Regulation of intracellular pH in the myocardium; relevance to pathology.

Intracellular pH affects the contractile function of the heart, metabolic reactions, ion exchange and calcium homoeostasis. Numerous studies have concluded that a fall of extracellular pH, by whatever mechanism, causes a fall of contractility by alteration of intracellular pH. Measurement of cytosolic intracellular pH using microelectrodes has confirmed that earlier deduction. Acidosis reduces the slow calcium current and the release of calcium from the sarcoplasmic reticumul but, because the cytosolic calcium does not fall, the major site of action of hydrogen ions appears to be on the calciu

Hypertonic shrinking but not hypotonic swelling increases sodium concentration in rat brain synaptosomes.

Neurotransmitter release is dependent on both calcium and sodium influx. Hypotonic swelling and hypertonic shrinking of neurons evokes calcium-independent exocytosis of neurotransmitters into the synaptic cleft. To date, there are not too much data available on relationship between extracellular osmolarity and sodium concentration in presynaptic endings. In the present study we investigated the effects of hypotonic swelling and hypertonic shrinking on sodium levels, as measured using fluorescent dyes SBFI-AM and Sodium Green in rat brain synaptosomes. Reduction of incubation medium osmolarity

Invited review: Neuroprotective properties of certain beta-adrenoceptor antagonists used for the treatment of glaucoma.

Although it is known that ganglion cell death causes loss of vision in glaucoma, the pathogenesis of the disease is complex, probably involving an initial ischemic insult to the ganglion cell axons and glial cells with the ganglion cell bodies eventually being affected. It may therefore be necessary to blunt many stages in the pathogenesis of the disease to obtain a clinically effective neuroprotective strategy. In animal experiments, one cause of ganglion cell death in ischemia is an overactivation of glutamate receptors and a subsequent rise in intracellular levels of sodium and calcium ions

Hyperglycemia-induced cardiac contractile dysfunction in the diabetic heart.

The development of a diabetic cardiomyopathy is a multifactorial process, and evidence is accumulating that defects in intracellular free calcium concentration [Ca2+]i or its homeostasis are related to impaired mechanical performance of the diabetic heart leading to a reduction in contractile dysfunction. Defects in ryanodine receptor, reduced activity of the sarcoplasmic reticulum calcium pump (SERCA) and, along with reduced activity of the sodium-calcium exchanger (NCX) and alterations in myofilament, collectively cause a calcium imbalance within the diabetic cardiomyocytes. This in turn is …

Re-thinking angina.

William Heberden in 1772 published "some account of the disorder of the breast" which contains the essential elements of angina pectoris as we understand it today. The number of existing cases in the U.S. population today is 6.4 million. Myocardial ischemia manifested by angina pectoris can be either acute or chronic. Patients with chronic stable angina will be the focus of this supplement. The majority of patients are symptomatic but approximately 25% can be asymptomatic. The clinical manifestations of myocardial ischemia generally are chest discomfort, arrhythmias, and LV dysfunction. Myocar

Chronic inhibition of the Na+/H+ - exchanger causes regression of hypertrophy, heart failure, and ionic and electrophysiological remodelling.

BACKGROUND AND PURPOSE: Increased activity of the Na+/H+ -exchanger (NHE-1) in heart failure underlies raised [Na+]i causing disturbances of calcium handling. Inhibition of NHE-1, initiated at the onset of pressure/volume overload, prevents development of hypertrophy, heart failure and remodelling. We hypothesized that chronic inhibition of NHE-1, initiated at a later stage, would induce regression of hypertrophy, heart failure, and ionic and electrophysiological remodelling. EXPERIMENTAL APPROACH: Development of heart failure in rabbits was monitored electrocardiographically and echocardiogr

Role of sodium in mitochondrial membrane depolarization induced by P2X7 receptor activation in submandibular glands.

The effect of ATP on mitochondrial membrane depolarization in rat submandibular glands was investigated. Exposure of the cell suspension to high concentrations of ATP induced a sustained depolarization of mitochondrial membrane. This effect was blocked in the presence of magnesium and reproduced by low concentrations of 2',3'-O-(4-benzoylbenzoyl)adenosine 5'-triphosphate (BzATP), suggesting the implication of the P2X(7) purinergic receptor. This point was confirmed by comparison of the response to ATP by wild-type and P2X(7) knock-out (P2X(7)R(-/-)) mice. Mitochondria took up calcium after ATP

Phosphoinositide hydrolysis induced by depolarization and sodium channel activation in mouse cerebrocortical slices.

Carbachol, a muscarinic receptor agonist and the sodium channel-activating agents, scorpion venom, veratridine, batrachotoxin and aconitine, were shown to stimulate the formation of [3H]inositol phosphates in [3H]inositol-labelled miniprisms, obtained from the cerebral cortex of the mouse. The inositol response to the Na+ channel-activating agents was inhibited by the sodium channel blocker tetrodotoxin (TTX), while the response induced by carbachol was partially resistant to TTX. The response to scorpion venom and the TTX-insensitive portion of the response to carbachol was additive, indicati

Regulation of the cytosolic pH set point for activation of the Na+/H+ antiport in human platelets: the roles of the Na+/Ca2+ exchange, the Na(+)-K(+)-2Cl- cotransport and cellular volume.

To explore further the mechanisms that regulate the Na+/H+ antiport in human platelets, we examined the effect of Na+ pump inhibition by ouabain and K+ removal from the extracellular medium on parameters of this transport system. Treatment with ouabain resulted in increased cytosolic free Ca2+ and Na+, coupled with an alkaline shift in the cytosolic pH set point for the Na+/H+ antiport. Inhibition of the Na+ pump by the removal of K+ from the medium increased the cytosolic Na+ but not the cytosolic Ca2+; yet this treatment also produced a substantial alkaline shift in the cytosolic pH set poin

Role of Mitochondrial Calcium Dysregulation in Alzheimer's Disease Pathogenesis.

Mitochondrial calcium has emerged as a critical player in Alzheimer's disease (AD), closely linked to neuronal dysfunction and cognitive decline seen in patients. Intracellular calcium signaling is essential for processes like synaptic plasticity, neuronal survival, and differentiation. When this balance is disturbed, it can trigger early pathological changes in AD, including the accumulation of amyloid-β (Aβ) peptides and the development of neurofibrillary tangles (NFTs), the hallmark features of the disease. Calcium imbalance in mitochondria disrupts their function, leading to reduced adenos

Pathways for the movements of ions during calcium-free perfusion and the induction of the 'calcium paradox'.

The intracellular sodium content of cardiac cells in fish and amphibia, measured with either an isotope technique or with sodium-sensitive micro-electrodes, rises steeply from around 15 mmol/l in calcium-containing solution to as much as 70 mmol/l, during exposure to a Ca2+-free solution. This increase is associated with the development of spontaneous and prolonged action potentials so that the membrane may stabilise around -20 mV. On reperfusion with calcium-containing medium the membrane repolarises before a strong contracture develops. Inhibition of the Na-pump increases both the sodium gai

Changes in total body calcium and sodium in osteomalacia.

Serial measurements were made of total body calcium and sodium, using whole body neutron activation analysis, in six patients being treated for osteomalacia. All showed a rise in total body calcium ranging from 3 to 30%; this continued for up to 24 months after the start of treatment, though complete healing had apparently occurred after 6 months on the basis of histological examination of biopsy specimens of bone. As the total body calcium rose there was a fall in the non-exchangeable sodium pool in bone.

The Na+/Ca2+ exchanger: molecular structure, functional regulation, and mode-selective modulation.

The Na+/Ca2+ exchanger (NCX) is a key regulator of intracellular Ca2+ homeostasis, mediating bidirectional ion transport in response to electrochemical gradients. NCX plays essential roles in diverse physiological processes, including cardiac contraction, neuronal signaling, and hormone secretion, while its dysregulation is implicated in a wide range of pathophysiological conditions such as heart failure, ischemia, and neurodegenerative diseases. Recent advances in structural biology, particularly cryo-electron microscopy, have provided detailed insights into the molecular architecture of euka

Na+, K(+)-ATPase and Na+/Ca2+ exchange isoforms: physiological and physiopathological relevance.

Different isoforms of the (Na+ + K+)-ATPase are expressed in different cell types in which they contribute to specialized properties. Their biochemistry and physiology are complex. These isozymes vary in their sensitivity to cardiac glycosides and to intracellular Na+ and Ca2+ concentrations. Their functional expression at the membrane level in the different parts of kidney, heart, and brain varies with species and during ontogenesis. In rat heart, at birth and postpartum, there are quantitative and qualitative changes in the expression of the (Na+ + K+)-ATPase and Na+/Ca2+ exchange isoforms.

Flecainide sensitizes conduction to hyponatremia through an ephaptic mechanism.

BACKGROUND: Studies suggest that voltage-gated sodium channel (SC) loss-of-function (LoF), often through the use of SC blockers, such as tricyclic anti-depressants, some recreational drugs, and importantly, class 1c anti-arrhythmics, sensitizes cardiac conduction to hyponatremia. However, the mechanism driving conduction velocity (CV) sensitivity to sodium ion (Na+) concentration ([Na+]) is unknown. We recently demonstrated CV-[Na+] sensitivity in haploinsufficient Scn5a+/- mouse and reduced CV-[Na+] sensitivity when ephaptic coupling (extracellular conduction by electric fields) is also reduc

Activation of β3-adrenergic receptor inhibits ventricular arrhythmia in heart failure through calcium handling.

Ventricular arrhythmia in chronic heart failure (CHF) is considered to be associated with stimulation of β-adrenergic receptors (β-ARs). Three classes of β-ARs have been identified; importantly, distinct from β1 and β2 subtypes, β3-AR could inhibit arrhythmia. Intracellular Ca2+ is considered as a predominant effecter of arrhythmia during heart failure. However, the exact role of β3-AR in arrhythmia and Ca2+ regulation in CHF is not clear yet. Therefore, we studied the effect of BRL37344, a specific β3-AR activator, on CHF-related ventricular arrhythmia and cellular Ca2+ transport. Rabbits wit

Fabrication of patterned calcium cross-linked alginate hydrogel films and coatings through reductive cation exchange.

Calcium cross-linked alginate hydrogels are widely used in targeted drug delivery, tissue engineering, wound treatment, and other biomedical applications. We developed a method for preparing homogeneous alginate hydrogels cross-linked with Ca(2+) cations using reductive cation exchange in homogeneous iron(III) cross-linked alginate hydrogels. Treatment of iron(III) cross-linked alginate hydrogels with calcium salts and sodium ascorbate results in reduction of iron(III) cations to iron(II) that are instantaneously replaced with Ca(2+) cations, producing homogeneous ionically cross-linking hydro

Urinary concentrating defect in mice lacking Epac1 or Epac2.

cAMP is a universal second messenger regulating a plethora of processes in the kidney. Two downstream effectors of cAMP are PKA and exchange protein directly activated by cAMP (Epac), which, unlike PKA, is often linked to elevation of [Ca2+]i. While both Epac isoforms (Epac1 and Epac2) are expressed along the nephron, their relevance in the kidney remains obscure. We combined ratiometric calcium imaging with quantitative immunoblotting, immunofluorescent confocal microscopy, and balance studies in mice lacking Epac1 or Epac2 to determine the role of Epac in renal water-solute handling.
