The ultrastructure of sensory nerve endings in the human knee joint capsule.

The ultrastructure of sensory nerve endings in the human knee joint capsule was studied. Three types of nerve endings were found: free nerve endings (FNE), Ruffini corpuscles and Pacini corpuscles. In the joint capsule, FNE are located below the synovial layer and within the fibrous layer near blood vessels. These nerve terminals derive from myelinated A delta-fibres or from unmyelinated C-fibres. Their structure is almost identical to FNE in human hairy and non-hairy skin. Ruffini corpuscles are present within the fibrous layer and the ligaments of the capsule in three variations: small Ruffi

Calretinin immunoreactivity in the prenatally developing olfactory systems of the tree shrew Tupaia belangeri.

The prenatal patterns of calretinin immunoreactivity were studied in the olfactory systems of Tupaia belangeri. We investigated the peripheral and primary central parts of the vomeronasal system and of the main olfactory system from the 19th to the 43rd (last) day of gestation and compared the findings with the known calretinin immunoreactivity patterns in adult T. belangeri and the published data on other mammals. The onset of calretinin immunoreactivity was noted in the main olfactory system on the 23rd day of gestation and, in the vomeronasal system, on the 25th day of gestation: single pre

Distinctive glial cells in the dorsal root ganglion: their morphology and functions.

Satellite glial cells in the dorsal root ganglion are integral to the biology of sensory neurons. This review explores their unique fine structures, as well as their roles in pain signaling and neuronal differentiation. Satellite glial cells exhibit remarkable plasticity, including stem cell-like properties and the ability to influence neuronal morphology and function. Less-studied glial types, such as axonic satellite glial cells and newly identified glial populations, also offer insights into glial cell diversity and specialization. By focusing on the cellular and molecular mechanisms underl

Mast cell-sensory neuron crosstalk in allergic diseases.

Mast cells (MCs) are tissue-resident immune cells, well-positioned at the host-environment interface for detecting external antigens and playing a critical role in mobilizing innate and adaptive immune responses. Sensory neurons are afferent neurons innervating most areas of the body but especially in the periphery, where they sense external and internal signals and relay information to the brain. The significance of MC-sensory neuron communication is now increasingly becoming recognized, especially because both cell types are in close physical proximity at the host-environment interface and a

Morphological relations between the receptor neurons, sustentacular cells and Schwann cells in the olfactory mucosa of the salamander.

The morphological characteristics of olfactory receptor neurons and their relations to the sustentacular cells and to the sheath cells of Schwann in the olfactory mucosa of the salamander (Ambystoma maculatum et tigrinum) were studied using a modification of the rapid Golgi technique. The bipolar receptor neurons had a fusiform-shaped cell body whose apical pole gave rise to a surface-reaching dendrite and a basal pole which gave rise to an axon. The length and width of the dendrite, although variable, were positively correlated with the relative depth at which the cell body was located in the

Parallel processing of afferent olfactory sensory information.

KEY POINTS: The functional synaptic connectivity between olfactory receptor neurons and principal cells within the olfactory bulb is not well understood. One view suggests that mitral cells, the primary output neuron of the olfactory bulb, are solely activated by feedforward excitation. Using focal, single glomerular stimulation, we demonstrate that mitral cells receive direct, monosynaptic input from olfactory receptor neurons. Compared to external tufted cells, mitral cells have a prolonged afferent-evoked EPSC, which serves to amplify the synaptic input. The properties of presynaptic glutam

Bradykinin receptor expression and bradykinin-mediated sensitization of human sensory neurons.

Bradykinin is a peptide implicated in inflammatory pain in both humans and rodents. In rodent sensory neurons, activation of B1 and B2 bradykinin receptors induces neuronal hyperexcitability. Recent evidence suggests that human and rodent dorsal root ganglia (DRG), which contain the cell bodies of sensory neurons, differ in the expression and function of key GPCRs and ion channels; whether bradykinin receptor expression and function are conserved across species has not been studied in depth. In this study, we used human DRG tissue from organ donors to provide a detailed characterization of bra

Pancreatic β-Cells Communicate With Vagal Sensory Neurons.

BACKGROUND AND AIMS: Destroying visceral sensory nerves impacts pancreatic islet function, glucose metabolism, and diabetes onset, but how islet endocrine cells interact with sensory neurons has not been studied. METHODS: We characterized the anatomical pattern of pancreatic sensory innervation by combining viral tracing, immunohistochemistry, and reporter mouse models. To assess the functional interactions of β-cells with vagal sensory neurons, we recorded Ca2+ responses in individual nodose neurons in vivo while selectively stimulating β-cells with chemogenetic and pharmacologic approaches.

Distribution of alpha-synuclein in the rat cranial sensory ganglia, and oro-cervical regions.

BACKGROUND: Alpha-synuclein (Syn), an unfolded soluble cytosolic protein, is known as a disease-associated protein in the brain. However, little is known about distribution of this protein in the peripheral nervous system. In this study, expression of Syn was investigated in the sensory ganglia of the cranial nerves V, IX and X. METHODS: To analyze distribution of Syn and its co-expression with calcitonin gene-related peptide (CGRP) or the transient receptor potential cation channel subfamily V member 1 (TRPV1), immunohistochemical techniques were used in the rat cranial sensory ganglia and t

Sensory representation and detection mechanisms of gut osmolality change.

Ingested food and water stimulate sensory systems in the oropharyngeal and gastrointestinal areas before absorption1,2. These sensory signals modulate brain appetite circuits in a feed-forward manner3-5. Emerging evidence suggests that osmolality sensing in the gut rapidly inhibits thirst neurons upon water intake. Nevertheless, it remains unclear how peripheral sensory neurons detect visceral osmolality changes, and how they modulate thirst. Here we use optical and electrical recording combined with genetic approaches to visualize osmolality responses from sensory ganglion neurons. Gut hypoto

Selectively Imaging Cranial Sensory Ganglion Neurons Using AAV-PHP.S.

Because of their ease of use, adeno-associated viruses (AAVs) are indispensable tools for much of neuroscience. Yet AAVs have been used relatively little to study the identities and connectivity of peripheral sensory neurons, principally because methods to selectively target peripheral neurons have been limited. The introduction of the AAV-PHP.S capsid with enhanced tropism for peripheral neurons (Chan et al., 2017) offered a solution, which we further elaborate here. Using AAV-PHP.S with GFP or mScarlet fluorescent proteins, we show that the mouse sensory ganglia for cranial nerves V, VII, IX

Cough frequency and cough-receptor sensitivity are increased in man at altitude.

1. Travellers to high altitude often complain of paroxysmal cough, which has not been previously investigated. We recorded overnight cough frequency and cough-receptor sensitivity to inhaled citric acid in a group of climbers travelling to 5300 m or higher. 2. Cough frequency, monitored in ten subjects, increased from a median of 0 coughs at sea level (range 0-1) to 5 coughs at 5000 m (range 0-13) and to over 60 coughs in subjects ascending to 7000 m. Citric acid cough threshold, measured in 42 subjects, was unchanged on arrival at 5300 m compared with sea level (geometric mean difference 1.26

The characteristics of the electro-olfactogram (EOG): its loss and recovery following olfactory nerve section in rainbow trout (Salmo gairdneri).

Electro-olfactograms (EOGs) were recorded from both sensory and non-sensory epithelia on the olfactory lamellae and from other areas within the rosette of rainbow trout (Salmo gairdneri). The trout EOG induced by amino acids was a monophasic negative voltage composed of a phasic component which declined to a steady level (tonic component) that was maintained throughout stimulus duration. The time period (2 min) for complete recovery of the EOG was obtained by applying two identical stimuli (10(-5) mol/liter L-serine for 5 s) successively at increasing intervals. EOG response to amino acids inc

Distribution of P2X3 receptors in the rat trigeminal ganglion after inferior alveolar nerve injury.

The ATP-gated cation channel receptor P2X3 is associated with nociceptive primary sensory neurons. We have, using immunohistochemistry, examined the expression of P2X3 in rat trigeminal ganglia 4-22 days after ligation/section or chronic constriction of the mandibular inferior alveolar nerve. In the normal trigeminal ganglion the anti-P2X3 receptor antibody labeled 37-58% of all neurons. Double labeling demonstrated that about 70-95% of the small neurons that bind the isolectin I-B4 displayed P2X3-immunoreactivity, and that about 40% of larger RT97-positive nerve cells were P2X3 receptor-immun

A capsaicin-receptor homologue with a high threshold for noxious heat.

Pain-producing heat is detected by several classes of nociceptive sensory neuron that differ in their thermal response thresholds. The cloned capsaicin receptor, also known as the vanilloid receptor subtype 1 (VR1), is a heat-gated ion channel that has been proposed to mediate responses of small-diameter sensory neurons to moderate (43 degrees C) thermal stimuli. VR1 is also activated by protons, indicating that it may participate in the detection of noxious thermal and chemical stimuli in vivo. Here we identify a structurally related receptor, VRL-1, that does not respond to capsaicin, acid o

Do axon reflexes exist within the lung?

Circumstantial evidence is given in support of the hypothesis that vagal lung receptors are involved in local axon reflexes. All the components of such mechanisms are present, and teleologically it would make sense for them to exist. It is suggested that rapidly-adapting or irritant receptors produce local muscle relaxation as well as the centrally-mediated reflex bronchoconstriction in other parts of the lung. Conversely, it is suggested that slowly-adapting pulmonary stretch receptors produce the opposite effect - local bronchoconstriction and centrally-mediated reflex bronchodilatation. The
