Placentation defects are highly prevalent in embryonic lethal mouse mutants.

Large-scale phenotyping efforts have demonstrated that approximately 25-30% of mouse gene knockouts cause intrauterine lethality. Analysis of these mutants has largely focused on the embryo and not the placenta, despite the crucial role of this extraembryonic organ for developmental progression. Here we screened 103 embryonic lethal and sub-viable mouse knockout lines from the Deciphering the Mechanisms of Developmental Disorders program for placental phenotypes. We found that 68% of knockout lines that are lethal at or after mid-gestation exhibited placental dysmorphologies. Early lethality (

Placentation in Sigmodontinae: a rodent taxon native to South America.

BACKGROUND: Sigmodontinae, known as "New World rats and mice," is a large subfamily of Cricetidae for which we herein provide the first comp … This survey of wild-trapped specimens from five genera is a useful starting point for the study of placentation in an important subfamily of South American rodents. We note, however, that some of these rodents can be captive bred and recommend that future studies focus on the study of time dated pregnancies.

Defective implantation and placentation: laying the blueprint for pregnancy complications.

Normal implantation and placentation is critical for pregnancy success. Many pregnancy-related complications that present late in gestation (such as pre-eclampsia and preterm labour) appear to have their origins early in pregnancy with abnormalities in implantation and placental development. Implantation is characterized by invasion of the maternal tissues of the uterus by fetal trophoblast, and the degree to which trophoblast invades these tissues appears to be a major determinant of pregnancy outcome. Excessive invasion can lead to abnormally firm attachment of the placenta to the myometrium…

The biology of the twinning process: how placentation influences outcome.

Placentation of twins has important bearing on the fetal development and neonatal outcome. Because of limitation in space of the endometrial cavity, twin placentas frequently collide during their expansion. This may restrict their growth and also lead to a somewhat increased rate of placenta previa. Moreover, abnormal cord insertions such as marginal and velamentous cords are much more frequent in multiple gestation. Absence of one umbilical artery is also much more frequent in twins. The best prognosis is had when two independent placentas develop, the dichorionic separate organs. Fusion of t…

Placentation Disruption by Perfluorooctanoic Acid and Perfluorooctanesulfonate in Human Trophoblast Organoids.

Prenatal exposure to perfluorooctanoic acid (PFOA) and perfluorooctanesulfonate (PFOS) is associated with low birth weight, a condition often resulting from placental dysfunction. However, whether and how PFOA and PFOS affect human placentation and placental-specific functions remains unclear. In this study, we reconstructed a human trophoblast organoid model, incorporating a near-physiological proportion of extravillous trophoblast (EVT). The organoids were exposed to PFOA or PFOS for 7 days. Exposure to PFOA at 10 nM significantly increased the proportion of villous cytotrophoblast (CTB) cel…

Third-trimester resolution of low placentation and risk of postpartum hemorrhage.

OBJECTIVE: Low placentation is associated with increased risk of postpartum hemorrhage (PPH). There is a paucity of data on the association between second trimester low placentation that later resolves, and PPH. Our objective was to investigate the association of resolved low placentation and other prenatal ultrasound markers, and PPH with delivery. STUDY DESIGN: This was a retrospective matched case-control (1:1) study of full term, singleton pregnancies complicated by PPH who delivered vaginally at our academic center between 2018-2022. All patients received a transvaginal ultrasound in the…

Histone modifications in embryo implantation and placentation: insights from mouse models.

Embryo implantation and placentation play pivotal roles in pregnancy by facilitating crucial maternal-fetal interactions. These dynamic processes involve significant alterations in gene expression profiles within the endometrium and trophoblast lineages. Epigenetics regulatory mechanisms, such as DNA methylation, histone modification, chromatin remodeling, and microRNA expression, act as regulatory switches to modulate gene activity, and have been implicated in establishing a successful pregnancy. Exploring the alterations in these epigenetic modifications can provide valuable insights for the…

Understanding placentation in ruminants: a review focusing on cows and sheep.

Mammals differ regarding their placentae, but in all species placental trophoblasts interact intimately with the uterine endometrium to mediate the transfer of nutrients from the mother to the embryo/fetus through the closely juxtaposed microcirculatory systems of the uterus and placenta. Placentation in ruminants is intermediate between the non-invasive type, as observed in the epitheliochorial placenta of pigs, and the invasive type, as observed in the haemochorial placentae of mice and humans. In ruminants, placental trophoblast cells invade uterine endometrial tissue, but invasion is belie…

Function and Hormonal Regulation of GATA3 in Human First Trimester Placentation.

Pregnancies resulting from fresh in vitro fertilization (IVF) cycles exposed to supraphysiologic estrogen levels have been associated with h … hormonally regulated and plays an important functional role in trophoblast migration, invasion, and placentation. We found that GATA3 expression was hormonally regulated by estradiol in HTR8/SVneo first trimester trophoblast cells; however, no change in expression was seen with progesterone treatment. Furthermore, GATA3 knockdown resulted in decreased HTR8/SVn …

Dose effect of gestational ethanol exposure on placentation and fetal growth.

INTRODUCTION: Prenatal ethanol exposure compromises fetal growth by impairing placentation. Invasive trophoblastic cells, which mediate placentation, express the insulin-IGF regulated gene, aspartyl-asparaginyl β-hydroxylase (ASPH), which has a critical role in cell motility and invasion. The aims of this study were to characterize effects of ethanol on trophoblastic cell motility, and assess ethanol dose-dependent impairments in placentation and fetal development. METHODS: Pregnant Long Evans dams were fed with isocaloric liquid diets containing 0%, 8%, 18% or 37% ethanol (caloric content) f…

The potential perinatal origin of placentation disorders in the young primigravida.

The fetus is exposed to high plasma concentrations of unbound estrogens and progesterone throughout pregnancy. However, secretory or decidual changes in the fetal uterus occur relatively infrequently before birth, suggesting a variable endometrial progesterone response at the time of birth. Arguably, partial progesterone resistance that persists into adolescent years may compromise the physiological transformation of the spiral arteries and predispose for defective placentation in the case of pregnancy. Decidualization of the endometrial stromal compartment and junctional zone myometrium prece…

The "Great Obstetrical Syndromes" are associated with disorders of deep placentation.

Defective deep placentation has been associated with a spectrum of complications of pregnancy including preeclampsia, intrauterine growth restriction, preterm labor, preterm premature rupture of membranes, late spontaneous abortion, and abruptio placentae. The disease of the placental vascular bed that underpins these complications is commonly investigated with targeted biopsies. In this review, we critically evaluate the biopsy technique to summarize the salient types of defective deep placentation, and propose criteria for the classification of defective deep placentation into 3 types based …

Distribution and maturity of dendritic cells in diseases of insufficient placentation.

PROBLEM: The immunological equilibrium at the feto-maternal interphase contributes towards late gestational diseases like growth restriction (IUGR) pre-eclampsia (PE) and hemolysis, elevated liver enzymes, low platelets (HELLP)-syndrome. The state of activation of decidual dendritic cells (DC) has emerged as one of the central players influencing this immunological equilibrium. METHOD OF STUDY: Paraffin-embedded tissue sections from 27 pregnancies were immunostained for DC markers DEC-205, DC-SIGN, DC-LAMP and costained for DC-SIGN/CD56 and DC-SIGN/ vascular endothelial growth factor receptor

Patterns of trophoblast migration in deep uterine arteries in accreta placentation.

INTRODUCTION: During normal placentation, extravillous trophoblast (EVT) colonises and, in synergy with maternal leukocytes, transforms the walls of uterine spiral arteries in the decidua and inner myometrium. In placenta accreta spectrum (PAS), migration of extravillous trophoblasts (EVT) is abnormally deep, reaching larger upstream arteries in myometrium. As little is known about their interactions in accreta areas, scar tissue and deeper arteries were examined for colonisation and remodelling by trophoblast and maternal inflammatory cells. METHODS: Samples (n = 79) were fresh hysterectomy …
