Generation and Characterization of Native and Sialic Acid-Deficient IgE.

Efficient characterization of IgE antibodies and their glycan structures is required for understanding their function in allergy and in the emerging AllergoOncology field for antibody immunotherapy. We report the generation, glyco-profiling and functional analysis of native and sialic acid-deficient glyco-engineered human IgE. The antibodies produced from human embryonic kidney cells were purified via a human IgE class-specific affinity matrix and structural integrity was confirmed by SDS-PAGE and size-exclusion chromatography (SEC). Purified IgEs specific for the tumor-associated antigens Cho

Genetic defects in the hexosamine and sialic acid biosynthesis pathway.

BACKGROUND: Congenital disorders of glycosylation are caused by defects in the glycosylation of proteins and lipids. Classically, gene defects with multisystem disease have been identified in the ubiquitously expressed glycosyltransferases required for protein N-glycosylation. An increasing number of defects are being described in sugar supply pathways for protein glycosylation with tissue-restricted clinical symptoms. SCOPE OF REVIEW: In this review, we address the hexosamine and sialic acid biosynthesis pathways in sugar metabolism. GFPT1, PGM3 and GNE are essential for synthesis of nucleot

Sialic acid, homocysteine and CRP: potential markers for dementia.

To investigate whether sialic acid could discriminate between healthy age matched controls and patients with dementias of the Alzheimer's type (AD), and pure vascular dementia (VaD). 27 patients and 51 controls were administered the Mini-Mental State Examination (MMSE) and had blood analyzed for levels of total sialic acid, total homocysteine (tHcy), and C-reactive protein (CRP). Significant differences were found between the mean MMSE scores for patients with dementia compared with controls. Sialic acid levels were significantly higher in patients with AD compared with controls and homocystei

Identification of a novel sialic acid transporter in Haemophilus ducreyi.

Haemophilus ducreyi, the causative agent of chancroid, produces a lipooligosaccharide (LOS) which terminates in N-acetyllactosamine. This glycoform can be further extended by the addition of a single sialic acid residue to the terminal galactose moiety. H. ducreyi does not synthesize sialic acid, which must be acquired from the host during infection or from the culture medium when the bacteria are grown in vitro. However, H. ducreyi does not have genes that are highly homologous to the genes encoding known bacterial sialic acid transporters. In this study, we identified the sialic acid transpo

Sialic acid metabolism's dual function in Haemophilus influenzae.

Many bacterial commensals and pathogens use the sialic acids as carbon and nitrogen sources. In Escherichia coli, the breakdown of these sugars is catalysed by gene products of the nan (Nacylneuraminate) operon; other microorganisms may use a similar catabolic strategy. Despite the known ligand and antirecognition functions of the sialic acids, the contribution of their catabolism to infection or host colonization has never been directly investigated. We addressed these questions with Haemophilus influenzae type b, which metabolizes relatively few carbohydrates, using the infant-rat infection …

A Japanese case of infantile sialic acid storage disease.

We report a 4-year-old Japanese girl with infantile sialic acid storage disease. She presented with failure to thrive, coarse facial features, hepatosplenomegaly, severe mental retardation and spastic quadriplegia. Electron microscopic examination of cultured skin fibroblasts revealed multiple vacuoles and inclusion material representing distended lysosomes, thus suggesting a lysosomal storage disorder. A high concentration of free sialic acid was present in the urine and cultured fibroblasts, but bound sialic acid was not increased. The activity of a variety of lysosomal enzymes was not dimin

Fibronectin and sialic acid levels in human meningiomas and gliomas.

In this study, fibronectin and sialic acid levels have been assayed in human meningiomas and gliomas. The mean fibronectin and sialic acid levels for human meningiomas were 22.01 +/- 9.70 micrograms/mg protein and 19.58 +/- 4.89 micrograms/mg protein, respectively, and for human gliomas were 27.30 +/- 13.70 micrograms/mg protein and 25.67 +/- 11.60 micrograms/mg protein, respectively, versus 9.23 +/- 5.40 micrograms/mg protein and 13.50 +/- 4.30 micrograms/mg protein for normal brain tissues. Fibronectin and sialic acid levels were significantly higher in human meningiomas (P < 0.01 and P < 0.

Erythrocyte sialic acid in human sickle-cell disease.

Membrane-bound erythrocyte surface sialic acid concentrations in sickle-cell human patients (SS) and in control patients with sickle-cell trait (AS) and normal genes (AA) were assayed. The erythrocyte sialic acid concentration was significantly (P less than 0.001) higher in sickle-cell patients than in non-sicklers and although the concentration was higher in AS than AA individuals the difference was not significant (P greater than 0.05). The anaemic state of the SS patients was shown by a significantly (P less than 0.001) lower PCV than in non-sicklers. This finding is discussed in relation t

Erythrocyte surface sialic acid in Ndama and Zebu cattle.

Erythrocyte surface sialic acid concentrations were measured in the trypano-tolerant Ndama and trypano-susceptible Zebu breeds of cattle. All animals were uninfected and had no records of previous exposure to infection by African pathogenic trypanosomes. Ndama adults had significantly (P less than 0.001) higher erythrocyte surface sialic acid concentrations than Ndama calves. Ndama adults and calves had significantly (P less than 0.001) higher erythrocyte surface sialic acid concentrations than Zebu adults. There were no significant differences between the packed cell volume (PCV), haemoglobin

Erythrocyte membrane sialic acid in new-born infants.

Erythrocyte surface sialic acid was measured in 94 full-term new-born infants and in 20 premature infants of different ages, varying from 1 to 16 post-natal days. After the first 3 days of life, the amount of sialic acid, both per cell and per gramme of haemoglobin, was rapidly reduced to 70-80% of the initial value; these lower levels were maintained on the following days. The decrease observed on the 4th day may represent an expression of the mechanism by which a large amount of fetal red blood cells is eliminated from the circulation during the same period. Therefore, the relationship betwe

The SARS-CoV-2 Spike Glycoprotein Directly Binds Exogeneous Sialic Acids: A NMR View.

The interaction of the SARS CoV2 spike glycoprotein with two sialic acid-containing trisaccharides (α2,3 and α2,6 sialyl N-acetyllactosamine) has been demonstrated by NMR. The NMR-based distinction between the signals of those sialic acids in the glycans covalently attached to the spike protein and those belonging to the exogenous α2,3 and α2,6 sialyl N-acetyllactosamine ligands has been achieved by synthesizing uniformly 13 C-labelled trisaccharides at the sialic acid and galactose moieties. STD-1 H,13 C-HSQC NMR experiments elegantly demonstrate the direct interaction of the sialic acid resi…

Determination of sialic acid by the thiobarbituric acid reaction in sweet whey and its fractions.

Determination of sialic acid in sweet whey is useful as the concentration of sialic acid reflects the amount of glycomacropeptide (GMP) present. In this study, the concentration of total sialic acid was determined by the thiobarbituric acid reaction after dialysis of samples in water, and the concentration of GMP sialic acid was estimated by gel chromatography on Sephacryl S-200. Concentrations of total and GMP sialic acid determined in a sweet whey sample prepared from fresh milk were 2.0 and 1.5 microg/mg of dry weight, respectively. Analysis of commercial samples showed that the concentrati

Sperm binding to oviductal epithelial cells in the rat: role of sialic acid residues on the epithelial surface and sialic acid-binding sites on the sperm surface.

The aim of this study was to assess the participation of carbohydrate residues in the adhesion of spermatozoa to the oviductal epithelium in the rat. We first examined, by lectin labeling, the distribution of glycoconjugates in rat oviducts obtained under various hormonal environments. Several classes of glycoconjugates were abundant in the epithelium, and the expression of some of these molecules varied differentially in ampulla and isthmus, along the estrous cycle and with estradiol and progesterone treatment. Proestrous rats were intraoviductally injected with lectins Dolichos biflorus, Ery
