Preparation of Highly Functionalized ortho-Naphthoquinone Methides and Their Subsequent Intermolecular [4 + 2]-Cycloadditions with Olefins.

Many natural products isolated from Rubiaceae plants, such as mollugin and rubioncolin B, contain 1-hydroxy-2-methoxycarbonylnaphthohydropyran skeletons. To access this framework, we developed a novel method for the preparation of highly functionalized ortho-naphthoquinone methides (o-NQMs) and subsequent intermolecular [4 + 2]-cycloaddition reactions with olefins. o-NQM precursors were prepared via Hauser-Kraus annulation of cyanophthalides with an α,β-unsaturated ester. Treatment of these precursors with a Lewis acid generated the corresponding o-NQMs, which underwent intermolecular [4 + 2]-…

[Chemical studies on the analgesic indole alkaloids from the traditional medicine (Mitragyna speciosa) used for opium substitute].

The leaves of a tropical plant, Mitragyna speciosa Korth. (Rubiaceae), have been traditionally used as a substitute for opium. By phytochemical studies on the constituents of the plant growing in Thailand as well as in Malaysia, several 9-methoxy-Corynanthe-type monoterpenoid indole alkaloids including new natural products were isolated. The structures of these new compounds were elucidated by the modern spectroscopic methods and/or chiral-total syntheses. The chiral total synthesis of (-)-mitragynine, a major component of this plant, was achieved. Potent opioid agonistic properties of mitragy…

Enzymatic dehydrogenation and flavin-mediated photocatalytic skeletal rearrangement of strictosamide, a key intermediate in camptothecin biosynthesis.

Camptothecin is a valuable monoterpene indole alkaloid (MIA) with antitumor activity, and strictosamide represents the first specialized intermediate in camptothecin biosynthesis. The proposed camptothecin biosynthetic pathway involves several oxidation-reduction reactions, while there have been no reports on the involvement of 2-oxoglutarate-dependent dioxygenases (2OGDs) in Ophiorrhiza pumila. In this study, the genome-wide identification of 2OGDs was performed in O. pumila, and a total of 140 2OGD genes were identified. Functional characterization revealed that five Op2OGDs catalyze the deh

Neuropharmacological effects of extracts from Sickingia williamsii.

Sickingia williamsii Standl. (Rubiaceae) is used in Peruvian folk medicine for its analgesic and anti-inflammatory activity. In this study we have examined the pharmacological profiles of petroleum ether, chloroform, chloroform-methanol (9:1) and methanol extracts of the tree. Both chloroform-methanol (250, 500 or 750 mg kg-1, p.o.) and methanol (125, 250 or 500 mg kg-1, p.o.) extracts significantly reduced both the locomotor activity and motor co-ordination of mice; they also prolonged sleep induced by pentobarbital, although no significant cataleptic response was observed. These extracts did…
