alpha-Methyltyrosine blocks methylamphetamine-induced degeneration in the rat somatosensory cortex.

Neurochemical and histological studies suggest that methylamphetamine (MA) administered continuously or in high doses is toxic to dopaminergic and serotonergic nerve terminals. Degeneration of the dopaminergic or serotonergic cell bodies themselves has not been reported, however. In the present study, administration of a single 100 mg/kg dose of MA was toxic to a subpopulation of neurons in the somatosensory cortex, an area of the brain which does not contain catecholaminergic or serotonergic cell bodies. This dose of MA also produced a long-lasting depletion of serotonin (5-HT) but not norepi…

Conscious Tactile Perception Depends on Early Processing in Primary Somatosensory Cortex.

The primary somatosensory cortex (S1) has long been implicated in tactile perception, yet its precise role in conscious tactile detection remains uncertain. The current study investigated the causal and time-specific involvement of S1 in tactile detection using single-pulse transcranial magnetic stimulation (spTMS). In two experiments, spTMS was applied over contralateral S1, an active control site (inferior parietal lobe; IPL), or under a sham condition at short (25 and 75 ms; Experiment 1) or longer (130 ms; Experiment 2) intervals following electrotactile stimulation of the finger. Particip…

Bilateral somatosensory cortex disinhibition in complex regional pain syndrome type I.

OBJECTIVE: In a previous study, we found bilateral disinhibition in the motor cortex of patients with complex regional pain syndrome (CRPS). This finding suggests a complex dysfunction of central motor-sensory circuits. The aim of our present study was to assess possible bilateral excitability changes in the somatosensory system of patients with CRPS. METHODS: We measured paired-pulse suppression of somatosensory evoked potentials in 21 patients with unilateral CRPS I involving the hand. Eleven patients with upper limb pain of non-neuropathic origin and 21 healthy subjects served as controls.

Bidirectional axonal plasticity during reorganization of adult rat primary somatosensory cortex.

Cortical sensory maps contain discrete functional subregions that are separated by borders that restrict tangential activity flow. Interestingly, the functional organization of border regions remains labile in adults, changing in an activity-dependent manner. Here, we investigated if axon remodeling contributes to this reorganization. We located the border between the forepaw and lower jaw representation (forepaw/lower jaw border,(1) FP/LJ border) in SI of adult rats, and used a retrograde axonal tracer (cholera toxin subunit B(2), Ctb) to determine if horizontal axonal projections change afte

Reorganization of the hand somatosensory cortex following perinatal unilateral brain injury.

Functional magnetic resonance imaging was used to map the hand somatosensory cortices of nine hemiparetic young adult patients with perinatal unilateral brain injury in the sensorimotor area and five normal subjects. Stimulation of the paretic hand by periodic manual squeezing produced activation in the contralateral hemisphere of three patients and in the ipsilateral hemisphere of three other patients. Paretic hand stimulation produced no activation in either hemisphere of the remaining three patients. Therefore, one-third of the patients demonstrated functional "plasticity" of the brain in t

Caudal Granular Insular Cortex to Somatosensory Cortex I: A Critical Pathway for the Transition of Acute to Chronic Pain.

Allodynia (perceiving touch as painful) is an enduring symptom of neuropathic pain. While acute pain is initiated by afferent signaling from the periphery to spinal cord, pain chronification recruits ongoing activity in supraspinal sites. One such site that has been proposed to be important in pain chronification is the caudal granular insular cortex (CGIC). The present studies of allodynia in response to sciatic nerve injury in male and female rats focus on the role of CGIC in pain chronification by analyzing: circuit-specific mGreenLantern expression to define CGIC-to-somatosensory cortex I …

Limits on plasticity in somatosensory cortex of adult rats: hindlimb cortex is not reactivated after dorsal column section.

1. To better understand the limits and extents of plasticity in sensory systems of adult mammals, we unilaterally sectioned the dorsal funiculus at thoracic levels in nine adult rats to deactivate ascending afferents from the hindpaw and lower body. After postsurgical recovery periods of 3 h to 3 mo, the region of primary somatosensory cortex (S1) representing the limbs and trunk was extensively mapped with microelectrodes. 2. Recording sites were later identified as being within the hindlimb representation and other parts of S1 by relating locations of microlesions to the cytochrome oxidase p…

Obesity differentially effects the somatosensory cortex and striatum of TgF344-AD rats.

Lifestyle choices leading to obesity, hypertension and diabetes in mid-life contribute directly to the risk of late-life Alzheimer's disease … spectrum of AD symptomology. We hypothesized regions primarily composed of gray matter, such as the somatosensory cortex (SSC), would be differentially affected compared to regions primarily composed of white matter, such as the striatum. We found increased myelin and oligodendrocytes in the somatosensory cortex of rats fed the HCHF diet with an absence of neuronal lo …

Neuronal Correlates of Tactile Working Memory in Prefrontal and Vibrissal Somatosensory Cortex.

Tactile working memory engages a broad network of cortical regions in primates. To assess whether the conclusions drawn from primates apply to rodents, we examined the vibrissal primary somatosensory cortex (vS1) and the prelimbic cortex (PL) in a delayed comparison task. Rats compared the speeds of two vibrissal vibrations, stimulus1 and stimulus2, separated by a delay of 2 s. Neuronal firing rates in vS1 and PL encode both stimuli in real time. Across the delay, the stimulus1 representation declines more precipitously in vS1 than in PL. Theta-band local field potential (LFP) coherence betwee…

Maturation of thalamocortical synapses in the somatosensory cortex depends on neocortical AKAP5 expression.

Sensory cortex topographic maps consist of organized arrays of thalamocortical afferents (TCAs) that project into distinct areas of the cortex. Formation of topographic maps in sensory cortices is a prerequisite for functional maturation of the neocortex. Studies have shown that the formation of topographic maps and the maturation of thalamocortical synapses in the somatosensory cortex depend on the cyclic adenosine 5'-monophosphate-(cAMP)-protein kinase A (PKA) signaling pathway. AKAP5 is a scaffold protein (also called AKAP79 in humans or AKAP150 in rodents; AKAP79/150) that serves as a sign…

Connectivity of mouse somatosensory and prefrontal cortex examined with trans-synaptic tracing.

Information processing in neocortical circuits requires integrating inputs over a wide range of spatial scales, from local microcircuits to long-range cortical and subcortical connections. We used rabies virus-based trans-synaptic tracing to analyze the laminar distribution of local and long-range inputs to pyramidal neurons in the mouse barrel cortex and medial prefrontal cortex (mPFC). In barrel cortex, we found substantial inputs from layer 3 (L3) to L6, prevalent translaminar inhibitory inputs, and long-range inputs to L2/3 or L5/6 preferentially from L2/3 or L5/6 of input cortical areas,

Organization of myelin in the mouse somatosensory barrel cortex and the effects of sensory deprivation.

In rodents, the barrel cortex is a specialized area within the somatosensory cortex that processes signals from the mystacial whiskers. We investigated the normal development of myelination in the barrel cortex of mice, as well as the effects of sensory deprivation on this pattern. Deprivation was achieved by trimming the whiskers on one side of the face every other day from birth. In control mice, myelin was not present until postnatal day 14 and did not show prominence until postnatal day 30; adult levels of myelination were reached by the end of the second postnatal month. Unbiased stereolo…

Connectivity of ectopic neurons in the molecular layer of the somatosensory cortex in autoimmune mice.

Approximately 50% of New Zealand Black mice (NZB/BINJ) and 80% of NXSM-D/EiJ mice prenatally develop neocortical layer I ectopias, mostly in somatosensory cortices. These cortical anomalies are similar to those seen in the brains of individuals with dyslexia. Neurofilament staining revealed a radial column of tightly packed fiber bundles in the layers underlying ectopias. This suggested that the connectivity of the ectopic neurons was aberrant. The present study used the tracers 1,1'-dioctadecyl- 3,3,3',3'-tetramethylindo- carbocyanine perchlorate (DiI) and biotinylated dextran amine (BDA) to

Synchrony within Aβ mechanoreceptor subtypes governs signal propagation to primary somatosensory cortex.

Fast-conducting Aβ slowly adapting type I low threshold mechanoreceptors (SAI-LTMRs) and Aβ rapidly adapting type I (RAI-)LTMRs are critical … spike trains with mean rates of 20 Hz in either subtype when monitoring responses in mouse primary somatosensory cortex (S1). Doing so revealed that propagation of steady-state signals from sustained responses of Aβ SAI-LTMRs to neural activity (spiking and local field potential) in S1 is dependent on synchronous activation of multiple Aβ SAI-LTMRs. Asynchronous acti …

Tactile force evokes a biphasic BOLD response in ipsilateral primary somatosensory cortex.

Tactile perception is fundamental to how we engage with and interpret our surroundings. While the contralateral primary somatosensory cortex (S1) is well known to play a role in processing tactile information, lesser understood responses in the ipsilateral hemisphere have been observed. We aim to characterize the blood-oxygen-level-dependent (BOLD) response pattern of ipsilateral S1 to unilateral tactile stimuli. Three stimulus force levels were applied in an event-related design. As expected, we observed an apparent difference in BOLD responses between the contralateral and ipsilateral hemisp…
