Cold acclimation-recruited nonshivering thermogenesis: the Syrian hamster is not an exception.

Biochemical evidence from in vitro studies of brown adipose tissue in Syrian hamsters indicates a significant degree of recruitment of the tissue as an effect of cold acclimation. However, earlier in vivo studies indicate a lack of recruitment of nonshivering thermogenesis in the intact animal as a result of cold acclimation. Because of this apparent discrepancy, the occurrence of cold acclimation-recruited nonshivering thermogenesis in hamsters was investigated. Hamsters were cold acclimated to 6 degrees C or remained at 24 degrees C (controls), and their thermogenic response was investigated…

THE ONTOGENY OF SHIVERING THERMOGENESIS IN THE RED-WINGED BLACKBIRD (AGELAIUS PHOENICEUS).

The ontogeny of shivering thermogenesis was investigated in the altricial red-winged blackbird (Agelaius phoeniceus). Two indices of heat production the rate of oxygen consumption (V(dot)O2) of the bird and the electromyographic (EMG) activity of the pectoralis (PECT) and gastrocnemius (GAST) muscles were measured simultaneously in adult and nestling red-winged blackbirds as they were subjected first to thermoneutral temperatures and subsequently to progressively colder ambient temperatures (Ta). The ontogenetic changes in both indices indicated that the capability for thermogenesi…

Ephedrine, xanthines and prostaglandin-inhibitors: actions and interactions in the stimulation of thermogenesis.

The pivotal role of the sympathoadrenal system in the defense of le milieu interieur has, in the last 15 years, been extended to include the fat stores-a notion that forms the basis of current strategies for thermogenic stimulation in obesity therapy. The search for effective and safe sympathetic stimulants has been directed at two main levels: (i) the development of novel beta-agonists selective for thermogenesis, and (ii) the evaluation of drugs already in clinical use for other purposes (e.g. ephedrine) which could conceivably increase the release of catecholamines to levels that enhance th…

EBF2 condensates enable adipose thermogenesis through ZFP423 sequestration and epigenetic remodeling.

Brown adipose tissue (BAT) dissipates energy through non-shivering thermogenesis and holds promise as a therapeutic target for obesity. The transcription factor Early B Cell Factor 2 (EBF2) is a key regulator of the thermogenic gene program, yet its underlying transcriptional regulatory mechanisms and potential for pharmacological targeting remain incompletely defined. Here, we identify a low-complexity C-terminal domain (CTD) within EBF2 that drives biomolecular condensation critical for thermogenic activation. Deletion of the CTD (ΔCTD) or mutation of conserved proline residues disrupts EBF2…

Sleep deprivation stimulates adaptive thermogenesis by activating AMPK pathway in mice.

Sleep deprivation (SD) can affect the adaptive thermogenesis in laboratory rodents, but the molecular mechanism and the crosstalk with other organs remain largely unknown. In order to investigate the effects and mechanisms of SD on thermoregulation and energy metabolism, here we measured the changes of body weight, body fat mass, body temperature, resting metabolic rate (RMR), and thermogenic gene expression in brown adipose tissue (BAT), white adipose tissue (WAT), skeleton muscle and liver in C57BL/6J mice during 7-day SD with rotating rod sleep deprivation device. Results showed that compar…

Brown fat thermogenesis and branched-chain amino acids in metabolic disease.

Since the 1960s, researchers have recognized an association between elevated plasma branched chain amino acids (BCAA) and metabolic disease, … sis. Previous investigations into BAT have emphasized fatty acids and glucose as substrates for BAT thermogenesis. Here, we address the importance of BAT in systemic BCAA metabolism, driven via the newly identified mitochondrial BCAA carrier (MBC), as well as the impact of BAT-driven BCAA clearance on glucose homeostasis and metabolic disease. The newly identif …

Corin Deficiency Alters Adipose Tissue Phenotype and Impairs Thermogenesis in Mice.

Atrial natriuretic peptide (ANP) is a key regulator in body fluid balance and cardiovascular biology. In addition to its role in enhancing natriuresis and vasodilation, ANP increases lipolysis and thermogenesis in adipose tissue. Corin is a protease responsible for ANP activation. It remains unknown if corin has a role in regulating adipose tissue function. Here, we examined adipose tissue morphology and function in corin knockout (KO) mice. We observed increased weights and cell sizes in white adipose tissue (WAT), decreased levels of uncoupling protein 1 (Ucp1), a brown adipocyte marker in W…

Magnolol promotes thermogenesis and attenuates oxidative stress in 3T3-L1 adipocytes.

OBJECTIVE: The aim of this study was to explore the browning and antioxidative effects of magnolol in 3T3-L1 adipocytes, as recruitment of beige-like adipocytes (browning) by natural compounds is being considered as a promising strategy to fight against obesity. METHODS: Magnolol-induced browning effect was evaluated by determining the expression levels of specific marker genes and proteins using real-time polymerase chain reaction and immunoblotting, respectively. Induction of thermogenesis and suppression of oxidative stress in 3T3-L1 adipocytes were further validated by immunofluorescence.…

Regulator of Calcineurin 1 helps coordinate whole-body metabolism and thermogenesis.

Increasing non-shivering thermogenesis (NST), which expends calories as heat rather than storing them as fat, is championed as an effective way to combat obesity and metabolic disease. Innate mechanisms constraining the capacity for NST present a fundamental limitation to this approach, yet are not well understood. Here, we provide evidence that Regulator of Calcineurin 1 (RCAN1), a feedback inhibitor of the calcium-activated protein phosphatase calcineurin (CN), acts to suppress two distinctly different mechanisms of non-shivering thermogenesis (NST): one involving the activation of UCP1 expr…

Distinct sympathetic projections to brown fat regulate thermogenesis and glucose tolerance.

Brown adipose tissue (BAT) contributes to thermoregulation and glucose metabolism, but how these functions are coordinated remains unclear. While thermogenesis in the activated BAT typically coincides with increased blood flow and glucose uptake1-5, several pathophysiological and nutritional states dissociate these processes6,7, suggesting they are governed by distinct sympathetic circuits. Here we identify subpopulations of sympathetic neurons in the stellate ganglion that mediate distinct functions of intrascapular BAT (iBAT) in mice. Two main types of sympathetic neurons project to iBAT: th…

Cold exposure aggravates vein occlusion through non-shivering thermogenesis-induced thrombocytopoiesis.

Vein occlusion (VO), including deep venous thrombosis (DVT) and retinal vein occlusion (RVO), is a common cause of multiple diseases that severely compromise the quality of life of affected individuals. Epidemiological evidence indicates that VO prevalence increases in cold seasons, yet the underlying mechanism remains unknown. Here, we show that cold exposure markedly elevates peripheral platelet counts, thereby aggravating VO in mouse models. Cold-augmented thrombocytopoiesis depends on the activation of adipose thermogenesis and subsequent increase in circulating free fatty acid (FFA) level…

Heme biosynthesis regulates BCAA catabolism and thermogenesis in brown adipose tissue.

With age, people tend to accumulate body fat and reduce energy expenditure 1 . Brown (BAT) and beige adipose tissue dissipate heat and increase energy expenditure via the activity of the uncoupling protein UCP1 and other thermogenic futile cycles 2,3 . The activity of brown and beige depots inversely correlates with BMI and age 4-11 , suggesting that promoting thermogenesis may be an effective approach for combating age-related metabolic disease 12-15 . Heme is an enzyme cofactor and signaling molecule that we recently showed to regulate BAT function 16 . Here, we show that heme biosynthesis i…

Leptin Improves Parameters of Brown Adipose Tissue Thermogenesis in Lipodystrophic Mice.

Lipodystrophy syndromes (LD) are a heterogeneous group of very rare congenital or acquired disorders characterized by a generalized or partial lack of adipose tissue. They are strongly associated with severe metabolic dysfunction due to ectopic fat accumulation in the liver and other organs and the dysregulation of several key adipokines, including leptin. Treatment with leptin or its analogues is therefore sufficient to reverse some of the metabolic symptoms of LD in patients and in mouse models through distinct mechanisms. Brown adipose tissue (BAT) thermogenesis has emerged as an important …

Chronic arsenic exposure impairs adaptive thermogenesis in male C57BL/6J mice.

The global prevalence of type 2 diabetes (T2D) has doubled since 1980. Human epidemiological studies support arsenic exposure as a risk factor for T2D, although the precise mechanism is unclear. We hypothesized that chronic arsenic ingestion alters glucose homeostasis by impairing adaptive thermogenesis, i.e., body heat production in cold environments. Arsenic is a pervasive environmental contaminant, with more than 200 million people worldwide currently exposed to arsenic-contaminated drinking water. Male C57BL/6J mice exposed to sodium arsenite in drinking water at 300 μg/L for 9 wk experien…

Grb10 promotes lipolysis and thermogenesis by phosphorylation-dependent feedback inhibition of mTORC1.

Identification of key regulators of lipid metabolism and thermogenic functions has important therapeutic implications for the current obesit … essed lipolysis, and reduced thermogenic function. The effects of Grb10 deficiency on lipolysis and thermogenesis were diminished by rapamycin administration in vivo. Our study has uncovered a unique feedback mechanism regulating mTORC1 signaling in adipose tissues and identified Grb10 as a key regulator of adiposity, thermogenesis, and energy expenditure.

[The effect of palmitic, oleic and linoleic acids on thermogenesis in the model experiment in sows].

An animal model experiment was conducted with nine adult sows to study the effects of long-chain fatty acids on thermogenesis when different fatty acids were replaced for 30% of the energy of a basal diet based on cereals and soybean meal. The acids were fed as commercial products containing as main constituent either palmitic acid, oleic acid, or linoleic acid, according to a latin square design in experimental periods 2 to 4. In periods 1 and 5 the sows were submitted to basal diet alone. Digestibility of palmitic acid was only 36%, whereas the unsaturated fatty acids were highly absorbed (9…

Comparison of brown adipose tissue thermogenesis induced by congeners and isomers of phenylpropanolamine.

The present experiment compared the effects of intraperitoneal injection (.0223 mMol/kg) of several phenethylamine congeners and isomers including amphetamine (AMP), ephedrine (EPH), methoxyphenamine (MET), norpseudoephedrine (NOR), pseudoephedrine (PS) and phenylpropanolamine (PPA) on in vivo interscapular brown adipose tissue temperature in adult male rats. Comparisons of isomer potency revealed that the 1-isomer was more thermogenic than the d-isomer for EPH and PPA but not for AMP, NOR and PS. Congener potency order was: AMP greater than PPA greater than EPH greater than NOR = MET greater
