Neuronal damage of the substantia nigra in HIV-1 infected brains.

Extrapyramidal motor disorders are frequently noted in HIV-1-infected patients. In the present study, the substantia nigra was analyzed morphometrically to detect neuronal changes which might contribute to the pathogenetic mechanisms causing extrapyramidal motor dysfunction in HIV-1-infected patients. The numerical density and the size of pigmented, non-pigmented small, and non-pigmented large neurons in four nuclei of the substantia nigra pars compacta (antero-medial, antero-intermediolateral, postero-lateral, and postero-medial nuclei) in HIV-1-infected patients and in age-matched normal con…

Parkinsonism due to predominant involvement of substantia nigra in Japanese encephalitis.

OBJECTIVE: To study the clinical correlates of lesions seen predominantly in the substantia nigra in some patients with Japanese encephalitis (JE). BACKGROUND: JE typically involves thalamus, brainstem, spinal cord, and cerebral cortex. Rarely, basal ganglia and cerebellum may be affected. Lesions are often widespread and discrete. Predominant involvement of substantia nigra in JE has not been previously reported. METHODS: Of 52 patients with JE seen in an endemic zone, five were selected on the basis of isolated lesions in the substantia nigra on MRI; all were subjected to detailed clinical…

Alterations of striatal cholinergic receptors after lesioning of the substantia nigra.

Dopamine deficiency syndrome is known to cause cholinergic hyperactivity. Therefore, it was hypothesized that the said phenomenon may be due to enhanced cholinergic receptor functions. In the present study we examined the changes in striatal dopaminergic and cholinergic receptors in unilateral substantia nigra lesioned rats that showed vigorous ipsilateral rotation (total turns > 300) in response to apomorphine (1 mg kg(-1) ip). [3H] Spiperone ([3H]-SP) and [3H]-quinuclidinyl benzilate ([3H]-QNB) bindings were performed in the striata of the lesioned animals. There was no significant differenc…

Localisation of glutamate receptors in the substantia nigra pars compacta of the monkey.

The distribution of ionotropic glutamate receptor subunits GluR1, GluR2/3 and NMDAR1, and meta-botropic receptor mGluR1 alpha was studied in the monkey substantia nigra. High levels of immunoreactivity to GluR1, GluR2/3 and NMDAR1, and moderate levels of immunoreactivity to mGluR1 alpha were observed in the substantia nigra pars compacta. GluR1 and GluR2/3 were mostly in cell bodies and larger stem dendrites, whilst NMDAR1 and mGluR1 alpha were present on medium sized and small dendrites, respectively. The substantia nigra receives glutamatergic afferents from the subthalamic nucleus and the f…

Preliminary evidence for neurodegenerative changes in the substantia nigra of Rett syndrome.

Rett syndrome (RS), which affects approximately 1 in 10,000 young females, is characterized by cognitive deterioration, ataxia, apraxia, rigidity, and stereotyped hand movements. Neuropathological features include reduction in brain size and hypopigmentation of neurons of the substantia nigra pars compacta (SNpc). Neurochemical and imaging studies support nigrostriatal involvement. The results of our preliminary studies show abnormalities in neurons of the substantia nigra (SN), including decreased numbers of neurons, ubiquitin-stained neuronal inclusion bodies, decreased immunostaining for tr…

Dopa-decarboxylation in the striata of rats with unilateral substantia nigra lesions.

The source and site of the DOPA decarboxylation to dopamine in Parkinson's disease (PD) and animal models of PD are controversial. Since most of aromatic L-amino acid decarboxylase (AADC) are lost along with the degenerating dopaminergic neurons, we addressed the possibility that other decarboxylases or a novel protein that is structurally different from AADC decarboxylate L-DOPA in the denervated striatum. Immunotitration of the extracts from the denervated striatum with AADC antibody showed that all activity can be attributed to AADC-immunoreactive protein. We then investigated if there are

Memory-contingent saccades and the substantia nigra postulate for essential blepharospasm.

Essential blepharospasm and cranial dystonia are related focal dystonias of unknown aetiology. Blepharospasm induced by acute dopamine depletion in parkinsonism restricts saccade initiation possibly via the substantia nigra pars reticulata (SNpr). If essential blepharospasm and cranial dystonia similarly restrict saccades, then a selective, somatotopically arranged pathway such as the SNpr may be involved. To test this possibility memory-contingent and visually-guided saccades were measured in patients with essential blepharospasm and cranial dystonia. The latency of both forms of saccades was…

Role of the substantia nigra in audiogenic seizures: a neuroethological analysis in the rat.

The audiogenic crisis (AC) includes an initial phase of bursts of circular, stereotyped running. Since it has been shown that the nigrostriatal system is important in the control of lesion- and drug-induced stereotyped rotational behavior, the effects of unilateral electrolytic lesions of the substantia nigra (SN) on the AC were investigated. Behavior was recorded using a detailed ethogram by means of systematic observational techniques and analyzed quantitatively in terms of sequential structure by multivariate statistical methods. An index of susceptibility was computed on the basis of the f…

Specificity of GABA as a stimulator of dopamine release in rat substantia nigra.

The effects of amino acids and the benzodiazepine, flurazepam, on spontaneous and potassium-stimulated [3H]dopamine (DA) release were studied in slices of rat substantia nigra. None of the amino acids tested liberated [3H]DA directly. Flurazepam increased [3H]DA output by 51% and this effect was partially blocked by bicuculline but not by picrotoxin. GABA, L-glutamate and flurazepam, but none of the other compounds, potentiated the release of [3H]DA evoked by 20 mM KCl. The facilitation produced by flurazepam, but not by GABA or L-glutamate, was weakly antagonized by bicuculline and not picrot…

HFE genotype influences substantia nigra iron accumulation and Parkinson's disease progression.

Increased iron in the substantia nigra has been thought to be a mechanism potentially related to the etiology and/or progression of Parkinson's disease (PD). We hypothesized that genetic variants of HFE, a major iron regulatory gene, would influence substantia nigra iron accumulation in PD. The HFE genotype was obtained from 195 subjects (102 PD and 83 Controls) who participated in the PD biomarker program (PDBP) in central Pennsylvania, United States. For this study, carriers of two SNPs (HFE H63D and/or C282Y) were considered as variants and the others as wildtype. Susceptibility MRI metrics…

Striatal Infarction Elicits Secondary Extrafocal MRI Changes in Ipsilateral Substantia Nigra.

Focal ischemia may induce pathological alterations in brain areas distant from the primary lesion. In animal models, exofocal neuron death i … images with a digitized anatomic atlas revealed localization of secondary lesions primarily in the substantia nigra pars compacta. Apparent diffusion coefficient (ADC) values in the secondary lesions showed a delayed sharp decline through day 10. Normalization of ADC values was observed at late measurements. Taken together, our study demonstrates that striatal in …

Substantia nigra neuromelanin-MR imaging differentiates essential tremor from Parkinson's disease.

BACKGROUND: Essential tremor (ET) is a very common movement disorder that has no diagnostic markers. Differentiation with Parkinson's disease (PD) can be clinically challenging in some cases, with a high rate of misdiagnosis. Magnetic resonance imaging (MRI) studies have been able to identify neuromelanin changes in the substantia nigra (SN) of PD patients, but they have thus far not been investigated in ET. In this study, we aimed to characterize neuromelanin-MR signal changes in ET and evaluate its diagnostic accuracy in the differential diagnosis with PD. METHODS: The inclusion criteria we…

Intraventricular insulin increases dopamine transporter mRNA in rat VTA/substantia nigra.

The hormone insulin can down-regulate the function and synthesis of the re-uptake transporter for norepinephrine (NET) in vivo and in vitro. In the present study we tested whether this action of insulin is generalized to another member of the catecholamine transporter family. We determined the level of dopamine transporter (DAT) mRNA expression in the ventral tegmental area (VTA)/substantia nigra compacta (SNc) of rats which were chronically treated with vehicle or insulin via the third cerebral ventricle (i.c.v.). DAT mRNA was significantly elevated in the VTA/SNc of rats treated with insulin…

GDNF modulates HO-1 expression in substantia nigra postnatal cell cultures.

Heme oxygenase-1 (HO-1) has been strongly highlighted because of its induction in many cell types by toxic stimuli, including oxidative stress. The intense HO-1 immunostaining in the substantia nigra of Parkinson disease (PD) patients suggests its involvement in the pathogenesis of this neurodegenerative disease. In this work we investigated HO-1 expression in rat substantia nigra postnatal cell cultures under conditions mimicking dopamine toxicity and its modulation by glial cell line-derived neurotrophic factor (GDNF), a potent neuroprotective factor for dopaminergic neurons. In neuron-glia …

Oxidative stress and microglial activation in substantia nigra following striatal MPP+.

The present study aims to study sequential alterations occurring in both dopaminergic neurons and microglia in substantia nigra (SN) following intrastriatal injection of 1-methyl-4-phenylpridium ion (MPP+) in rats. Heme oxygenase-1 (HO-1), a marker of oxidative stress, first appeared in dopaminergic neurons in SN at 1 day post-lesion. Subsequently, microglia in SN exhibited morphological changes indicative of activation. At 7 days post-lesion, those findings increased severity and 7a significant reduction in the number of dopaminergic neurons was observed. The present finding suggests that ext…
