OBJECTIVE: To study the case series for intussusception associated with the vaccination of rotavirus vaccine in children.
METHODS: The study of spontaneous adverse event monitoring such as intussusception due to rotavirus vaccine was carried out from the year 2011 through 2015. The individual case safety reports (ICSRs) of this event were collated, assessed and recorded as per the requirement of Suspected Adverse Drug Reactions Reporting form of Pharmacovigilance Programme of India (PvPI).
RESULTS: In the present study, 10 ICSRs of intussussception due to rotavirus vaccine were reported to P

Live attenuated oral rotavirus vaccines were tested for proof-of-concept in the early 1980s, the first vaccine (RotaShield, Wyeth) was introduced in 1998 but was subsequently withdrawn because of association with intussusception, and the two currently licensed vaccine (Rotarix, GlaxoSmithKline, and RotaTeq, Merck) were introduced in 2006. Before licensure both vaccines were extensively tested for safety (for intussusception) and efficacy in trials comprising in over 60,000 infants each. Rotarix is a single-strain human rotavirus vaccine (RV1) and RotaTeq is a combination of five bovine-human r…

The important burden of rotavirus infections in infants largely justifies vaccine prevention. Two attenuated oral vaccines were licensed in Europe in 2006 and have proven effective against severe rotavirus gastroenteritis in infants, yet few countries have implemented vaccination. This hesitation may be related to a very low risk of intussusception in vaccinees, but is mainly due to national differences in cost-effectiveness.

Single rotavirus infections have been reported to protect humans against subsequent illnesses caused by both homotypic and heterotypic rotaviruses. On the basis of these observations, a G1P[8] strain of human rotavirus named 89-12 was attenuated by multiple passages in cell culture and was developed into a vaccine candidate (herein referred to as the RIX4414 vaccine). This vaccine is currently licensed in >100 countries worldwide, many of which have a universal recommendation for all infants. The US Food and Drug Administration approved the RIX4414 vaccine for use in the United States in April

Vaccination against rotavirus disease has been a global preventive health priority since rotaviruses were first discovered in the 1970s. The first licensed rotavirus vaccine, RRV-TV, was removed from the market shortly after licensure because of an unexpected serious adverse event, intussusception. A pentavalent rotavirus vaccine, PRV, is the second FDA-licensed rotavirus vaccine and does not signal the risk observed with RRV-TV. The properties and large safety study results of PRV are reviewed here.

In July 1999, CDC recommended that health-care providers and parents postpone use of the rhesus rotavirus vaccine-tetravalent (RRV-TV) (RotaShield, Wyeth Laboratories, Inc., Marietta, Pennsylvania), for infants, at least until November 1999. This action was based on reports to the Vaccine Adverse Event Reporting System of intussusception (a type of bowel obstruction that occurs when the bowel folds in on itself) among 15 infants who received rotavirus vaccine. Also at that time, the manufacturer, in consultation with the Food and Drug Administration, voluntarily ceased further distribution of

By the end of 2017, 32 (68%) of 47 countries in the World Health Organization's African Region had introduced rotavirus vaccine into their national immunization programs, including 27 countries that received financial support from the Gavi, the Vaccine Alliance. Several early introducing African countries previously evaluated the impact, vaccine effectiveness, and/or cost effectiveness of their routine rotavirus vaccination programs and found that rotavirus vaccine was effective and resulted in substantial declines in hospitalizations due to rotavirus. This Special Issue of Vaccine provides ad

The present statement provides information concerning the clinical rotavirus disease and rotavirus vaccines in Canada. Since the implementation of publically funded rotavirus vaccine programs in Canada, increasing evidence has been accumulating globally as to the effectiveness of rotavirus vaccines in the prevention of acute gastroenteritis. Current data estimate vaccine effectiveness to be in the order of 85% for preventing severe disease, including hospitalizations and emergency department visits, when vaccine coverage is high. Also, substantial herd protection in older children has been doc…

The first rotavirus vaccine was associated with a small, albeit statistically significant, risk of intussusception in immunized infants. Before results of new vaccine studies are available, we need to define the real background risk of intussusception in participating countries. A 5-year retrospective study conducted in Panama found an average rate of 1 case per 3300 infants younger than 1 year of age, with substantial yearly variations ranging from 1:2500 to 1:5000.

Rotavirus gastroenteritis continues to cause substantial morbidity and mortality worldwide, despite widespread breastfeeding and use of oral rehydration therapy. This burden of disease indicates that an effective, safe rotavirus vaccine is needed, and in 1998 the first rhesus-human reassortant rotavirus tetravalent vaccine, Rotashield, was licensed in the United States. However, the recommendations for its use were withdrawn in 1999 because of the recognition of an uncommon but serious adverse event, intussusception. A workshop in September 2001 was held to review the subsequent developments a

Worldwide, rotaviruses have been a significant cause of dehydrating gastroenteritis. This contributed to increased infant morbidity and mortality in Jamaica. We enrolled 1804 Jamaican infants in the international randomized, placebo-controlled, pentavalent (G1, G2, G3, G4 and P1) rotavirus vaccine trial. This pentavalent vaccine was found to significantly reduce rotavirus gastroenteritis attributable emergency room visits and hospitalizations, without increasing the rates of intussusception, or other serious adverse events in Jamaican infants. It is recommended that the rotavirus vaccine be in

Rotavirus gastroenteritis primarily affects children younger than 5 years of age and is the leading cause of diarrhea-related hospitalizations worldwide. The substantial morbidity associated with this disease and the major burden on healthcare resources underscore the need for an effective vaccine. Two recently developed vaccines (RotaTeq [rotavirus vaccine, live, oral, pentavalent], and Rotarix [rotavirus vaccine, live]) share some characteristics of an ideal rotavirus vaccine. High efficacy, excellent tolerability, and no increased risk of intussusception were shown in separate clinical tria
