Impact of carboxymethyl cellulose on mucin-tea polyphenol interactions: insights into protein-polysaccharide-polyphenol ternary systems.

Polysaccharides and polyphenols frequently coexist in food systems and are consumed simultaneously, yet how these interactions influence polyphenol behavior in the gastrointestinal tract remains insufficiently understood. In this study, the effects of carboxymethyl cellulose (CMC) with different concentrations, molecular weights and degrees of substitution on mucin-tea polyphenol (TP) interactions were systematically investigated. Turbidity and zeta-potential measurements indicated that the addition of CMC reduced aggregation of mucin-TP complexes and increased the negative surface charge. Flu

Pancreatic Lipase Inhibition by Polyphenols in Persimmon Leaf Tea.

Persimmon (Diospyros kaki) leaf tea is a traditional beverage rich in polyphenols and has been associated with various health benefits; however, its potential to inhibit dietary lipid digestion remains to be fully elucidated. This study evaluated the pancreatic lipase-inhibitory effect of hot-water extracts of persimmon leaves (persimmon leaf tea) using both a synthetic substrate (p-nitrophenyl palmitate, pNPP) and a physiologically relevant triacylglycerol substrate in an oil-in-water emulsion system. Persimmon leaf tea exhibited concentration-dependent inhibition in both assays, indicating l

Mechanism of PRRSV infection and antiviral role of polyphenols.

Porcine reproductive and respiratory syndrome (PRRS) is associated with the endemic outbreak of fever, anorexia, and abortion in pregnant sows, resulting in an enormous economic impact on the global swine industry. Current mainstream prophylactic agents and therapies have been developed to prevent PRRSV infection; however, they have limited efficacy. Therefore, there is an urgent need to develop novel antiviral strategies to prevent PRRSV infection and transmission. The identification of new PRRSV entry mediators, such as MYH9 and HSPA8; viral apoptotic mimicry; and TIM-induced macropinocytosi

Targeting Oxidative Stress with Polyphenols to Fight Liver Diseases.

Reactive oxygen species (ROS) are important second messengers in many metabolic processes and signaling pathways. Disruption of the balance between ROS generation and antioxidant defenses results in the overproduction of ROS and subsequent oxidative damage to biomolecules and cellular components that disturb cellular function. Oxidative stress contributes to the initiation and progression of many liver pathologies such as ischemia-reperfusion injury (LIRI), non-alcoholic fatty liver disease (NAFLD), and hepatocellular carcinoma (HCC). Therefore, controlling ROS production is an attractive ther

Polyphenols as Potential Metal Chelation Compounds Against Alzheimer's Disease.

Alzheimer's disease (AD) is the most common neurodegenerative disease affecting more than 50 million people worldwide. The pathology of this multifactorial disease is primarily characterized by the formation of amyloid-β (Aβ) aggregates; however, other etiological factors including metal dyshomeostasis, specifically copper (Cu), zinc (Zn), and iron (Fe), play critical role in disease progression. Because these transition metal ions are important for cellular function, their imbalance can cause oxidative stress that leads to cellular death and eventual cognitive decay. Importantly, these transi

Metal Complexation Mechanisms of Polyphenols Associated to Alzheimer's Disease.

Polyphenols are a class of compounds, produced by plants, which share the ability to act as potent antioxidants. First investigations on polyphenols' antioxidant activity are dated almost twenty years ago when their relationship and implication with the prevention and treatment of cancer was proposed for the first time. Later, in the early 2000s, the neuroprotective effects of several polyphenols were demonstrated. Nowadays, the benefits of a plethora of polyphenols have been studied and their ameliorating effects in several disease conditions, like cancer, cardiac and neuronal diseases are wi

Dietary Polyphenols: A Multifactorial Strategy to Target Alzheimer's Disease.

Ageing is an inevitable fundamental process for people and is their greatest risk factor for neurodegenerative disease. The ageing processes bring changes in cells that can drive the organisms to experience loss of nutrient sensing, disrupted cellular functions, increased oxidative stress, loss of cellular homeostasis, genomic instability, accumulation of misfolded protein, impaired cellular defenses and telomere shortening. Perturbation of these vital cellular processes in neuronal cells can lead to life threatening neurological disorders like Alzheimer's Disease, Parkinson's Disease, Hunting

Polyphenols journey through blood-brain barrier towards neuronal protection.

Age-related complications such as neurodegenerative disorders are increasing and remain cureless. The possibility of altering the progression or the development of these multifactorial diseases through diet is an emerging and attractive approach with increasing experimental support. We examined the potential of known bioavailable phenolic sulfates, arising from colonic metabolism of berries, to influence hallmarks of neurodegenerative processes. In silico predictions and in vitro transport studies across blood-brain barrier (BBB) endothelial cells, at circulating concentrations, provided evide

Effects of polyphenols from grape seeds on renal lithiasis.

Nephrolithiasis is a complex disease that results from a combination of factors related to both urine composition and kidney morphoanatomy. Development of calcium oxalate monohydrate papillary calculi is linked to initial subepithelial calcification of renal papilla. Progressive tissue calcification depends on preexisting injury and involves reactive oxygen species. Many plant extracts that protect against oxidative stress manifest antilithiasic activity. Our study focused on determining the effects of polyphenols on a lithiasis rat model. Rats were pretreated with polyphenols and grape seed e

Natural Polyphenols Inhibit Lysine-Specific Demethylase-1 in vitro.

Natural polyphenols, such as resveratrol, have beneficial functions on major human diseases such as cancer, diabetes, and cardiovascular disease. Besides acting as antioxidants, some of these polyphenols can also target proteins to modulate specific biological pathways. The lysine-specific histone demethylase LSD1 plays important roles in cell growth, differentiation and nutrient metabolism. Here, we studied the effect of natural polyphenols resveratrol, curcumin, quercetin and analogs on LSD1. Using in vitro LSD1 enzymatic assays, we show that resveratrol, curcumin and quercetin displayed a p

Polyphenolic dendrimers as carriers of anticancer siRNA.

Dendrimers have emerged as an important group of nanoparticles to transport drugs, DNA, or RNA into target cells in cancer and other diseases. Various functional modifications can be imposed on dendrimers to increase the efficacy and specificity in delivering their cargo to the target cells and decrease their toxicity. In the present work, we evaluated the potential of carbosilane polyphenolic dendrimers modified with caffeic acid (CA) and polyethylene glycol (PEG) to deliver proapoptotic Mcl-1 and Bcl-2 siRNAs to A549 cancer cells. Dendrimers formed stable complexes with siRNAs as assessed by

Polyphenols as next-generation prebiotics targeting intestinal mucosal tolerance.

Polyphenols, long recognized for their antioxidant and anti-inflammatory properties, are widely consumed as dietary components or as constituents of functional foods and nutraceuticals. Emerging evidence, however, supports a paradigm shift beyond a purely redox-centered perspective, repositioning polyphenols as functionally relevant prebiotic phytochemicals with significant immunomodulatory capacity. This review frames polyphenols as next-generation prebiotics, emphasizing their capacity to act through coordinated, multi-layered mechanisms that extend beyond the stimulation of intestinal micro

Role and mechanisms of plant polyphenols in ovarian aging.

The ovary is a vital organ in female animals, playing a crucial role in regulating reproductive and endocrine functions. Ovarian aging is characterized by declining oocyte quality and ovarian reserve quantity, ultimately leading to infertility and increased risk of age-related diseases. Oxidative stress, inflammation, endocrine disorders, and ovarian microenvironmental disruption are central mechanisms contributing to ovarian aging. Despite the growing incidence of delayed childbearing, there are limited effective interventions to preserve ovarian function. Plant polyphenols, a diverse group o

Site-specific impact of polyphenols on the gastrointestinal microbiome.

The gastrointestinal (GI) tract hosts a diverse microbiota composed of trillions of microorganisms that play crucial roles in maintaining human homeostasis, health, and overall well-being. Dietary polyphenols, primarily found in edible plants, exhibit intricate interactions with the GI microbiota. These polyphenols undergo biotransformation by microbial activity and can modulate the growth of microorganisms, either promoting or inhibiting their proliferation. The composition of microbes in different segments of the GI tract is highly variable and directly influences polyphenols' metabolism. Co
