Identification of expression profiles of sorghum genes in response to greenbug phloem-feeding using cDNA subtraction and microarray analysis.

The phloem-feeding by greenbug (Schizaphis graminum) elicits unique interactions with their host plants. To investigate the expression profiles of sorghum genes responsive to greenbug feeding, two subtractive cDNA libraries were constructed through different combinatorial subtractions in a strong greenbug resistance sorghum M627 line and a susceptible Tx7000 line with or without greenbug infestation. A total of 3,508 cDNAs were selected from the two cDNA libraries, and subsequent cDNA microarray and northern blot analyses were performed for identification of sorghum genes responsive to greenbu…

Elevation-Driven Adaptive Shifts in Phloem Anatomy and Non-Structural Carbohydrates: Differential Responses of Pinus koraiensis and Acer mono.

Phloem is crucial for long-distance transport, but its coordinated phenological, anatomical, and nonstructural carbohydrate (NSC) responses to environmental changes remain unclear. Along a 750-1150 m elevational gradient on the Changbai Mountains of China, we monitored phloem growth, anatomy, and total NSC (soluble sugars and starch) in Pinus koraiensis and Acer mono during the 2024 growing season. Elevation delayed phloem formation and compressed the growing season in both species. However, the two wood types diverged in adaptive strategies. P. koraiensis employed an active structural compens…

Spider toxin (Hvt) gene cloned under phloem specific RSs1 and RolC promoters provides resistance against American bollworm (Heliothis armigera).

Spider venoms are neurotoxin proteins that can kill insects. Spider toxin Hvt gene was cloned under two phloem specific RSs1 and RolC promoters, transformed into tobacco plants through Agrobacterium-mediated transformation and tested against Heliothis armigera larvae. Transgenic plants were confirmed through PCR. First instar larvae of H. armigera were released on detached leaves of transformed and non-transformed plants. Insect bioassays showed 93-100% mortality of H. armigera larvae within 72 h on the leaves of transgenic plants while all larvae survived and continued feeding on detached lea…

Both heavy metal-amendment of soil and aphid-infestation increase Cd and Zn concentrations in phloem exudates of a metal-hyperaccumulating plant.

Plants that are able to hyperaccumulate heavy metals show increased concentrations of these metals in their leaf tissue. However, little is known about the concentrations of heavy metals and of organic defence metabolites in the phloem sap of these plants in response to either heavy metal-amendment of the soil or biotic challenges such as aphid-infestation. In this study, we investigated the effects of heavy metal-exposure and of aphid-infestation on phloem exudate composition of the metal hyperaccumulator species Arabidopsis halleri L. O'Kane & Al-Shehbaz (Brassicaceae). The concentrations of…

Design, synthesis, antifungal activity, and phloem mobility of novel aromatic amide phenylthiazole-4-carboxylic acid derivatives as potential succinate dehydrogenase inhibitors.

BACKGROUND: Although succinate dehydrogenase inhibitors (SDHIs) display potent antifungal activity, the majority possess weak phloem mobility, limiting their efficacy against root and vascular diseases. To develop novel SDHI candidates with phloem mobility, 36 aromatic (heterocyclic) amide phenylthiazole-4-carboxylic acid derivatives were synthesized and subjected to antifungal evaluation. RESULTS: In vitro bioassays showed that compounds E11 and E21 exhibited high antifungal activities against Sclerotinia sclerotiorum, with median effective concentration (EC50) values of 0.34 and 0.19 μg mL-…

Nitric oxide generation in Vicia faba phloem cells reveals them to be sensitive detectors as well as possible systemic transducers of stress signals.

Vascular tissue was recently shown to be capable of producing nitric oxide (NO), but the production sites and sources were not precisely determined. Here, NO synthesis was analysed in the phloem of Vicia faba in response to stress- and pathogen defence-related compounds. The chemical stimuli were added to shallow paradermal cortical cuts in the main veins of leaves attached to intact plants. NO production in the bare-lying phloem area was visualized by real-time confocal laser scanning microscopy using the NO-specific fluorochrome 4,5-diaminofluorescein diacetate (DAF-2 DA). Abundant NO genera…

Quantification of Callose Deposition in the Phloem of Woody Stems Using Supervised Machine Learning-Driven Automated Image Analysis with the IlastiKlean R Package.

Callose deposition in the phloem is an innate part of plant development and a response to biotic and abiotic stress, aiding in stress mitigation but potentially also compromising phloem functionality. Measuring callose using aniline blue staining is widely employed, but accurate quantification is hindered by image qualities such as texture and fluorescent artifacts. Here, we describe a method to quantify callose levels in the phloem of woody plants using aniline blue staining, confocal microscopy, and automated supervised machine learning-driven image analysis supported by the IlastiKlean R pa…

Isotope Measurements of Phloem-Transported Organic Matter: Tools to Obtain Information on Gas Exchange Physiology and Transport Processes, and to Better Understand Tree Ring Archives.

Stable isotope analysis of phloem-transported organic matter provides valuable insights into plant physiological processes. This paper reviews methodologies for phloem sampling suitable for isotope measurements, including phloem bleeding from bark incisions, exudation into aqueous solutions, and centrifugation techniques. I highlight critical considerations for preventing sample contamination and degradation across these methods. I demonstrate how phloem isotope measurements offer crown-integrated information on gas exchange physiology, connecting stomatal conductance and photosynthetic assimi…

Identification of high levels of phytochelatins, glutathione and cadmium in the phloem sap of Brassica napus. A role for thiol-peptides in the long-distance transport of cadmium and the effect of cadmium on iron translocation.

Phytochelatins (PCs) are glutathione-derived peptides that function in heavy metal detoxification in plants and certain fungi. Recent research in Arabidopsis has shown that PCs undergo long-distance transport between roots and shoots. However, it remains unknown which tissues or vascular systems, xylem or phloem, mediate PC translocation and whether PC transport contributes to physiologically relevant long-distance transport of cadmium (Cd) between shoots and roots. To address these questions, xylem and phloem sap were obtained from Brassica napus to quantitatively analyze which thiol species …

Copper in xylem and phloem saps from rice (Oryza sativa): the effect of moderate copper concentrations in the growth medium on the accumulation of five essential metals and a speciation analysis of copper-containing compounds.

Copper (Cu) is an essential element for cereals, playing important roles as a cofactor of several enzymes. Copper and four other metals (Fe, Mn, Zn and Mo) taken up by roots are efficiently delivered to the shoots via xylem and phloem. Here we investigated the concentrations of Cu, Fe, Mn, Zn and Mo in the xylem and phloem saps as well as in tissues of rice (Oryza sativa L.) seedlings when they were grown under different Cu levels in culture solution. Although the Cu concentrations in the roots and the Mn concentrations in the mature shoot tissues were increased with the increase of the Cu lev…

Concentrations of metals and potential metal-binding compounds and speciation of Cd, Zn and Cu in phloem and xylem saps from castor bean plants (Ricinus communis) treated with four levels of cadmium.

We examined the concentrations of metals (Cd, Zn, Cu, Fe and Mn) and potential metal-binding compounds [nicotianamine (NA), thiol compounds and citrate] in xylem and phloem saps from 4-week-old castor bean plants (Ricinus communis) treated with 0 (control), 0.1, 1.0, and 10 μM Cd for 3 weeks. Treatment with 0.1 and 1 μM Cd produced no visible damage, while 10 μM Cd retarded growth. Cadmium concentrations in both saps were higher than those in the culture solution at 0.1 μM, similar at 1.0 μM and lower at 10 μM. Cd at 10 μM reduced Cu and Fe concentrations in both saps. NA concentrations measur…

Study on the Design, Synthesis, Bioactivity and Translocation of the Conjugates of Phenazine-1-carboxylic Acid and N-Phenyl Alanine Ester.

The natural pesticide phenazine-1-carboxylic acid (PCA) is known to lack phloem mobility, whereas Metalaxyl is a representative phloem systemic fungicide. In order to endow PCA with phloem mobility and also enhance its antifungal activity, thirty-two phenazine-1-carboxylic acid-N-phenylalanine esters conjugates were designed and synthesized by conjugating PCA with the active structure N-acylalanine methyl ester of Metalaxyl. All target compounds were characterized by 1H NMR, 13C NMR and HRMS. The antifungal evaluation results revealed that several target compounds exhibited moderate to potent …

Prior Cereal Leaf Beetle (Oulema spp.) Window-Pane Feeding Has Weak Systemic Legacy on the Wheat Dwarf Virus Vector Psammotettix alienus in Barley.

Plant-mediated interactions can modify herbivore behaviour and vector-borne plant disease dynamics, yet the systemic consequences of prior cereal leaf beetle (Oulema spp.) window-pane feeding for later phloem feeder vectors remain unclear. We hypothesised that prior Oulema window-pane feeding induces systemic changes in barley that alter the subsequent host acceptance and feeding behaviour of Psammotettix alienus. Plants received one of three 24 h treatments on one leaf: Oulema pre-colonisation, standardised artificial vascular injury or an undamaged control. Leafhopper responses were measured…

The impact of seasonal variations in inner bark width and xylem growth on irreversible annual stem diameter measurements.

To better interpret continuous xylem growth measurements from dendrometers, we evaluated how the widths of developing xylem and phloem, along with seasonal trends in inner bark width (conducting phloem, non-conducting phloem and the youngest periderm), contribute to irreversible annual stem diameter changes in Pinus nigra, Fraxinus ornus and Ostrya carpinifolia. Histometric analyses were performed on microcores taken from tree stems in 2020. The seasonal patterns of xylem and phloem formation, as well as changes in inner bark width and xylem growth, varied both within and among species. Of the…

Anatomical Features of Foliar Vasculature as Flux-Capacity Proxies.

We present a procedure for preparing leaf tissue for minor-vein analysis using light and transmission electron microscopy that facilitates quantification of phloem and xylem features that correlate with photosynthetic capacity or transpiration rates, respectively, and thus provide proxies for foliar sugar- and/or water-transport capacity. We suggest here that the numbers and individual sizes of phloem cells of foliar minor veins (sometimes normalized by vein density) and transfer-cell wall ingrowths provide quantitative proxies for the capacity of the leaf to load and export photosynthates. Am…

Detecting Rapid Changes in Carbon Transport and Partitioning with Carbon-11 (11C).

Noninvasive techniques to measure phloem transport of carbon will be crucial to efforts to engineer improved crop yields, which are highly dependent on carbon partitioning. Phloem, which is buried in the interior of the plant, is highly sensitive to tissue damage. Here we describe nondestructive methods using carbon-11, fed to leaves as 11CO2, as a tracer to track export of recently fixed carbon from leaves, transport speed through the phloem, and distribution or partitioning throughout the plant.

Effect of mineral nutritional status on shoot-root partitioning of photoassimilates and cycling of mineral nutrients.

Mineral nutrients taken up by the roots are, as a rule, transported in the xylem to the shoot, and photoassimilates transported in the phloem to the roots. According to the Thornley model of photosynthate partitioning, nutrient deficiencies should favour photosynthate partitioning to the roots. Examples are cited to show that this preferential partitioning is dependent on phloem mobility and hence on nutrient cycling from shoot to roots. Thus, root growth is enhanced under nitrogen and phosphorus deficiencies, but not under deficiencies of nutrients of low mobility in the phloem, such as calci…

Measurement of Inner Bark and Leaf Osmolality.

Sugar transport in the phloem is driven by turgor pressure gradients, which are created by osmotic gradients resulting from sugars loaded into the phloem at the source tissue and unloaded at the sink tissue. Therefore, osmolality is a key parameter that can be used to evaluate sugar status and to indicate the driving force for phloem transport. Here, we describe how osmotic concentration measurements from inner bark and needles of trees can be made. This protocol presents the procedure used by Lintunen et al. [15] and Paljakka et al. [16], and extends it with practical advice and discussion of…
