Methods for Monitoring ABCA1-Dependent Sterol Release.

Releasing sterols to the extracellular milieu is an important part of sterol homeostasis in cells and in the body. ATP-binding cassette transporter A1 (ABCA1) plays an essential role in cellular phospholipid and sterol release to lipid-free or lipid-poor apolipoprotein A-I (apoA-I), the major apolipoprotein in high-density lipoprotein (HDL), and constitutes the first step in the formation of nascent HDL. Loss-of-function mutations in the ABCA1 gene lead to a rare disease known as Tangier disease that causes severe deficiency in plasma HDL level. Mammalian cells receive exogenous cholesterol ma…

Intestinal sterol transporters and cholesterol absorption inhibition.

PURPOSE OF REVIEW: Statin therapy is the mainstay of lipid-lowering therapy; however, many patients, particularly those at high risk, do not achieve sufficient LDL-cholesterol (LDL-C) lowering. Thus, there remains an unmet medical need for more effective and well tolerated lipid-lowering agents. Guidelines recommend combining additional lipid-lowering agents with a complementary mode of action for these patients. One approach to complementing statin therapy is combination with inhibitors that block the intestinal absorption of dietary and biliary cholesterol. This review summarizes what is cur

Hepatic management of toxic sterols after acute deletion of Cyp51 from cholesterol synthesis.

Lanosterol 14α-demethylase (CYP51), a key enzyme in cholesterol synthesis, is essential for normal liver function. Reduced CYP51 activity leads to metabolism-associated liver disease and ultimately hepatocellular carcinoma, yet the hepatocellular processes most affected and their crosstalk with other liver cell types remain poorly defined. Here, we present a new inducible liver-specific Cyp51 KO mouse (iLKO) model designed to study how acute disruption of cholesterol synthesis is managed in the adult liver. Doxycycline-inducible deletion minimizes developmental confounders of albumin-Cre model

Noncholesterol sterols in bile and stones of patients with cholesterol and pigment stones.

Human bile and cholesterol gallstones contain sterols including methylated (lanosterol and other dimethyl and monomethyl sterols), and demethylated cholesterol precursor sterols (delta 8-lathosterol, lathosterol, and desmosterol), plant sterols (campesterol and sitosterol), and cholestanol. The aim of the study was to analyze the noncholesterol sterols in gallbladder bile and stones from female and male patients with cholesterol stones (CS) and pigment stones (PS) to ascertain whether any sterol fraction contributes to the gallstone formation. Classification of gallstones to CS and PS was perf…

[A method of quantitative determination of delta 5,7-sterols in unsaponifiable lipid fractions].

The technique is offer for rapid quantitative definition of sterols having a system of delta 5,7-conjugated double bonds which conditions biological activity of D vitamin--the result of photoisomerisation of the sterols. The technique bases on the analysis of UV absorption spectra of unsaponifiable lipid fraction of several kinds of mutant yeast strains and the model mixtures of sterols. The results obtained concerning the estimation of delta 5,7-sterols containing in total fraction are confirmed by gas-liquid-chromatography and mass-spectrometry data.

Role of hydrophilic bile acids and of sterols on cholelithiasis in the hamster.

The effect of various dietary additions such as cholesterol, beta-sitosterol, bile acids, and bile acid analogs on gallstone formation was studied in the hamster. Gallstones were formed in 50% of the animals fed a high glucose, fat-free diet. Administration of 0.2% cholesterol or 1% beta-sitosterol had no effect on the incidence of gallstones. Ursodeoxycholic acid (0.5%) and its analog ursodeoxy-oxazoline [2-(3 alpha, 7 beta-dihydroxy-24-nor-5 beta-cholanyl)-4,4-dimethyl-2- oxazoline] were ineffective in preventing gallstones. Hyodeoxycholic acid and hyodeoxy-oxazoline [2-(3 alpha,6 alpha-dihy

Analysis of Phloem Sterols in Plants using EDTA-Facilitated Exudation and GC-MS.

Phytosterols occur in the phloem of vascular plants as free, acylated, and glycosylated derivatives. The phloem is increasingly recognized as an active site of sterol synthesis and transport used to support plant metabolism, while also providing a potential nutritional source for phloem-feeding insects. This chapter describes a method for the collection and quantitative analysis of phloem sterols using an EDTA-facilitated exudation procedure followed by gas chromatography-mass spectrometry (GC-MS). Excised leaves are incubated in an EDTA solution to induce phloem exudation, and sterols from co…

Exploration of Hypolipidemic Effects of Sterols from Pleurotus tuber-regium(Fr.) Sing Sclerotium.

The extraction technology of sterol was confirmed by ethanol reflux and saponification in this study. The orthogonal test was employed to assess the impact of extraction time, solid-liquid ratio, ethanol concentration and extraction temperature on the yield of sterol extraction. Hyperlipidemia model mice were established by feeding a high-fat and -sugar diet, and different doses of sterol extracts were given to the mice by gavages. The optimal extraction conditions were identified as an extraction time of 80 min, a solid-liquid ratio of 1:10, an ethanol concentration of 95%, and an extraction

Deep learning reveals endogenous sterols as allosteric modulators of the GPCR-Gα interface.

Endogenous intracellular allosteric modulators of GPCRs remain largely unexplored, with limited binding and phenotype data available. This g … chemical assays, and physiological readouts, identified endogenous hydrophobic metabolites, notably sterols, as direct intracellular allosteric modulators of Ste2p. Molecular simulations coupled with biochemical signaling assessment in site-directed Ste2p mutants further confirmed that metabolites binding to GPCR-Gα obstruct downstream signaling, possibly …

Identification of C31 and C32 sterols in Pneumocystis carinii hominis-infected human lungs.

Two sterols in autopsied whole lung specimens obtained from Pneumocystis carinii pneumonia patients were detected by gas-liquid chromatography and their structures were elucidated by mass spectrometry and nuclear magnetic resonance spectrometry. Both were in the lanosterol series; the C31 sterol, with a methyl group at C-24, was identified as euphorbol, and the more abundant C32 sterol, with an ethyl group at C-24, is given the trivial name pneumocysterol.

Phytosterolaemia, xanthomatosis and premature atherosclerotic arterial disease: a case with high plant sterol absorption, impaired sterol elimination and low cholesterol synthesis.

A fourth case is described in which phytosterolaemia, earlier diagnosed as familial hypercholesterolaemia, was associated with normocholesterolaemia, hypersplenism and premature atherosclerotic arterial disease requiring a three-vessel coronary bypass at the age of 29 years. During a follow-up of 5 years 22-26% and 27-30% of serum and bile sterols were plant sterols, respectively. In addition to campesterol and beta-sitosterol, stigmasterol and a fourth major plant sterol, tentatively identified as avenasterol, were found in bile, and in free and esterified forms in all serum lipoproteins. Ana…

Modulation of human Niemann-Pick C1-like 1 gene expression by sterol: Role of sterol regulatory element binding protein 2.

Niemann-Pick C1-like 1 (NPC1L1) is an essential intestinal component of cholesterol absorption. However, little is known about the molecular regulation of intestinal NPC1L1 expression and promoter activity. We demonstrated that human NPC1L1 mRNA expression was significantly decreased by 25-hydroxycholesterol but increased in response to cellular cholesterol depletion achieved by incubation with Mevinolin (an inhibitor of 3-hydroxy-3-methylglutaryl-CoA reductase) in human intestinal Caco-2 cells. We also showed that a -1741/+56 fragment of the NPC1L1 gene demonstrated high promoter activity in

Metoprolol Inhibits Developmental Brain Sterol Biosynthesis in Mice.

De novo sterol synthesis is a critical homeostatic mechanism in the brain that begins during early embryonic development and continues throughout life. Multiple medications have sterol-biosynthesis-inhibiting side effects, with potentially detrimental effects on brain health. Using LC-MS/MS, we investigated the effects of six commonly used beta-blockers on brain sterol biosynthesis in vitro using cell lines. Two beta-blockers, metoprolol (MTP) and nebivolol, showed extreme elevations of the highly oxidizable cholesterol precursor 7-dehydrocholesterol (7-DHC) in vitro across multiple cell lines

Sterol Sponge Mechanism Is Conserved for Glycosylated Polyene Macrolides.

Amphotericin-like glycosylated polyene macrolides (GPMs) are a clinically and industrially important family of natural products, but the mechanisms by which they exert their extraordinary biological activities have remained unclear for more than half a century. Amphotericin B exerts fungicidal action primarily via self-assembly into an extramembranous sponge that rapidly extracts ergosterol from fungal membranes, but it has remained unclear whether this mechanism is applicable to other GPMs. Using a highly conserved polyene-hemiketal region of GPMs that we hypothesized to represent a conserved

Antileishmanial effect of mevastatin is due to interference with sterol metabolism.

Visceral leishmaniasis (VL) is one of the most severe forms of leishmaniasis which is fatal if left untreated. Sterol biosynthetic pathway in Leishmania is currently being explored for its therapeutic potential. In the present study, we have evaluated the antileishmanial efficacy of mevastatin, a known inhibitor of 3-hydroxy-3-methyl glutaryl-CoA reductase (HMGR) enzyme. Mevastatin inhibited Leishmania donovani promastigotes and intracellular amastigotes with an 50% inhibitory concentration (IC50) value of 23.8 ± 4.2 and 7.5 ± 1.1 μM, respectively, without exhibiting toxicity towards host cell

SIRT2 inhibition achieves neuroprotection by decreasing sterol biosynthesis.

Huntington's disease (HD), an incurable neurodegenerative disorder, has a complex pathogenesis including protein aggregation and the dysregulation of neuronal transcription and metabolism. Here, we demonstrate that inhibition of sirtuin 2 (SIRT2) achieves neuroprotection in cellular and invertebrate models of HD. Genetic or pharmacologic inhibition of SIRT2 in a striatal neuron model of HD resulted in gene expression changes including significant down-regulation of RNAs responsible for sterol biosynthesis. Whereas mutant huntingtin fragments increased sterols in neuronal cells, SIRT2 inhibitio…

Identification of mosquito sterol carrier protein-2 inhibitors.

A mosquito sterol carrier protein-2, AeSCP-2, has been shown to aid in the uptake of cholesterol in mosquito cells. The discovery of chemical inhibitors of AeSCP-2 is reported here. AeSCP-2 inhibitors (SCPIs) belong to several chemotypes of hydrophobic compounds. Those inhibitors competed with cholesterol for AeSCP-2, binding with relatively high binding affinities. In cultured insect cells, SCPIs reduced cholesterol uptake by as much as 30% at 1-5 microM concentrations. SCPIs were potent larvicides to the yellow fever mosquito, Aedes aegypti, and to the tobacco hornworm, Manduca sexta, with 5
