Determining Sex in Xenopus laevis Tadpoles.

Xenopus laevis has a ZZ/ZW type sex determination system, and a PCR-based method was developed in 2007 to distinguish ZZ males from ZW females. As X. laevis can regenerate tail tissue as tadpoles, this method can be used to determine sex from DNA extracted from tadpole tail clips, which enables accurate identification of sex. Male X. laevis reach sexual maturity faster than females, so we routinely select male tadpoles for establishing transgenic lines. It may also be useful to stratify behavioral or drug testing results by biological sex. Here, I present a quick and simple multiplex PCR-based

True hermaphroditism and mixed gonadal dysgenesis in young children: a clinicopathologic study of 10 cases.

True hermaphroditism (TH) refers to individuals who have both unequivocal ovarian tissue and testicular elements regardless of their karyotypes; whereas mixed gonadal dysgenesis (MGD) refers to individuals who usually have a differentiated gonad on one side and a streak gonad or streak testis on the other side. A differential diagnosis between the TH and MGD has important clinical implications for gender assignment and the decision for early gonadectomy; however, variable clinical and histological features frequently lead to the confusion of TH with MGD. We reviewed the clinicopathological fea

XX-male syndrome bearing the sex-determining region Y.

The case of a 25-year-old man who presented for evaluation of infertility is described. The physical examination revealed testicular atrophy without gynecomastia. Repeated seminal analyses showed azoospermia, and serum hormonal levels suggested a state of a hypergonadotropic hypogonadism. Chromosomal analysis demonstrated 46XX. Polymerase chain reaction revealed the existence of a sex-determining region Y. The etiology of this rare sex reversal syndrome is discussed and cases reported in Japan are reviewed.

Sex Determination and Differentiation in Teleost: Roles of Genetics, Environment, and Brain.

The fish reproductive system is a complex biological system. Nonetheless, reproductive organ development is conserved, which starts with sex determination and then sex differentiation. The sex of a teleost is determined and differentiated from bipotential primordium by genetics, environmental factors, or both. These two processes are species-specific. There are several prominent genes and environmental factors involved during sex determination and differentiation. At the cellular level, most of the sex-determining genes suppress the female pathway. For environmental factors, there are temperat

Genomic predisposition is associated with the direction of sex chromosome evolution.

Sex chromosome evolution is among the most dynamic genomic innovations in vertebrates, yet why some lineages evolve XY while others evolve ZW remains unclear. Using chromosome-level genomes from 19 geckos, we found that gecko sex chromosomes originated independently from 16 ancestral chromosomes. Their origins are nonrandom in both time and direction. Five of 11 datable origins coincide near the Middle Miocene Climatic Transition (~10 million years ago). Ancestral gene content is associated with the direction of evolution, with testis-enriched regions tending to evolve into ZW and testis-deple

Gonadal sex determination in vertebrates: rethinking established mechanisms.

Sex determination and differentiation are fundamental processes that are not only essential for fertility but also influence the development of many other organs, and hence, are important for species diversity and survival. In mammals, sex is determined by the inheritance of an X or a Y chromosome from the father. The Y chromosome harbours the testis-determining gene SRY, and it has long been thought that its absence is sufficient for ovarian development. Consequently, the ovarian pathway has been treated as a default pathway, in the sense that ovaries do not have or need a female-determining

A Conserved Somatic Sex Determination Cascade Instructs Trait-Specific Sexual Dimorphism in Horned Dung Beetles.

Sex-specific trait expression represents a striking dimension of morphological variation within and across species. The mechanisms instructing sex-specific organ development have been well studied in a small number of insect model systems, suggesting striking conservation in some parts of the somatic sex determination pathway while hinting at possible evolutionary lability in others. However, further resolution of this phenomenon necessitates additional taxon sampling, particularly in groups in which sexual dimorphisms have undergone significant elaboration and diversification. Here, we functi

Identification and functional verification of Y-chromosome-specific gene typo-gyf in Bactrocera dorsalis.

Genes on the Y chromosome play important roles in male sex determination and development. The identification of Y-chromosome-specific genes not only provides a theoretical basis for the study of male reproductive development, but also offers genetic control targets for agricultural pests. However, Y-chromosome genes are rarely characterized due to their high repeatability and high heterochromatinization, especially in the oriental fruit fly. In this study, 1 011 Y-chromosome-specific candidate sequences were screened from 2 to 4 h Bactrocera dorsalis embryo datasets with the chromosome quotien

Complete Depletion of Primordial Germ Cells Results in Masculinization of Monopterus albus, a Protogynous Hermaphroditic Fish.

Primordial germ cells (PGCs) play an important role in sexual fate determination and gonadal development in gonochoristic fish, such as zebrafish and medaka. However, little is known about the function of PGCs in hermaphroditic fish. Rice field eel (Monopterus albus), a protogynous hermaphroditic fish, is an economically valuable aquaculture species. We eliminated PGCs in rice field eels during embryogenesis via morpholino-mediated knockdown dead end (dnd). The PGCs-depleted gonads developed into testis-like structures with Sertoli cells and Leydig cells. The gene expression pattern of 15-mont

Sex Chromosome Turnover in Bent-Toed Geckos (Cyrtodactylus).

Lizards and snakes (squamates) are known for their varied sex determining systems, and gecko lizards are especially diverse, having evolved sex chromosomes independently multiple times. While sex chromosomes frequently turnover among gecko genera, intrageneric turnovers are known only from Gekko and Hemidactylus. Here, we used RADseq to identify sex-specific markers in two species of Burmese bent-toed geckos. We uncovered XX/XY sex chromosomes in Cyrtodactylus chaunghanakwaensis and ZZ/ZW sex chromosomes in Cyrtodactylus pharbaungensis. This is the third instance of intrageneric turnover of se

Sex chromosome and sex locus characterization in goldfish, Carassius auratus (Linnaeus, 1758).

BACKGROUND: Goldfish is an important model for various areas of research, including neural development and behavior and a species of significant importance in aquaculture, especially as an ornamental species. It has a male heterogametic (XX/XY) sex determination system that relies on both genetic and environmental factors, with high temperatures being able to produce female-to-male sex reversal. Little, however, is currently known on the molecular basis of genetic sex determination in this important cyprinid model. Here we used sequencing approaches to better characterize sex determination and

Effect of Prenatal Exposure to Corticosterone on Broad-Snouted Caiman (Caiman latirostris) Hatchlings.

Living organisms face stressful situations and use endocrine mechanisms, such as glucocorticoid production, to maintain homeostasis. Although glucocorticoids are essential for basal function, their secretion increases when organisms face stressful situations. In species with temperature-dependent sex determination (TSD), like Caiman latirostris, an increase in these hormones could also alter the sex ratio. In this study, we applied a dose of exogenous corticosterone (70 ng/egg) at developmental stage 20 to assess its effects on sex determination, incubation period (IP), and morphometric traits

Gonadal transdifferentiation in teleosts during normal and altered physiology-a review.

Teleost species display a diverse range of reproductive strategies, consisting mainly of gonochorism and hermaphroditism, either sequentially or simultaneously. This leads to species-specific sex determination processes. Owing to the vast range of teleost species, sex determination and differentiation mechanisms vary in their sexual patterns; therefore, sexual plasticity in gonadal development is inevitable. Under certain environmental conditions, such as fluctuations in temperature, pH, density, social interaction, and hypoxia, gonadal fate is liable to change through a phenomenon referred to…

An explanation for the prevalence of XY over ZW sex determination in species derived from hermaphroditism.

The many independent transitions from hermaphroditism to separate sexes (dioecy) in flowering plants and some animal clades must often have involved the emergence of a heterogametic sex-determining locus, the basis of XY and ZW sex determination (i.e., male and female heterogamety). Current estimates indicate that XY sex determination is much more frequent than ZW, but the reasons for this asymmetry are unclear. One proposition is that separate sexes evolve through the invasion of sterility mutations at closely linked loci, in which case XY sex determination evolves if the initial male sterili

The Male-Biased Expression of miR-2954 Is Involved in the Male Pathway of Chicken Sex Differentiation.

Many expression data showed miRNAs have a potential function on regulating gonadal differentiation in animals, but their function is rarely studied in vivo, especially in chickens. Using the comprehensive expression profiles analysis, the specific male-biased miR-2954, which is significantly higher expressed in male embryos and gonads at all detected stages, was firstly screened during the early stages of chicken embryogenesis and gonadogenesis. In sex-reversed female gonads treated with aromatase inhibitors, the expression of miR-2954 was increased, which was consistent with the up-regulation

Highly rapid and diverse sex chromosome evolution in the Odorrana frog species complex.

Sex chromosomes in poikilothermal vertebrates are characterized by rapid and diverse evolution at the species or population level. Our previous study revealed that the Taiwanese frog Odorrana swinhoana (2n = 26) has a unique system of multiple sex chromosomes created by three sequential translocations among chromosomes 1, 3, and 7. To reveal the evolutionary history of sex chromosomes in the Odorrana species complex, we first identified the original, homomorphic sex chromosomes, prior to the occurrence of translocations, in the ancestral-type population of O. swinhoana. Then, we extended the i

Sex reversal of pejerrey (Odontesthes bonariensis), a species with temperature-dependent sex determination, in a seminatural environment.

This study examined the changes in sex ratios and sex reversal rates in pejerrey Odontesthes bonariensis that occur with the progression of the spawning season in a seminatural setting. Four groups of hatchery-produced pejerrey larvae were stocked in floating cages in La Salada de Monasterio lake (Pampas region), a natural habitat of this species, and reared from hatching beyond gonadal sex determination with minimum human interference. Cage 1 was stocked at the beginning of the spring spawning season and the other cages were stocked with monthly delays until cage 4 in early summer. The genoty
