports a cis κ1 -(- OONO) geometry. PN1 transforms thermally into an isomeric form (PN2) with κ2 -O,O'-(- OONO) coordination, which undergoes O-O bond homolysis to generate a putative cupryl (LCuII -O. ) intermediate and NO2. . These transient species do not recombine to give a nitrato (NO3- ) product but instead proceed to effe

A Cytochrome P450 TxtE Model System with Mechanistic and Theoretical Evidence for a Heme Peroxynitrite Active Species.

The cytochrome P450 homolog, TxtE, efficiently catalyzes the direct and regioselective aromatic nitration of the indolyl moiety of L-tryptophan to 4-nitro-L-tryptophan, using nitric oxide (NO) and dioxygen (O2) as co-substrates. Pathways for such direct and selective nitration of heteroaromatic motifs present platforms for engineering new nitration biocatalysts for pharmacologically beneficial targets, among a medley of other pivotal industrial applications. Precise mechanistic details concerning this pathway are only weakly understood, albeit a heme iron(III)-peroxynitrite active species has

A Mitochondria-Targeting and Peroxynitrite-Activatable Ratiometric Fluorescent Probe for Precise Tracking of Oxidative Stress-Induced Mitophagy.

Mitochondria are the energy factory of cells and can be easily damaged by reactive oxygen species (ROS) because of the frequent occurrence of oxidative stress. Abnormality in mitophagy is often associated with many diseases including inflammation, cancer, and aging. While previously developed fluorescent probes mainly focus on detecting just ROS or mitophagy, quite rare studies have endeavored to comprehensively capture the entire mitophagic process, encompassing both the production of ROS and the induction of mitophagy. Herein, we report a new ratiometric fluorescent probe NA-DP for tracking

Screening and optimization of a water-soluble near-infrared fluorescent probe for drug-induced liver injury monitoring.

Peroxynitrite (ONOO-) is a potential biomarker of drug-induced liver injury (DILI) and is involved in the process of DILI. Therefore, developing a reliable detection method for ONOO- will greatly contribute to ensuring drug safety and improving treatment efficiency. Here, based on the previous work, two kinds of NIR fluorescence probes PN and SPN were developed with phenyl-hydrazine as the ONOO- recognition group, which based on two fluorophores RN and SRN that are stable to ONOO-. A sensitive NIR probe SPN with good water solubility, low detection limit and good biocompatibility was selected

Novel near-infrared fluorescence probe with large Stokes shift for monitoring CCl4-induced toxic hepatitis.

Toxic hepatitis which is induced by chemical substance is a serious threat to human health. More and more studies have shown that peroxynitrite (ONOO-) is related with the development of toxic hepatitis. So it is important to find a tool to study ONOO- change during the diagnosis and therapy of toxic hepatitis. Herein, a series of novel near-infrared (NIR) fluorescence dyes (DDM-R) with long emission wavelength (740-770 nm) and large Stokes shift (~200 nm) are developed. Among the dyes, DDM-OH with great spectral performance and facilely modified feature is used to construct probe DDM-ONOO-. T

Nitric Oxide Dioxygenase Activity of a Nitrosyl Complex of Cobalt(II) Porphyrinate in the Presence of Hydrogen Peroxide via Putative Peroxynitrite Intermediate.

The reaction of a cobalt porphyrin complex, [(F8TPP)Co], 1 {F8TPP = 5,10,15,20- tetrakis(2,6-difluorophenyl)porphyrinate dianion} in dichloromethane with nitric oxide (NO) led to the nitrosyl complex, [(F8TPP)Co(NO)], 2. Spectroscopic studies and structural characterization revealed it as a bent nitrosyl of {CoNO}8 description. It was stable in the presence of dioxygen. However, it reacts with H2O2 in acetonitrile (or THF) solution at -40 °C (or -80 °C) to result in the corresponding Co(III)-nitrate complex, [(F8TPP)Co(NO3)], 3. The reaction presumably proceeds via the formation of a Co-peroxy

Proteomic analysis of peroxynitrite-induced protein nitration in isolated beef heart mitochondria.

Mitochondria are exposed to reactive nitrogen species under physiological conditions and even more under several pathologic states. In order to reveal the mechanism of these processes we studied the effects of peroxynitrite on isolated beef heart mitochondria in vitro. Peroxynitrite has the potential to nitrate protein tyrosine moieties, break the peptide bond, and eventually release the membrane proteins into the solution. All these effects were found in our experiments. Mitochondrial proteins were resolved by 2D electrophoresis and the protein nitration was detected by immunochemical methods
