Exposure to perchlorate, nitrate, and thiocyanate among pregnant women in Norway.

BACKGROUND: The sodium-iodide symporter (NIS) inhibitors perchlorate, thiocyanate, and nitrate are ubiquitous environmental contaminants that competitively inhibit iodine uptake into the thyroid gland as well as iodine transport across the placenta. Globally, iodine deficiency remains common, and inhibition of NIS, reducing iodine transport, may increase the risk of impaired thyroid function. AIM: To characterize pregnant women's exposure to NIS inhibitors by measuring the urine concentrations of perchlorate, thiocyanate, and nitrate, and explore their determinants. We also examined individua

The therapeutic potential of thiocyanate and hypothiocyanous acid against pulmonary infections.

Hypothiocyanous acid (HOSCN) is an endogenous oxidant produced by peroxidase oxidation of thiocyanate (SCN-), an ubiquitous sulfur-containing pseudohalide synthesized from cyanide. HOSCN serves as a potent microbicidal agent against pathogenic bacteria, viruses, and fungi, functioning through thiol-targeting mechanisms, independent of currently approved antimicrobials. Additionally, SCN- reacts with hypochlorous acid (HOCl), a highly reactive oxidant produced by myeloperoxidase (MPO) at sites of inflammation, also producing HOSCN. This imparts both antioxidant and antimicrobial potential to SC

Biochemical mechanisms and therapeutic potential of pseudohalide thiocyanate in human health.

Thiocyanate (SCN(-)) is a ubiquitous molecule in mammalian biology, reaching up to mM concentrations in extracellular fluids. Two- electron oxidation of SCN(-) by H2O2 produces hypothiocyanous acid (HOSCN), a potent anti-microbial species. This reaction is catalyzed by chordate peroxidases (e.g., myeloperoxidase and lactoperoxidase), occurring in human secretory mucosa, including the oral cavity, airway, and alimentary tract, and regulates resident and transient flora as part of innate immunity. Increasing SCN(-) levels limits the concentrations of a family of 2-electron oxidants (H2O2, hypoha

Normative concentrations of urine thiocyanate in cassava eating communities in Nigeria.

Exposure to cyanide is a major public health problem where highly cyanogenic cassava foods are consumed. Thiocyanate (SCN), the biomarker of exposure to cyanide is present in several foods, and produced endogenously. Concentrations of urine SCN were measured in endemic and non-endemic areas of ataxic polyneuropathy in Nigeria. Cassava food consumption in the endemic area was twice that of non-endemic areas. Geometrical mean (95% CI) urine SCN was 20 µmol/l (18-24) for no consumption of cassava foods, 56 µmol/l (49-64) for daily consumption, 56 µmol/l (48-65) for twice daily consumption and 85

Concentration of the antibacterial precursor thiocyanate in cystic fibrosis airway secretions.

A recently discovered enzyme system produces antibacterial hypothiocyanite (OSCN(-)) in the airway lumen by oxidizing the secreted precursor thiocyanate (SCN(-)). Airway epithelial cultures have been shown to secrete SCN(-) in a CFTR-dependent manner. Thus, reduced SCN(-) availability in the airway might contribute to the pathogenesis of cystic fibrosis (CF), a disease caused by mutations in the CFTR gene and characterized by an airway host defense defect. We tested this hypothesis by analyzing the SCN(-) concentration in the nasal airway surface liquid (ASL) of CF patients and non-CF subjects

Application of potassium dichromate and iron-thiocyanate in the spectrophotometric investigations of phenothiazines.

A spectrophotometric procedure for the determination of phenothiazines in pure form and in a number of their pharmaceutical preparations has been developed that offers the advantages of simplicity, accuracy, precision and sensitivity over many other methods. The method is based on the oxidation of phenothiazines by a known excess amount of potassium dichromate followed by the estimation of unreacted amount of dichromate by reacting with excess of iron(II) and measuring the iron(III) formed by complexing with thiocyanate. The reacted oxidant corresponds to the drug content. Different variables

Oxidative conversion of thiocyanate to cyanide by oxyhemoglobin during acid denaturation.

Human oxyhemoglobin (Hb-O2) converts thiocyanate to cyanide under acidic conditions, in a hemoglobin and thiocyanate concentration-dependent manner. Other naturally occurring hemoglobin derivatives and hematin did not result in cyanide-producing activity. Pre-denaturation completely destroyed cyanide production. In the presence of 10% phosphoric acid, 12 microM cyanide was produced by 1 mM Hb-O2 (per heme) and 1 mM thiocyanate. This conversion was observed over a wide range of acidic pH levels at 50 degrees C, with an optimal pH of 4. The production of cyanide under strong acidic conditions wa

Electrical responses of the rat's retinal pigment epithelium to azide and thiocyanate.

In the rat under urethane anesthesia, a fast intravenous injection of a bolus of sodium azide elicited a transient cornea-positive change in transocular potential (azide response). A bolus injection of sodium thiocyanate (NaSCN) produced a cornea-negative response (SCN- response) with a similar rising phase as the azide response, but with a faster return from the peak. The peak amplitude depended on bolus volume, concentration, animal strain, and age. For more than 24 h, the azide and SCN- responses could be recorded repeatedly from a single rat with little variation in peak amplitudes. Follow

Modifiers of mercaptopyruvate sulfurtransferase catalyzed conversion of cyanide to thiocyanate in vitro.

The enzyme mercaptopyruvate sulfurtransferase appears to play an important role in the in vivo detoxification of cyanide. It does so by transferring sulfur to cyanide to produce thiocyanate, which is less toxic and may be excreted through the kidney. Several compounds were tested for their ability to affect the rate of enzyme catalyzed thiocyanate formation in vitro. The studies were carried out using both a partially purified bovine kidney extract and a highly purified enzyme preparation. Hypotaurine and methanesulfinic acid doubled sulfurtransferase activity in the partially purified extract

Validity of salivary thiocyanate as an indicator of cyanide exposure from smoking.

The purpose of this study was to evaluate the validity of salivary thiocyanate as an indicator of cyanide exposure from smoking. The salivary thiocyanate level showed wide variation, and the largest source of thiocyanate was hydrogen cyanide from cigarette smoke. Hydrogen cyanide in the mainstream of cigarette smoke was as high as 40-70 ppm, which is beyond the threshold limit value for occupational health (10 ppm). The hydrogen cyanide level in the sidestream smoke was lower less than 5 ppm. Thus the amount of cyanide intake was larger in active smokers than in passive ones. The correlation b

Ring-Opening Thiocyanation Reaction of Sulfonium Salts with Elemental Sulfur and TMSCN.

A simple and efficient strategy has been developed for the synthesis of organic thiocyanates through direct ring-opening thiocyanation reactions of aryl, alkenyl, and alkynyl sulfonium ylides using elemental sulfur and TMSCN. Using elemental sulfur as the sulfur source, this transformation proceeds under mild conditions without the need for metal reagents or malodorous materials. Notably, this approach features excellent versatility, as evidenced by its compatibility with scale-up synthesis and the late-stage modification of the corresponding products.

Computational exploration of the electrochemical oxidation mechanism of thiocyanate catalyzed by cobalt-phthalocyanines.

In this study, we focused on the mechanism of the electrocatalytic oxidation of thiocyanate, which in traditional electrodes typically requires high overpotentials. As models for reducing these overpotentials and catalyzing the reaction, we used a set of modified cobalt phthalocyanines (CoPc), known as electrocatalysts. Using DFT calculations, we explored how modifications to CoPc by adding electron-donating and withdrawing groups and the coordination of 4-amino thiophenol impact the oxidation process. The reaction mechanism for the electrooxidation of thiocyanate has remained elusive, where o

Electrochemical Synthesis of α-Thiocyanated/Methoxylated Ketones Using Enol Acetates.

We have developed the synthesis of α-substituted ketone compounds with enol acetates in an electrochemical way. By using cheap NH4SCN and MeOH as the radical sources, a series of valuable α-thiocyanates/methoxy ketones were synthesized under mild electrolysis conditions in acceptable yields with diverse functional group compatibility. Additionally, the scale-up experiment and synthetic transformations reveal potential applications in organic synthesis.

In vitro comparative cytotoxicity study of a novel biocidal iodo-thiocyanate complex.

Novel biocides, which avoid the induction of cross-resistance to antibiotics, are an urgent societal requirement. Here, we compared the cytotoxic and bactericidal effects of a new antimicrobial agent, the iodo-thiocyanate complex (ITC), with those of the common antiseptics, hydrogen peroxide (H2O2), povidone iodine (PVP-I) and Lugol's iodine (Lugol). The antimicrobials were co-incubated for 10 min with HeLa and Escherichia coli cells in the presence and absence of organic matter (Dulbecco's modified Eagle's medium, supplemented with 10% fetal bovine serum). The cytotoxic concentrations of ITC

Endemic goiter, thiocyanate overload, and selenium status in school-age children.

Iodine deficiency (ID) and related disorders are still major, yet unresolved health concerns. Recently, in a systematic survey of school-age children (SAC), we reported severe to moderate ID, in Ankara and three cities from Black Sea region of Turkey. The current study attempted to evaluate selenium (Se) status, thiocyanate (SCN-) overload, and their possible contribution to the goiter endemics and thyroid hormone profile observed in these cities. Thyroid ultrasonography was performed and serum Se, SCN(-), thyroid hormones, sensitive TSH (sTSH) levels, and, urinary iodine concentrations (UICs)
