Spin trapping agents (Tempol and POBN) protect HepG2 cells overexpressing CYP2E1 against arachidonic acid toxicity.

Polyunsaturated fatty acids such as arachidonic acid were previously shown to be toxic to HepG2 cells expressing CYP2E1 by a mechanism involving oxidative stress and lipid peroxidation. This study investigated the effects of the spin trapping agents Tempol and POBN on the arachidonic acid toxicity. Arachidonic acid caused toxicity and induced lipid peroxidation and mitochondrial membrane damage in cells overexpressing CYP2E1 but had little or no effect in control cells not expressing CYP2E1. The toxicity appeared to be both apoptotic and necrotic in nature. 4-Hydroxy-[2,2,6,6-tetramethylpiperi…

2-2'-Pyridylisatogen, a selective allosteric modulator of P2 receptors, is a spin trapping agent.

2-2'-Pyridylisatogen (PIT) has been reported to be a relatively selective irreversible antagonist of responses to adenosine 5'-triphosphate (ATP) in some smooth muscle preparations and to be an allosteric modulator of responses to ATP at recombinant P2Y receptors from chick brain. PIT is also a potent inhibitor of mitochondrial oxidative phosphorylation. However, the compound has a unique nitrone structure, so PIT was compared with dimethyl-pyrroline-N-oxide (DMPO) as a spin trapping agent for superoxide and hydroxyl radicals using electron spin resonance (ESR). PIT was found to be a potent sp…

PBN spin trapping of free radicals in the reperfusion-injured heart. Limitations for pharmacological investigations.

Post-ischemic reperfusion causes cardiac dysfunction and radical-induced lipid peroxidation (LPO) detectable by ESR spin trapping. This study deals with the applicability of the spin trapping technique to pharmacological investigations during myocardial reperfusion injury. The use of the spin trap phenylbutylnitrone (PBN, 3 mM) in isolated rat hearts demonstrated the release of alkoxyl radicals (aN = 1.39 mT, aHbeta = 0.19 mT) formed particularly within the first 15 min of reperfusion following 30 min of ischemia. The decline of radicals, after 10 min of reperfusion, was accompanied by recover…

Unveiling the mystery of chlorine radical in water by a simple methanol-based spin trapping.

Both hydroxyl radical (•OH) and chlorine radical (Cl•) are recognized as significant reactive species in aquatic and atmospheric sciences. While •OH can be readily detected with the conventional spin trap, 5,5-dimethyl-1-pyrroline N-oxide (DMPO), Cl• is often considered difficult to detect by electron paramagnetic resonance (EPR) technique and has therefore been largely underexploited. In this work, we clarify the complicated reactions of Cl• with DMPO, which pose challenges for the direct detection of Cl•. To overcome this limitation, we take advantage of the differences in the products forme

Unveiling singlet oxygen spin trapping in catalytic oxidation processes using in situ kinetic EPR analysis.

Singlet oxygen (1O2) plays a pivotal role in numerous catalytic oxidation processes utilized in water purification and chemical synthesis. The spin-trapping method based on electron paramagnetic resonance (EPR) analysis is commonly employed for 1O2 detection. However, it is often limited to time-independent acquisition. Recent studies have raised questions about the reliability of the 1O2 trapper, 2,2,6,6-tetramethylpiperidine (TEMP), in various systems. In this study, we introduce a comprehensive, kinetic examination to monitor the spin-trapping process in EPR analysis. The EPR intensity of t

Ethylene glycol generates free radical metabolites in rats: an ESR in vivo spin trapping investigation.

Ethylene glycol, best known as antifreeze, is most often ingested accidentally or as a substitute for alcochol by chronic alcohol abusers. The toxicity of ethylene glycol poisoning is due to its toxic metabolites rather than to ethylene glycol itself. In this study, electron spin resonance (ESR) spectroscopy has been used to study free radical generation in rats by acute ethylene glycol poisoning. The radical spin trapping technique was applied where the spin trapping agent alpha-(4-pyridyl-1-oxide)-N-tert-butylnitrone (POBN) reacted with free radical metabolites to form radical adducts in viv…

In vivo evidence of free radical formation after asbestos instillation: an ESR spin trapping investigation.

It has been postulated that the in vivo toxicity of asbestos results from its catalysis of free radical generation. We examined in vivo radical production using electron spin resonance (ESR) coupled with the spin trap alpha-(4-pyridyl-1-oxide)-N-t-butylnitrone (4-POBN); 180 day-old rats were intratracheally instilled with either 500 microg crocidolite or saline. Twenty-four hours later, histologic examination revealed a neutrophilic inflammatory response. ESR spectroscopy of the chloroform extract from lungs exposed to asbestos gave a spectrum consistent with a carbon-centered radical adduct,

Spin trapping study in the lungs and liver of F344 rats after exposure to ozone.

Fischer 344 rats were injected with the spin traps C-phenyl N-tert-butyl nitrone (PBN, 150 mg/kg bw, ip) or 4-pyridine-N-oxide N-tert-butyl nitrone (POBN, 775 mg/kg bw, ip), and exposed to clean air or 2 ppm ozone for two hours. The presence of spin adducts was determined by electron paramagnetic resonance (EPR) spectroscopy of chloroform extracts of lung and liver homogenates. No significant levels of adducts were detected in the lungs of air control animals. Benzoyl N-tert-butyl aminoxyl, attributed to direct reaction of ozone with PBN, and tert-butyl hydroaminoxyl, the scission product of t

EPR spin-trapping study of nitric oxide formation during bilateral carotid occlusion in the rat.

The formation of nitric oxide (NO) radicals was demonstrated by electron paramagnetic resonance spectroscopy in the rat during varying degrees of brain ischemia. Diethyldithiocarbamate and Fe-citrate were used as in vivo spin-trapping reagents. The signal of NO spin adducts increased in accordance with the degree of ischemic insults. The formation of NO radicals was inhibited by NG-nitro-L-arginine methyl ester.

The spin trap alpha-(4-pyridyl-1-oxide)-N-tert-butylnitrone stimulates peroxidase-mediated oxidation of deferoxamine. Implications for pharmacological use of spin-trapping agents.

The iron chelator deferoxamine (Desferal; DSFL) reacts with peroxidases and H2O2 to form the DSFL radical (DSFL.), which can be detected by EPR spectroscopy. We have found that DSFL. formation resulting from exposure to H2O2 and any of a number of different peroxidases is greatly enhanced in the presence of the nitrone spin trap alpha-(4-pyridyl-1-oxide)-N-tert-butylnitrone (4-POBN). This enhancement was seen at 4-POBN concentrations as low as 200 microM. We observed a modest enhancement of DSFL. formation with 2-methyl-2-nitrosopropane. However, no enhancement was seen with 5,5-dimethyl-1-pyr

Spin-orbit-coupled fractional oscillators and trapped Bose-Einstein condensates.

We study the ensemble of pseudo-spin 1/2 ultracold bosons, performing Lévy flights, confined in a parabolic potential. The (pseudo-) spin-orbit coupling (SOC) is additionally imposed on these particles. We consider the structure and dynamics of macroscopic pseudospin qubits based on Bose-Einstein condensates, obtained from the above "fractional" bosons. Under "fractional" we understand the substitution of the ordinary second derivative (kinetic energy term) in the Gross-Pitaevskii equation by a so-called fractional Laplacian, characterized by the Lévy index μ. We show that the joint action of

Spin trap salvage from endotoxemia: the role of cytokine down-regulation.

BACKGROUND: The spin trap alpha-phenyl-N-tert-butyl-nitrone (PBN) affords protection from the lethality of septic (lipopolysaccharide) shock. We hypothesized that PBN may work through down-regulation of the sepsis-induced cytokine cascade. METHODS: C3H/HEN mice received 30 mg/kg lipopolysaccharide 15 minutes after pretreatment with PBN or vehicle. Animals were monitored for differences in behavior, histopathologic studies, survival, and serum levels of tumor necrosis factor (TNF-alpha), interferon-gamma (IFN-gamma), and interleukin-6 (IL-6) after lipopolysaccharide. Northern blot analyses of

Spin Polarization in Strong-Field Ionization as Sensor of Trapped Electron Orbits.

We show how spin polarization of photoelectrons produced in strong-field ionization of noble-gas atoms can be used to detect highly unexpected electron orbits created in strong, circularly polarized near-infrared fields. In such fields, ionization is expected to generate photoelectrons rapidly moving away from the ionic core, freezing their initial spin due to the short-range nature of the spin-orbit interaction between the photoelectron and the core. However, we find clear evidence of spin flips in the photoelectron spectra at low photoelectron energies and confirm that they happen because a

Mechanistic studies of the reactions of nitrone spin trap PBN with glutathiyl radical.

We performed mechanistic studies of the reaction of PBN with the physiologically relevant glutathiyl radical, GS*, formed upon oxidation of the intracellular antioxidant, glutathione, GSH. The scavenging rate constant of GS* by PBN has been measured directly by laser flash photolysis and indirectly by competitive EPR of the spin adduct of PBN and another spin trap, DMPO (5,5-dimethyl-1-pyrroline N-oxide), and was found to be 6.7 x 107 M(-1) s(-1). Reverse decomposition of the paramagnetic PBN-glutathiyl radical adduct to the nitrone and thiyl radical was observed for the first time. The rate c

Electron paramagnetic resonance (EPR) spin trapping applied to Chardonnay wines: impact of phenolic content and ethanol.

Chemical oxidation is the main cause of damage during the bottle aging of white wines. It is due to Fenton-type reaction mechanisms, i.e., a radical oxidation cascade which leads to the formation of 1-hydroxyethyl radicals (1-HER) from ethanol. An optimized electron paramagnetic resonance (EPR) approach, using the Fenton reaction and N-tert-Butyl-α-(4-pyridyl-1-oxide) nitrone as spin trap, was used to study the formation kinetic of 1-HER. Important parameters of the reaction were identified and optimized, i.e., temperature, dissolved oxygen and free bisulfite content. We found that acetaldehyd

Immuno-spin trapping from biochemistry to medicine: advances, challenges, and pitfalls. Focus on protein-centered radicals.

BACKGROUND: Immuno-spin trapping (IST) is based on the reaction of a spin trap with a free radical to form a stable nitrone adduct, followed by the use of antibodies, rather than traditional electron paramagnetic resonance spectroscopy, to detect the nitrone adduct. IST has been successfully applied to mechanistic in vitro studies, and recently, macromolecule-centered radicals have been detected in models of drug-induced agranulocytosis, hepatotoxicity, cardiotoxicity, and ischemia/reperfusion, as well as in models of neurological, metabolic and immunological diseases. SCOPE OF THE REVIEW: To…

Clarification of the relationship between free radical spin trapping and carbon tetrachloride metabolism in microsomal systems.

It has been proposed that the C-phenyl-N-tert-butylnitrone/trichloromethyl radical adduct (PBN/.CCl3) is metabolized to either the C-phenyl-N-tert-butylnitrone/carbon dioxide anion radical adduct (PBN/.CO2-) or the glutathione (GSH) and CCl4-dependent PBN radical adduct (PBN/[GSH-.CCl3]). Inclusion of PBN/.CCl3 in microsomal incubations containing GSH, nicotinamide adenine dinucleotide phosphate (NADPH), or GSH plus NADPH produced no electron spin resonance (ESR) spectral data indicative of the formation of either the PBN/[GSH-.CCl3] or PBN/.CO2- radical adducts.
