Calcium-ROS-NO signalling integration in plant immunity: mechanistic interactions and feedback regulation.

This review establishes Ca2+, ROS and NO signalling as an integrated feedback network that links pathogen perception, calcium decoding, local and systemic defence, and maintenance of immune homeostasis across tissues. Calcium (Ca2+) signalling is a central component of plant immune responses and provides a mechanistic link between pathogen perception and downstream defence activation. Immune stimuli generate distinct spatial and temporal Ca2+ signatures that are decoded by calcium sensor proteins, including calcium-dependent protein kinases, calcineurin B-like protein-interacting protein kinas

Disulfide-bond-driven neck-ring-like conformation mediates SCREW recognition in plant immunity.

Secreted cysteine-rich peptides (CRPs) are vital plant signalling molecules, yet how their essential intramolecular disulfide bonds structurally mediate receptor complex activation is poorly understood. Here we present the crystal structure of an Arabidopsis immune complex comprising the CRP SMALL PHYTOCYTOKINES REGULATING DEFENSE AND WATER LOSS (SCREW), the receptor PLANT SCREW UNRESPONSIVE RECEPTOR (NUT) and the coreceptor BRASSINOSTEROID INSENSITIVE 1-ASSOCIATED RECEPTOR KINASE 1 (BAK1). Unlike typical multi-disulfide CRPs with compact folds, SCREW maintains a flexible loop constrained into

An apoplastic effector Uv7366 contributes to Ustilaginoidea virens virulence and elicits plant immunity.

Ustilaginoidea virens (Cook) Takahashi, the causal agent of rice false smut disease (RFS), causes severe losses in yield and grain quality worldwide. The ascomycete pathogen deploys a large arsenal of effectors to facilitate infection, but only a few have been functionally characterized. In this study, we characterized Uv7366, an apoplastic glycoside hydrolase family 12 (GH12) protein that was previously identified in the culture filtrate of U. virens. Purified Uv7366 triggered a reactive oxygen species (ROS) burst and strongly induced the transcription of multiple defense-associated genes, an

Cross-kingdom RNA interference mediated by insect salivary microRNAs may suppress plant immunity.

Communication between insects and plants relies on the exchange of bioactive molecules that traverse the species interface. Although proteinic effectors have been extensively studied, our knowledge of other molecules involved in this process remains limited. In this study, we investigate the role of salivary microRNAs (miRNAs) from the rice planthopper Nilaparvata lugens in suppressing plant immunity. A total of three miRNAs were confirmed to be secreted into host plants during insect feeding. Notably, the sequence-conserved miR-7-5P is specifically expressed in the salivary glands of N. lugen

Root-associated Streptomyces produce galbonolides to modulate plant immunity and promote rhizosphere colonization.

The rhizosphere, which serves as the primary interface between plant roots and the soil, constitutes an ecological niche for a huge diversity of microbial communities. Currently, there is little knowledge on the nature and the function of the different metabolites released by rhizospheric microbes to facilitate colonization of this highly competitive environment. Here, we demonstrate how the production of galbonolides, a group of polyene macrolides that inhibit plant and fungal inositol phosphorylceramide synthase (IPCS), empowers the rhizospheric Streptomyces strain AgN23, to thrive in the rh

Plant-produced Bluetongue chimaeric VLP vaccine candidates elicit serotype-specific immunity in sheep.

Bluetongue (BT) is a hemorrhagic non-contagious, biting midge-transmitted disease of wild and domestic ruminants that is caused by bluetongue virus (BTV). Annual vaccination plays a pivotal role in BT disease control in endemic regions. Due to safety concerns of the current BTV multivalent live attenuated vaccine (LAV), a safe efficacious new generation subunit vaccine such as a plant-produced BT virus-like particle (VLP) vaccine is imperative. Previously, homogenous BTV serotype 8 (BTV-8) VLPs were successfully produced in Nicotiana benthamiana plants and provided protective immunity in sheep

Biosynthesis and Regulation of Salicylic Acid and N-Hydroxypipecolic Acid in Plant Immunity.

Salicylic acid (SA) has long been known to be essential for basal defense and systemic acquired resistance (SAR). N-Hydroxypipecolic acid (NHP), a recently discovered plant metabolite, also plays a key role in SAR and to a lesser extent in basal resistance. Following pathogen infection, levels of both compounds are dramatically increased. Analysis of SA- or SAR-deficient mutants has uncovered how SA and NHP are biosynthesized. The completion of the SA and NHP biosynthetic pathways in Arabidopsis allowed better understanding of how they are regulated. In this review, we discuss recent progress

Plant A20/AN1 protein serves as the important hub to mediate antiviral immunity.

Salicylic acid (SA) is a key phytohormone that mediates a broad spectrum of resistance against a diverse range of viruses; however, the downstream pathway of SA governed antiviral immune response remains largely to be explored. Here, we identified an orchid protein containing A20 and AN1 zinc finger domains, designated Pha13. Pha13 is up-regulated upon virus infection, and the transgenic monocot orchid and dicot Arabidopsis overexpressing orchid Pha13 conferred greater resistance to different viruses. In addition, our data showed that Arabidopsis homolog of Pha13, AtSAP5, is also involved in v

Free radical-mediated systemic immunity in plants.

Systemic acquired resistance (SAR) is a form of defense that protects plants against a broad-spectrum of secondary infections by related or unrelated pathogens. SAR related research has witnessed considerable progress in recent years and a number of chemical signals and proteins contributing to SAR have been identified. All of these diverse constituents share their requirement for the phytohormone salicylic acid, an essential downstream component of the SAR pathway. However, recent work demonstrating the essential parallel functioning of nitric oxide (NO)-derived and reactive oxygen species (R

Defensins--components of the innate immune system in plants.

Plant defensins are small (c.a. 5 kDa), basic, cysteine-rich proteins with antimicrobial activities. They are ubiquitous in plants and form part of the innate immunity arsenal. Plant defensins are encoded by small multigene families and are expressed in various plant tissues, but are best characterized in seeds. They are typically produced as preproteins, however, a small subset are produced as larger precursors with C-terminal prodomains. To date, the three-dimensional solution structures of seven seed- and two floral-derived defensins have been elucidated by (1)H-NMR spectroscopy. Despite li

The plant immune system: From discovery to deployment.

Plant diseases cause famines, drive human migration, and present challenges to agricultural sustainability as pathogen ranges shift under climate change. Plant breeders discovered Mendelian genetic loci conferring disease resistance to specific pathogen isolates over 100 years ago. Subsequent breeding for disease resistance underpins modern agriculture and, along with the emergence and focus on model plants for genetics and genomics research, has provided rich resources for molecular biological exploration over the last 50 years. These studies led to the identification of extracellular and int

Perception of the plant immune signal salicylic acid.

Salicylic acid (SA) plays a central role in plant innate immunity. The diverse functions of this simple phenolic compound suggest that plants may have multiple SA receptors. Several SA-binding proteins have been identified using biochemical approaches. However, genetic evidence supporting that they are the bona fide SA receptors has not been forthcoming. Mutant screens revealed that NPR1 is a master regulator of SA-mediated responses. Although NPR1 cannot bind SA in a conventional ligand-binding assay, its homologs NPR3 and NPR4 bind SA and function as SA receptors. During pathogen challenge,

Immunization with plant-based vaccine expressing Toxoplasma gondii SAG1 fused to plant HSP90 elicits protective immune response in lambs.

Toxoplasma gondii is a protozoan parasite causing toxoplasmosis, a principal concern for public health and livestock industries. Effective vaccination strategies are crucial for controlling toxoplasmosis, particularly in the lamb, which are significant reservoirs of T. gondii. In addition, ovine toxoplasmosis also causes economic losses due to abortions and reproductive complications. In this study, we evaluated two immunization strategies to elucidate the immune protective potential of T. gondi major surface protein SAG1 fused to the plant heat shock proteins 90-kDa (pHsp90) adjuvant against

A NAC triad modulates plant immunity by negatively regulating N-hydroxy pipecolic acid biosynthesis.

N-hydroxy pipecolic acid (NHP) plays an important role in plant immunity. In contrast to its biosynthesis, our current knowledge with respect to the transcriptional regulation of the NHP pathway is limited. This study commences with the engineering of Arabidopsis plants that constitutively produce high NHP levels and display enhanced immunity. Label-free proteomics reveals a NAC-type transcription factor (NAC90) that is strongly induced in these plants. We find that NAC90 is a target gene of SAR DEFICIENT 1 (SARD1) and induced by pathogen, salicylic acid (SA), and NHP. NAC90 knockout mutants e

Phytophthora infestans RXLR effector PITG20303 targets a potato MKK1 protein to suppress plant immunity.

Pathogens secret a plethora of effectors into the host cell to modulate plant immunity. Analysing the role of effectors in altering the function of their host target proteins will reveal critical components of the plant immune system. Here we show that Phytophthora infestans RXLR effector PITG20303, a virulent variant of AVRblb2 (PITG20300) that escapes recognition by the resistance protein Rpi-blb2, suppresses PAMP-triggered immunity (PTI) and promotes pathogen colonization by targeting and stabilizing a potato MAPK cascade protein, StMKK1. Both PITG20300 and PITG20303 target StMKK1, as confi

The CALMODULIN-BINDING PROTEIN60 family includes both negative and positive regulators of plant immunity.

Two members of the eight-member CALMODULIN-BINDING PROTEIN60 (CBP60) gene family, CBP60g and SYSTEMIC ACQUIRED RESISTANCE DEFICIENT1 (SARD1), encode positive regulators of plant immunity that promote the production of salicylic acid (SA) and affect the expression of SA-dependent and SA-independent defense genes. Here, we investigated the other six family members in Arabidopsis (Arabidopsis thaliana). Only cbp60a mutations affected growth of the bacterial pathogen Pseudomonas syringae pv maculicola ES4326. In contrast to cbp60g and sard1 mutations, cbp60a mutations reduced pathogen growth, indi

Salicylic acid-dependent and -independent impact of an RNA-binding protein on plant immunity.

Plants overexpressing the RNA-binding protein AtGRP7 (AtGRP7-ox plants) constitutively express the PR-1 (PATHOGENESIS-RELATED-1), PR-2 and PR-5 transcripts associated with salicylic acid (SA)-mediated immunity and show enhanced resistance against Pseudomonas syringae pv. tomato (Pto) DC3000. Here, we investigated whether the function of AtGRP7 in plant immunity depends on SA. Endogenous SA was elevated fivefold in AtGRP7-ox plants. The elevated PR-1, PR-2 and PR-5 levels were eliminated upon expression of the salicylate hydroxylase nahG in AtGRP7-ox plants and elevated PR-1 levels were suppres

A Ralstonia solanacearum effector RipAD perturbs aquaporin NbPIP oligomeric status to suppress plant immunity.

Bacterial wilt caused by Ralstonia solanacearum is a devastating disease that affects a wide range of crops, including tobacco. The pathogen promotes infection by secreting type III effector (T3E) proteins into host plants to facilitate pathogenicity; however, the molecular mechanisms for many of these effectors remain elusive. Here, we characterize RipAD, a conserved T3E that significantly contributes to R. solanacearum virulence in Nicotiana tabacum and Nicotiana benthamiana. We demonstrate that RipAD dampens early immune responses, including pathogen-associated molecular pattern-triggered r
